        Correlations between lignin content and related genes, weathering deterioration, and soybean seed quality at pre-harvest

        The present study aimed to identify the relationship between lignin content in soybean pods, pod dehiscence rate, gene expression from phenylpropanoid pathway, cell wall lignifcation, and deterioration of seed due to weather. To evaluate the weathering deterioration, an experiment simulating 150 mm of rainfall was performed in greenhouse conditions. Gene expression was assessed by RT-qPCR. Results showed that soybean cultivars BRS Jiripoca and BRS 388 RR considered tolerant to weathering deterioration, presented higher levels of lignin content in pods. The data showed that dehiscence rate was neither a trait involved in weathering deterioration tolerance nor in lignin content in soybean pods. Lignin content and weathering deterioration were highly negatively correlated, probably due to changes in cell wall permeability to water accordingly to lignin level. Higher expression levels of genes involved in lignin biosynthesis and cell wall lignifcation were identifed in both tolerant cultivars, however, with opposite expression profles and in diferent developmental phases, suggesting a possible temporal strategy to cope with environmental adverse conditions.

        Mobilization Functions of the Bacteriocinogenic Plasmid pRJ6 of Staphylococcus aureus

        Plasmid pRJ6 is the first known bacteriocinogenic mobilizable (Mob) plasmid of Staphylococcus aureus. Its Mob region is composed of four mob genes (mobCDAB) arranged as an operon, a genetic organization uncommon among S. aureus Mob plasmids. oriTpRJ6 was detected in a region of 431 bp, positioned immediately upstream of mobC. This region, when cloned into pCN37, was able to confer mobilization to the recombinant plasmid only in the presence of pRJ6. The entire Mob region, including oriTpRJ6, is much more similar to Mob regions from several coagulase-negative staphylococci plasmids, although some remarkable similarities with S. aureus Mob plasmids can also be noted. These similarities include the presence within oriTpRJ6 of the three mcb (MobC binding sites), firstly described in pC221 and pC223, an identical nick site also found in these same plasmids, and a nearly identical srapC223 site (sequence recognized by MobA). pRJ6 was successfully transferred to S. epidermidis by conjugation in the presence of the conjugative plasmid pGO1. Altogether these findings suggest that pRJ6 might have been originally a coagulase-negative staphylococci plasmid that had been transferred successfully to S. aureus.

      • Identification and characterization of fluid escape structures (pockmarks) in the Estremadura Spur, West Iberian Margin

        <P><B>Abstract</B></P> <P>Located on the West Iberian margin, between Cabo Carvoeiro and Cabo da Roca, the Estremadura Spur is a trapezoidal promontory elongated in an east-west direction, extending until the Tore seamount. Recently a field with more than 70 pockmarks was discovered in the NW region of the Estremadura Spur outer shelf (Lourinhã Monocline). Pockmarks are the seabed culminations of fluid migration through the sedimentary column and their characteristic seabed morphologies correspond to cone-shaped circular or elliptical depressions. The characterization of these features and the understanding of the associated fluid escape process are the main objectives of this work. Here we characterize these structures to understand their structural and stratigraphic control based on: 1) Seismic processing and interpretation of the high resolution 2D single-channel sparker seismic dataset, 2) Bathymetric and Backscatter interpretation and 3) ROV direct observation of the seafloor.</P> <P>The analysis of the seismic profiles allowed the identification of six seismic units, disturbed by the migration and accumulation of fluids. The Estremadura Spur outer shelf has been affected by several episodes of fluid migration and fluid escape during the Pliocene-Quaternary that are expressed by a vast number of seabed and buried pockmarks. At present, the pockmarks are mainly inactive, as the seabed pockmarks are covered by recent sediments. It is concluded that the migration of fluids to the seabed occurred over the Pliocene-Quaternary, as indicated by the buried pockmarks at different depths below the seabed. The vertical stacking of various pockmarks suggests a cyclical fluid flow activity that can possibly be the result of the eustatic sea level variations and the subsequent changes of the hydrostatic pressure.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The Estremadura Spur pockmark field is the first fluid seepage system identified in the West Iberian Margin. </LI> <LI> The Estremadura Spur outer shelf has been affected by several episodes of fluid migration and fluid escape, during the Pliocene-Quaternary. </LI> <LI> At present-day the pockmarks are mainly inactive, as the seabed pockmarks are recovered by recent sediments. </LI> </UL> </P>
