Isolation, Optimization, and Partial Purification of Amylase from Chrysosporium asperatum by Submerged Fermentation

        ( Sanghvi Gaurav V. ),( Rina D. Koyani ),( Kishore S. Rajput ) 한국미생물 · 생명공학회 2011 Journal of microbiology and biotechnology Vol.21 No.5

A potent fungus for amylase production, Chrysosporium asperatum, was isolated from among 30 different cultures obtained from wood samples collected in the Junagadh forest, India. All of the isolated cultures were screened for their ability to produce amylase by submerged fermentation. Among the selected cultures, C. asperatum (Class Euascomycetes; Onygenales; Onygenaceae) gave maximum amylase production. In all of the different media tested, potato starch was found to be a good substrate for production of amylase enzyme at 30oC and pH 5.0. Production of enzyme reached the maximum when a combination of starch and 2% xylose, and organic nitrogen (1% yeast extract) and ammonium sulfate were used as carbon and nitrogen sources, respectively. There was no significant effect of metal ions on enzyme activity. The enzyme was relatively stable at 50oC for 20 min, and no inhibitory effect of Ca+2 ions on amylase production was observed.

A rare cause of unilateral upper and lower eyelid swelling: isolated conjunctival amyloidosis

        Gabriel Rodrigues,Vikram Sanghvi,Murad Lala 대한안과학회 2001 Korean Journal of Ophthalmology Vol.15 No.1

Amyloidosis is a term encompassing a group of disorders characterized by the extracellular deposition of a substance called amyloid in various tissues. A very rare cause of chronic ocular discomfort is amyloidosis. We treated a patient with the above complaint in whom both eyelids were diffusely swollen with no other chronic ophthalmic complaints. Transconjunctival biopsy revealed amyloidosis and was confirmed on congo-red staining. Since we are aware that surgical excision may cause eyelid cicatrisation or secondary xerophthalmia, we explained the pros and cons of surgery to the patient. In view of the 12-year history and lack of other symptoms, the patient preferred to be kept under observation. Follow-up at two years did not reveal any significant increase in the size of the swelling. Therefore, we recommend that in a patient with amyloidosis with no visual/ocular impairment, the condition should be left alone without surgical intervention.

FuseSharp: A Multi-Image Focus Fusion Method Using Discrete Wavelet Transform and Unsharp Masking

        Gargi J. Trivedi,Rajesh C. Sanghvi 한국전산응용수학회 2023 Journal of applied mathematics & informatics Vol.41 No.5

In this paper, a novel hybrid method for multi-focus image fusion is proposed. The method combines the advantages of wavelet transform-based methods and focus-measure-based methods to achieve an improved fusion result. The input images are first decomposed into different frequency sub-bands using the discrete wavelet transform (DWT). The focus measure of each sub-band is then calculated using the Laplacian of Gaussian (LoG) operator, and the sub-band with the highest focus measure is selected as the focused sub-band. The focused sub-band is sharpened using an unsharp masking filter to preserve the details in the focused part of the image. Finally, the sharpened focused sub-bands from all input images are fused using the maximum intensity fusion method to preserve the important information from all focus images. The proposed method has been evaluated using standard multi focus image fusion datasets and has shown promising results compared to existing methods.

MosaicFusion: merging modalities with partial differential equation and discrete cosine transformation

Gargi J. Trivedi,Rajesh C. Sanghvi 한국전산응용수학회 2023 Journal of Applied and Pure Mathematics Vol.5 No.5

In the pursuit of enhancing image fusion techniques, this research presents a novel approach for fusing multimodal images, specifically infrared (IR) and visible (VIS) images, utilizing a combination of partial differential equations (PDE) and discrete cosine transformation (DCT). The proposed method seeks to leverage the thermal and structural information provided by IR imaging and the fine-grained details offered by VIS imaging create composite images that are superior in quality and informativeness. Through a meticulous fusion process, which involves PDE-guided fusion, DCT component selection, and weighted combination, the methodology aims to strike a balance that optimally preserves essential features and minimizes artifacts. Rigorous evaluations, both objective and subjective, are conducted to validate the effectiveness of the approach. This research contributes to the ongoing advancement of multimodal image fusion, addressing applications in fields like medical imaging, surveillance, and remote sensing, where the marriage of IR and VIS data is of paramount importance.

      • Resveratrol Prevents β-Cell Dedifferentiation in Nonhuman Primates Given a High-Fat/High-Sugar Diet

        Fiori, Jennifer L.,Shin, Yu-Kyong,Kim, Wook,Krzysik-Walker, Susan M.,Gonzá,lez-Mariscal, Isabel,Carlson, Olga D.,Sanghvi, Mitesh,Moaddel, Ruin,Farhang, Kathleen,Gadkaree, Shekhar K.,Doyle, Maire American Diabetes Association 2013 Diabetes Vol.62 No.10

Eating a “Westernized” diet high in fat and sugar leads to weight gain and numerous health problems, including the development of type 2 diabetes mellitus (T2DM). Rodent studies have shown that resveratrol supplementation reduces blood glucose levels, preserves β-cells in islets of Langerhans, and improves insulin action. Although rodent models are helpful for understanding β-cell biology and certain aspects of T2DM pathology, they fail to reproduce the complexity of the human disease as well as that of nonhuman primates. Rhesus monkeys were fed a standard diet (SD), or a high-fat/high-sugar diet in combination with either placebo (HFS) or resveratrol (HFS+Resv) for 24 months, and pancreata were examined before overt dysglycemia occurred. Increased glucose-stimulated insulin secretion and insulin resistance occurred in both HFS and HFS+Resv diets compared with SD. Although islet size was unaffected, there was a significant decrease in β-cells and an increase in α-cells containing glucagon and glucagon-like peptide 1 with HFS diets. Islets from HFS+Resv monkeys were morphologically similar to SD. HFS diets also resulted in decreased expression of essential β-cell transcription factors forkhead box O1 (FOXO1), NKX6–1, NKX2–2, and PDX1, which did not occur with resveratrol supplementation. Similar changes were observed in human islets where the effects of resveratrol were mediated through Sirtuin 1. These findings have implications for the management of humans with insulin resistance, prediabetes, and diabetes.
