Quantitative Meaurement of Vibrio vulnificus Hemolysin Production

Vibrio vulnificus cytolysin/hemoIysin (VvhA) is produced during the early growth phase and abruptly inactivated in the late growth phase via self-oligomerization, which indicates that actual VvhA production, especially during the late growth phase, cannot be accurately determined by conventional functional assay methods, which measure hemolytic activity by only active VvhA monomers, In this study, we attempted to evaluate the usefulness of immunological methods for the quantitative determination of actual VvhA production, Although several electrophoretic methods were employed, the Western blot methods were not suitable for the quantitative measurement of VvhA production because of the presence of multiple or gigantic VvhA forms. The dot blot method was able to measure quantitatively actual VvhA production consistent with the transcription level of the whA gene. Accordingly, we recommend the Dot-blot method as a simple useful immunological method for the quantitative measurement of actual VvhA production.

Monodispersed $Al_2O_3-TiO_2$ Powder was prepared by metal-alkoxide hydrolsis. A homogeneous nucleation/growth occurred in the solutions containing ethanol, butanol and acetonitrile, and resulted in spherical, submicrometer-sized powder. The titania and the alumina crystals were formed at $800^{\circ}C$ and $1000^{\circ}C$, respectively. These crystala were subsequently reacted each other beyond $1320^{\circ}C$ and formed $Al_2TiO_5$. The relative densities of sintered bodies prepared with as-received powder were examined at the temperature range of 1300-$1500^{\circ}C$ and they were about 79% at $1300^{\circ}C$. The formation of aluminum titanata decreased the relative density at the temperature range of 1300-$1450^{\circ}C$, and at above $1450^{\circ}C$, the relative density started to increase again. It was observed that $\alpha-Al_2O_3$-doped aluminum titanate was more stable than pure aluminum titante at $1200^{\circ}C$.
Effect of Formaldehyde on the Water Resistance of MDF Cement Composites

Formaldehyde has widely been used for the cross-linking of polyvinyl alcohol polyvinyl alcohol polymer. The effects of formaldehyde on the water resistance of MDF cement composites were investigated as a function of types of catalyst, base or acid, and the amount of formaldehyde. The acetalization, reaction of OH group of PVA with aldehyde, was ended incompletely under base atmosphere. However, by addition of citric acid, the cross-linking of PVA polymer could be acheved through acetalization of PVA and formaldehyde. The effects of these different patterne according to the types of catalyst on the water resistance of MDF cement were studied by the preparation of PVA films and MDF composites. Thanks to the cross-linking reaction of PVA polymer chains by formaldehyde, the modified PVA films and MDF composites showed a good water-resistant propety. The modified MDF cement composite to which 3 wt% formaldehyde and 1 wt% cirtic acid were added showed 80% of initial flexural strength and good interfacial state between cement grain and polymer matrix. However, 4 wt% formaldehyde deteriorted the processing conditions, microstructures and eventually the flexural strength, causing sharp increase in the viscosity of sample dough during the mixing process. To study the relatins of flexural strength and interface of cement grain and polymer matrix, SEM and MIP measurement were performed.
Tween-80 Can Enlarge the Swarming Haloes of Human-Pathogenic Bacteria

Bacterial swarming is considered as a good in vitro model for colonization on and adhesion to the epithelial surfaces, and some efforts to enlarge swarming haloes have been made. Recently, Tween-80, a commercially available anionic surfactant, was reported to be able to enlarge the swarming haloes of some non-human-pathogenic bacteria on the surfaces of agars with relatively high viscosity by reducing surface tension, In this study, we attempted to observe whether Tween-80 can also enlarge the swarming haloes of some human-pathogenic bacteria. Proteus mirabilis, Serratia marcescens and Vibrio parahaemolyticus could swarm on the surfaces of semisolid agars containing over 0.7% Bacto agar, and their swarming haloes were 2.7-, 1.6- and 9.0-fold enlarged by Tween-80. In contrast, Escherichia coli, Vibrio vulnificus and Vibrio cholerae could swarm only on the surfaces of semisolid agars containing 0.3% Bacto agar, and their swarming haloes were 1.5-, 1.5- and 1.4-fold enlarged by Tween-80. Accordingly, Tween-80 can be particularly useful in enlarging the swarming haloes of V. parahaemolyticus on the surfaces of semisolid agars with relatively high viscosity.
