        Development of a SCAR Marker Linked to Male Fertility Traits in ‘Jinkyool’(Citrus sunki)

        감귤류 중 수술이 퇴화되어 웅성불임형질을 나타내는 ‘청견’ 품종에 정상적인 수술의 형태를 가진 웅성가임인 ‘진귤’ 품종을 교배하여 150개체의 F₁ 집단을 구축하여 수술이 퇴화되는 개체와 정상인 개체를 분리하였다. 분리된 F₁ 개체들을 사용하여 SRAP 기법과 집단 분리 분석법(BSA)을 조합하여 웅성 가임 연관 마커 개발에 활용하였 다. F1 집단 내 150개체 중 66개체가 퇴화 수술을 갖고 있으며 웅성 가임성과 웅성 불임성의 분리비는 1:1이며 χ² 값은 2.16(p=0.05)이었다. 197개의 SRAP 프라이머 조합들 중 웅성가임 특이밴드를 형성하는 3개의 SRAP 프라이머 조합(F4/R27, F39/R60, 및 F15/R37)을 선발하였으며, 이 중 F39/R60 프라이머에 특이적으로 증폭하는 DNA단편의 염기서열을 기본으로 하여 새롭게 작성한 양방향 프라이머 조합 중 웅성 가임 계통에서만 약 1.4 Kb의 특이밴드를 증폭하는 프라이머 조합, pMS 33U/pMS 1462L를 선발하여 SCAR 마커를 개발 하였다. 이러한 결과는 개발된 SCAR 마커로 무핵성 계통들의 육종 선발에 효율성을 높일 수 있을 것으로 기대된다. In Citrus, an F1 segregation population of 150 plants was constructed from a cross between ‘Kiyomi’ (C. unshiu × C. sinensis) carrying the male sterility trait and ‘Jinkyool’ (C. sunki). Sequence-related amplification polymorphism (SRAP) combined with bulked segregant analysis was used to develop markers linked to male fertility. In the F1 population, 66 out of 150 seedlings had aborted anthers and the ratio of male sterile plants to fertile plants in the progenies matched the expected Mendelian segregation ratio of 1:1 (χ² =2.16 at p=0.05). From the profiling of the 197 SRAP primer sets, three SRAP primer sets (F4/R27, F39/R60, and F15/R37) that were closely linked to the target trait were identified and successfully converted into a sequence characterized amplified region (SCAR) marker for selection of male fertility in citrus. The SCAR marker, using the pMS 33U/pMS 1462L primer set specifically, produced a single 1.4-Kb fragment that was linked to male fertility. Our results suggested that this SCAR marker can be useful for marker-assisted selection of male sterile individuals in breeding F₁ progenies in Citrus.

        Episodes of prolonged “trance-like state” in an infant with hypothalamic hamartoma

        Hypothalamic hamartoma (HH) is one of the most common causes of central precocious puberty (CPP) in first few years of life. It can present with either seizures or CPP, although both manifestations coexist in the majority of the children. Gelastic seizures (GS), or laughing spells, are usually the first type of seizures seen in patients with HH. Although a wide variety of seizure types are known to occur in children with HH, GS are most common and consistent seizure type. The clinical presentation of HH may vary with the size and position of the mass, although large tumours typically present with both CPP and seizures. Although CPP can be managed with medical therapy, seizures can be very difficult to treat, even with multiple antiepileptic drugs. Noninvasive gamma knife surgery has been used with some success for the treatment of refractory epilepsy. We present a case of HH with very early onset seizures and CPP. The patient had an atypical form of seizures described by the parents as a "trance-like state" in which the patient had prolonged episodes of unresponsiveness lasting for hours with normal feedings during the episodes. GS occurred late in the course and were refractory to various combinations of antiepileptic drugs. A brain magnetic resonance imaging showed a large sessile HH (>20 mm). Later in the course of the disease, the patient experienced cognitive and behavioural problems. The patient underwent gamma knife surgery at nearly 5 years of age and experienced a modest response in seizure frequency. This case highlights the presentation of HH as a previously unreported seizure morphology described as a prolonged "trance-like state."

        Metabolically engineered anthocyanin-producing lime provides additional nutritional value and antioxidant potential to juice

        Anthocyanins are synthesized via the flavonoid pathway through a complex expression of several genes such as MYB transcription factors. Anthocyanins protect plants against biotic and abiotic stresses. Herein, we studied the effect of expression of MYB (VvmybA1 cloned from the red grape and Ruby cloned from ‘Moro’ blood orange) transcription factors in “Mexican” lime on juice quality and leaf pigments, leaf metabolites, and phytohormones. Anthocyanins, furanocoumarins, flavonoids, and hydroxycinnamates were analyzed with high-performance liquid chromatography–mass spectrometry, whereas chlorophylls, carotenoids, and xanthophylls were analyzed using HPLC coupled with photodiode array detector (PDA). The rest of metabolites were analyzed using gas chromatography–mass spectrometry. Overexpression of VvmybA1 and Ruby resulted in accumulation of anthocyanins in leaves, flowers, and fruits of the transgenic plants. However, the level of anthocyanins in Ruby plants was significantly lower than that in VvmybA1 plants. The level of anthocyanins and the gene expression of VvmybA1 and Ruby in young leaves were higher than mature leaves. On the other hand, the level of several furanocoumarins, and hydroxycinnamates decreased in mature VvmybA1 leaves, indicating a drainage of p-coumaric acid due to the induction of anthocyanins biosynthesis. The level of chlorophyll decreased in mature VvmybA1 leaves, whereas zeaxanthin level increased, indicated a photoprotection role for anthocyanins. Most of polar and volatile metabolites also decreased VvmybA1 leaves, indicating a decrease in the photosynthetic efficiency. Benzoic acid and salicylic acid increased, whereas auxins decreased. The level of abscisic acid was not affected by the overexpression of VvmybA1 and the plants showed normal growth and development. Overexpression of VvmybA1 highly increased the antioxidant activity of the transgenic juice and leaves, whereas overexpression of Ruby showed only a slight increase. The pH, °Brix value, and TA of the transgenic juice were not affected by the expression of VvmybA1 or Ruby.
