      • Spatial Analysis of Rural Medical Facilities Using Huff Model : A Case Study of Lankao County, Henan Province

        Rural health care reformed in the new situation in China, indicating that the construction of medical facilities not only to solve difficult medical problems, but also to take into account the efficiency and fairness of the use of medical resources allocation. Facing these problems, the paper aims to assess the spatial distribution of rural medical services by using geographic information systems (GIS) and spatial accessibility indexes. Lankao County in Henan Province and 21 hospitals of township and county are selected as study samples. First adopted ArcGIS10.0 to collate data. Second, the authors established a network dataset to analysis the maximum coverage for hospital services space accessibility. At last, established Huff model and built a scientific evaluation for the current hospitals’ distribution. There are several findings from the case study. First, the distribution of medical facility is unbalanced in rural areas in term of per capita, such as opportunity for assessing medical services and the convenience of utilizing medical services. Second, by using the Huff Model to calculate the potential population in service regions, the Huff Model to calculate the potential population in service regions, the results showed the low efficiency of the use of medical resources and the problems in these regions.

      • An Analysis of Regional Economy and Resourcesand Environment Coordinated Development Based on PSR Model : A Case Study of Zhongyuan Urban Agglomerations

        With the acceleration of urbanization, the resources and environment problem has become more and more prominent in Zhongyuan urban agglomerations. Based on evaluation model of resources and environment coordination degree of Pressure-State-Response (PSR), this study adopted the coordination degree analysis methods to calculate and evaluate the resources and environment coordinated development of nine central cities in Zhongyuan Urban Agglomerations from 2006 to 2013. Results showed that: (1) from 2006 to 2013, the statuses of the nine central cities in Zhongyuan Urban Agglomerations were in moderate coordination, elementary coordination, on the verge of imbalance and moderate imbalance, the economic development and the resources and environment statuses were not optimistic; (2) from 2006 to 2009, those cities’ statuses were in moderate imbalance, on the verge of imbalance and elementary coordination, the overall development coordination degree was not high, could not reach the state of coordinated development. Luoyang, Jiaozuo, Xuchang and Luohe’s coordinated development situation was not up to standard, whose status was in moderate imbalance; (3) from 2010 to 2013, the nine cities were on the verge of imbalance, elementary coordination and moderate coordination, coordinated development was improved.

        TNF-α-Induced SOX5 Upregulation Is Involved in the Osteogenic Differentiation of Human Bone Marrow Mesenchymal Stem Cells Through KLF4 Signal Pathway

        Postmenopausal osteoporosis (PMOP) is a common systemic skeletal disease characterized by reduced bone mass and microarchitecture deterioration. Although differentially expressed SOX5 has been found in bone marrow from ovariectomized mice, its role in osteogenic differentiation in human mesenchymal stem cells (hMSCs) from bone marrow in PMOP remains unknown. In this study, we investigated the biological function of SOX5 and explore its molecular mechanism in hMSCs from patients with PMOP. Our findings showed that the mRNA and protein expression levels of SOX5 were upregulated in hMSCs isolated from bone marrow samples of PMOP patients. We also found that SOX5 overexpression decreased the alkaline phosphatase (ALP) activity and the gene expression of osteoblast markers including Collagen I, Runx2 and Osterix, which were increased by SOX5 knockdown using RNA interference. Furthermore, $TNF-{\alpha}$ notably upregulated the SOX5 mRNA expression level, and SOX5 knockdown reversed the effect of $TNF-{\alpha}$ on osteogenic differentiation of hMSCs. In addition, SOX5 overexpression increased Kruppel-like factor 4 (KLF4) gene expression, which was decreased by SOX5 silencing. KLF4 knockdown abrogated the suppressive effect of SOX5 overexpression on osteogenic differentiation of hMSCs. Taken together, our results indicated that $TNF-{\alpha}$-induced SOX5 upregulation inhibited osteogenic differentiation of hMSCs through KLF4 signal pathway, suggesting that SOX5 might be a novel therapeutic target for PMOP treatment.

        Bovine Viral Diarrhea Virus Infection Induces Autophagy in MDBK Cells

        Bovine viral diarrhea virus (BVDV) is an enveloped, positive-sense, single-stranded RNA virus that belongs to the genus Pestivirus (Flaviviridae). The signaling pathways and levels of signaling molecules are altered in Madin-Darby Bovine Kidney (MDBK) cells infected with BVDV. Autophagy is a conservative biological degradation pathway that mainly eliminates and degrades damaged or superfluous organelles and macromolecular complexes for intracellular recycling in eukaryotic cells. Autophagy can also be induced as an effective response to maintain cellular homeostasis in response to different stresses, such as nutrient or growth factor deprivation, hypoxia, reactive oxygen species exposure and pathogen infection. However, the effects of BVDV infection on autophagy inMDBK cells remain unclear. Therefore, we performed an analysis of autophagic activity after BVDV NADL infection using real-time PCR, electron microscopy, laser confocal microscopy, and Western blotting analysis. The results demonstrated that BVDV NADL infection increased autophagic activity and significantly elevated the expression levels of the autophagy-related genes Beclin1 and ATG14 inMDBK cells. However, the knockdown of Beclin1 and ATG14 by RNA interference (RNAi) did not affect BVDV NADL infection-related autophagic activity. These findings provided a novel perspective to elaborate the effects of viral infection on the host cells.
