        Development and pharmacokinetic evaluation of spray-dried self-nanoemulsifying drug delivery system of sertraline

        The objective of the present study was to develop solid self-nanoemulsifying drug delivery system (SNEDDS) of poorly absorbed drug sertraline (SRT) and evaluation of its pharmacokinetic parameters in rats. The composition of optimized liquid SNEDDS was (25.42 % v/v) Labrafil M 2125 CS and Maisine 35-1 (1:1), (49.72 % v/v) Tween 80 and (24.86 % v/v) Lauroglycol 90 containing 25 mg SRT. Solid SNEDDS was prepared by spray-drying the liquid SNEDDS using dextran 40 as solid carrier. There was no significant difference (p[0.05) in the droplet size of reconstituted nanoemulsion between both liquid and solid SNEDDS. The surface characterization of spray-dried powder showed a satisfactory regular spherical shape of particles. The internal physical state of SRT was verified by X-ray diffraction analysis indicated the transformation of crystalline structure of SRT to amorphous and molecularly dispersed state. In vitro release of SRT from solid SNEDDS was highly significant (p\0.01) as compared to unformulated SRT. After oral administration of solid SNEDDS to adult Sprague–Dawley (SD) rats, the area under the curve (AUC) were 2.8- and 6.8-folds and the maximum plasma concentration (Cmax) were 3.5 -and 13-folds higher, respectively compared to those of conventional capsule and unformulated drug suspension. These results reveal that solid SNEDDS results in a significantly increased absorption of SRT compared with that from the marketed conventional capsule and aqueous suspension of SRT. Thus, this solid SNEDDS may provide a useful solid dosage form for oral delivery of poorly-water soluble lipophilic compounds.

        Measurement and Structural Modelling on Factors of Islamic Payment Gateway System Among Millennial Generation in Malaysia

        Purpose: This study aims to develop a conceptual model and validate the model using a unified theory of acceptance and use of technology to determine the Determinants of Intention to Use the Islamic Payment Gateway System among the Millennial Generation in Malaysia via a Structural Model. Based on past research, the UTAUT hypothesises four variable constructs: performance expectancy, effort expectancy, social Influence, and facilitating factors. These characteristics are predictive of behavioural intent. In terms of the study, the author added a religiosity variable instead of the four recommended moderators. Design/methodology/approach: A total of 303 field tests have been obtained from Malaysian millennial citizens using Islamic Fintech. Before the data is analysed into exploratory factor analysis, preliminary analysis such as normality and multicollinearity test has been conducted. Findings: All constructs, which are performance expectancy, effort expectancy, social Influence, facilitating condition, and behavioural intention with religiosity as the moderating, have no factor reduction, but all constructs identified two components under exploratory factor analysis. Moreover, all construct KMO tests more than 0.50. All constructs met unidimensionality, validity, reliability, and model fitness in CFA. All the requirements for model fit in this study have been achieved. Research limitations/implications: This study has significant consequences, particularly for the existing payment gateway system, which is already entering every area of human lifestyle with the millennial generation being targeted as the native user. Originality/value: This study fills the apparent gap in the existing payment gateway system literature by assessing religiosity as moderating through exploratory factor analysis.

        Protection of hepatotoxicity using Spondias pinnata by prevention of ethanol-induced oxidative stress, DNA-damage and altered biochemical markers in Wistar rats

        Background Traditional systems of medicine use herbal drugs for hepatoprotection. Thus, the study was designed to evaluate the hepatoprotective and antioxidant effects of Spondias pinnata bark extracts against ethanol-induced liver injury in Wistar rats. Methods Group I animals were treated with 1 mL/kg 0.3% carboxymethyl cellulose and Group II with 12 mL/kg 50% ethanol for 8 consecutive days. Groups III–VII animals were first treated with 400 mg/kg petroleum ether extract, chloroform extract, acetone extract (AE), ethanol extract (EE), and 100 mg/kg silymarin, and then 12 mL/kg 50% ethanol orally after 2 hours pretreatment each day for 8 consecutive days. Six hours after the last dose, blood was withdrawn. The hepatoprotective activity was assessed by several biochemical and antioxidant parameters. It was accomplished by the histopathology and DNA fragmentation study of liver tissues. Results Treatment with S. pinnata extracts, mainly AE and EE significantly (p < 0.05–0.01) and dose-dependently prevented the ethanol-induced increase in serum levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, cholesterol, bilirubin, and malondialdehyde, and decrease in reduced glutathione, catalase, superoxide dismutase, and albumin. They also attenuated the ethanol-induced DNA damage. Hepatoprotective potential of the extract was less than that of standard drug silymarin. Results of the study were well supported by the histopathological observations. Conclusion S. pinnata extracts AE and EE possess a potent hepatoprotective effect against ethanol-induced liver injury in Wistar rats, and protect them from hepatotoxicity by prevention of ethanol-induced oxidative stress, DNA-damage and altered biochemical markers.

        Flocculation kinetics and dewatering studies of quaternized cellulose derived from oil palm empty fruit bunches

        Flocculation kinetics and sludge dewatering of kaolin suspension as influenced by various q-EFBC flocculant dosing were studied. In this study, 62.5mg L1 q-EFBC exhibited the highest turbidity removal efficiency of 99.53± 0.08%. The adsorption rate of kaolin towards 12.5mg L1 to 112.5mg L1 q-EFBC dosing increased rapidly for t<60 s and became gradual before completion. The mass transfer coefficient was independent of dosage. The experimental data best-fitted the non-linear pseudo-first order due to the R2>0.99 and the lowest standard deviation. The highest rate constant of particle aggregation and breakage was consistent with the highest rate constant of particle collision, which led to the highest turbidity removal at the optimal dosage. The rate-limiting steps in the flocculation process were particle collision and aggregation since their rate constant was lower than the other kinetic constants. The lower values of SRF and TTF of treated sludge as compared to the untreated one confirmed the improvement in the dewaterability characteristic. The lowest TTF (37.44±1.44 s) and SRF (1.49×1010 m kg1) was observed for 62.5mg L1 q-EFBC. The high turbidity removal and improved sludge dewaterability indicate the potential application of q-EFBC for water treatment.
