Syntheses, Structures, and Characterization of Quaternary Tellurites, Li3MTe4O11 (M = Al, Ga, and Fe)

        Lü,, Minfeng,Jo, Hongil,Oh, Seung-Jin,Lee, Suheon,Choi, Kwang-Yong,Yu, Yang,Ok, Kang Min ACS AMERICAN CHEMICAL SOCIETY 2017 Inorganic Chemistry Vol.56 No.10

<P>Three new quaternary lithium metal tellurites, Li3MTe4O11 (M = Al, Ga, and Fe), have been synthesized through hydrothermal and solid-state reactions by heating a mixture of LiOH center dot H2O, TeO2, and M2O3. The structures of the title compounds have been determined by single-crystal and powder X-ray diffraction. Li3MTe4O11 reveal three-dimensional (3D) frameworks that consist of MO6 octahedra, TeO3 trigonal pyramids, and TeO4 polyhedra. The variable coordination mode of Te4+ within the framework leads to the formation of 1D channels that host Li+ cations on both tetrahedral and octahedral sites. The bulk and grain boundary Li+ ion conductivities for a Li3FeTe4O11 pellet in open air are estimated to be 1.0 x 10(-4) and 2.7 x 10(-6) S cm(-1), respectively, at room temperature from the impedance profile analysis. A lower activation energy of 19.9 kJ mol(-1) is obtained for the system, which is similar to that of Li10GeP2S12 (24 kJ mol(-1)). Detailed characterizations such as thermal, spectroscopic, and magnetic properties for the reported materials are also reported.</P>

Attenuation of Experimental Autoimmune Hepatitis in Mice with Bone Mesenchymal Stem Cell-Derived Exosomes Carrying MicroRNA-223-3p

        Lu, Feng-Bin,Chen, Da-Zhi,Chen, Lu,Hu, En-De,Wu, Jin-Lu,Li, Hui,Gong, Yue-Wen,Lin, Zhuo,Wang, Xiao-Dong,Li, Ji,Jin, Xiao-Ya,Xu, Lan-Man,Chen, Yong-Ping Korean Society for Molecular and Cellular Biology 2019 Molecules and cells Vol.42 No.12

MicroRNA-223-3p (miR-223-3p) is one of the potential microRNAs that have been shown to alleviate inflammatory responses in pre-clinical investigations and is highly encased in exosomes derived from bone mesenchymal stem cells (MSC-exosomes). MSC-exosomes are able to function as carriers to deliver microRNAs into cells. Autoimmune hepatitis is one of the challenging liver diseases with no effective treatment other than steroid hormones. Here, we examined whether MSC-exosomes can transfer miR-223-3p to treat autoimmune hepatitis in an experimental model. We found that MSC-exosomes were successfully incorporated with miR-223-3p and delivered miR-223-3p into macrophages. Moreover, there was no toxic effect of exosomes on the macrophages. Furthermore, treatments of either exosomes or exosomes with miR-223-3p successfully attenuated inflammatory responses in the liver of autoimmune hepatitis and inflammatory cytokine release in both the liver and macrophages. The mechanism may be related to the regulation of miR-223-3p level and STAT3 expression in the liver and macrophages. These results suggest that MSC-exosomes can be used to deliver miR-223-3p for the treatment of autoimmune hepatitis.

      • Cecropin Suppresses Human Hepatocellular Carcinoma BEL-7402 Cell Growth and Survival in vivo without Side-Toxicity

        Jin, Xiao-Bao,Wang, Ying-Jiao,Liang, Lu-Lu,Pu, Qiao-Hong,Shen, Juan,Lu, Xue-Mei,Chu, Fu-Jiang,Zhu, Jia-Yong Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.13

Conventional chemotherapy against hepatocellular carcinoma typically causes various side effects. Our previous study showed that cecropin of Musca domestica can induce apoptosis in human hepatocellular carcinoma BEL-7402 cells in vitro. However, whether cecropin inhibits BEL-7402 cell in vivo and the question of possible side effects remained undentified. The present study confirmed tumor-inhibitory effects of cecropin in vivo, and furthermore strongly suggested that cecropin cytotoxicity in BEL-7402 cells in vivo may be mainly derived from its pro-apoptotic action. Specifically, we found that cecropin exerted no obvious side effects in tumor-bearing mice as it had no significant hematoxicity as well as visceral toxicity. Therefore, cecropin may be a potential candidate for further investigation as an antitumor agent against hepatocellular carcinoma.

      • Function and optimization of colorectal cancer vaccine candidate spGA733-FcK and spGA733-Fc expressed in baculovirus-insect cell system

        Lu Qiao,Kyung-Jin Lee,Kisung Ko 한국응용곤충학회 2013 한국응용곤충학회 학술대회논문집 Vol.2013 No.10

The antigen GA733 is a cell-surface highly expressed glycoprotein on most human colorectal carcinomas. GA733 can be characterized as a cancer vaccine. In this study, GA733 was fused to the human immunoglobulin IgG Fc fragment to become recombinant gene GA733-Fc. Based on this, 4 recombinant genes were constructed as follows: GA733-Fc with signal peptide sequence and fusion of ER retention sequence (KDEL) (spGA733-FcK), GA733-Fc with signal sequence (spGA733-Fc), GA733-Fc fused to ER retention sequence (GA733-FcK) without signal peptide and GA733-Fc without signal peptide. Baculovirus-insect cell expression system is widely used for the high level production of recombinant proteins especially for glycoproteins. Constructed 4 recombinant genes were cloned to baculovirus express vectors. DH10Bac E.coli.-mediated transformation was used to generate recombinant bacmid DNA. Recombinant DNA was confirmed by PCR. Insect cell was transfected by bacmid to produce the recombinant baculovirus infects insect cell to produce recombinant protein. Western blot and sandwich ELISA showed the expression of recombinant proteins. Each cell lines (sf9 and HighFive) differed in recombinant proteins production level and protein secretion capability. N-Glycosylation analysis showed the function of signal peptide and ER retention sequence (KDEL). Taken together, baculovirus-insect cell system can be used to express recombinant GA733-Fc glycoproteins.

HYERS-ULAM STABILITY OF DERIVATIONS IN FUZZY BANACH SPACE: REVISITED

        Gang Lu,Yuanfeng Jin,Gang Wu,Sungsik Yun 한국수학교육학회 2018 純粹 및 應用數學 Vol.25 No.2

Lu et al. [27] dened derivations on fuzzy Banach spaces and fuzzy Lie Banach spaces and proved the Hyers-Ulam stability of derivations on fuzzy Banach spaces and fuzzy Lie Banach spaces. It is easy to show that the denitions of derivations on fuzzy Banach spaces and fuzzy Lie Banach spaces are wrong and so the results of [27] are wrong. Moreover, there are a lot of seroius problems in the statements and the proofs of the results in Sections 2 and 3. In this paper, we correct the denitions of biderivations on fuzzy Banach algebras and fuzzy Lie Banach algebras and the statements of the results in [27], and prove the corrected theorems.

      • Phase II Study on Javanica Oil Emulsion Injection (Yadanzi®) Combined with Chemotherapy in Treating Patients with Advanced Lung Adenocarcinoma

        Lu, Yan-Yan,Huang, Xin-En,Cao, Jie,Xu, Xia,Wu, Xue-Yan,Liu, Jin,Xiang, Jin,Xu, Lin Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.8

Purpose: To investigate the efficacy and safety of Javanica oil emulsion injection (Yadanzi$^{(R)}$) combined with pemetrexed and platinum (PP) for treating patients with advanced lung cancer. Patients and Methods: From June 2011 to June 2013, we recruited 58 patients with advanced lung cancer, and divided them into two groups. Twenty eight patients received Yadanzi$^{(R)}$ (from ZheJiang Jiuxu Pharmaceutical Co., Ltd.) together with PP chemotherapy (combined group), while the others were given only PP chemotherapy (control group). After two cycles of treatment, efficacy and safety of treatment were evaluated. Results: The overall respnse rate [(CR+PR+SD)/(CR+PR+SD+PD)] of the combined group was higher than that of control group (89.7% vs. 86.2%, p>0.05). Regarding rate of life improvement, it was 82.8% in combined group, and 51.7% in the control group (p<0.05). In terms of side effects, leukopenia in combined group was less frequent than that in control group (p<0.05). More patients in the control group were found to suffer liver toxicity. Conclusions: Javanica oil emulsion injection combined with chemotherapy could be considered as a safe and effective regimen in treating patients with advanced lung adenocarcinoma. It can improve the quality of life and reduce the possibility of leukopenia. Further clinical trials with a large sample size should be conducted to confirm whether addition of Yadanzi$^{(R)}$ to chemotherapy could increase the response rate, reduce toxicity, enhance tolerability and improve quality of life for patients with advanced lung cancer.

Mechanism of E2F1 in the proliferation, migration, and invasion of endometrial carcinoma cells via the regulation of BMI1 transcription

        Lu Yanyang,Wei Ying,Shen Xiaoqin,Tong Yixi,Lu Jin,Zhang Yahui,Ma Yun,Zhang Rong 한국유전학회 2023 Genes & Genomics Vol.45 No.11

Background Endometrial carcinoma (EC) is the most prevalent gynecological cancer. Transcription factor (TF) regulates a large number of downstream target genes and is a key determinant of all physiological activities, including cell proliferation, differentiation, apoptosis, and cell cycle. The transcription factor E2F1 shows prominent roles in EC. BMI1 is a member of Polycomb suppressor Complex 1 (PRC1) and has been shown to be associated with EC invasiveness. It is currently unclear whether E2F1 can participate in the proliferation, migration, and invasion processes of EC cells by regulating BMI1 transcription. Objective We investigated whether E2F1 could participate in the proliferation, migration, and invasion processes of EC cells by regulating BMI1 transcription, in order to further clarify the pathogenesis and etiology of EC, and provide reference for identifying potential therapeutic targets and developing effective prevention and treatment strategies for this disease. Methods Human endometrial epithelial cells (hEECs) and human EC cell lines were selected. E2F1 expression was assessed by Western blot. E2F1 was silenced in AN3CA or overexpressed in HEC-1 by transfections, or E2F1 was silenced and BMI1 was overexpressed in AN3CA by cotransfection. Cell proliferation, migration, and invasion were detected by MTT, wound healing, and Transwell assays. The binding sites between E2F1 and BMI1 promoters were predicted through JASPAR website, and the targeted binding was verified by dual-luciferase report and ChIP assays. Results E2F1 was up-regulated in human EC cell lines, with its expression highest in AN3CA, and lowest in HEC-1. AN3CA invasion, migration, and proliferation were repressed by E2F1 knockdown, while those of HEC-1 cells were promoted by E2F1 overexpression. E2F1 overexpression increased the activity of wild type BMI1 reporter vector promoter, while this promotion was weakened after mutation of the predicted binding site in the BMI1 promoter. In the precipitated E2F1, BMI1 promoter site level was higher than that of IgG immunoprecipitant. BMI1 silencing suppressed AN3CA cell growth. BMI1 overexpression partially abrogated E2F1 silencing-inhibited EC cell growth. Conclusion E2F1 promoted EC cell proliferation, invasion, and migration by promoting the transcription of BMI1.

Study on 3D Characterized Profile and Point Accuracies of Ground Micro-Pyramid-Structured Si Surface

Jin Xie,Yong-Xian Lu,Xu-Ran Liu,Yan-Jun Lu 한국정밀공학회 2013 International Journal of Precision Engineering and Vol. No.

It is very difficult to evaluate 3D profile accuracy of micro-machined surface due to the 3D characterization of its measured point cloud. Hence, three ideal point cloud models, which are grid point model, slash profile model and horizontal profile model, were constructed to register 3D measured point cloud of micro-machined surface and evaluate the accuracies of characterized profile and points. First, #400 and #600 diamond wheel V-tips were employed to fabricate non-integrated and integrated micro-pyramidstructured surfaces with 173 μm in depth and 0.865 in aspect ratio in Computer Numerical Control (CNC) grinding system, respectively; then a white light interferometer was utilized to measure the micro-ground surfaces; finally, registration accuracy and registration efficiency were investigated with regard to characterized profile and point accuracies. The results show that registration accuracy increases and registration efficiency decreases with increasing model point number, but they have little relation with the posture of measured point cloud. Registration error may converge to micro-ground form error when model point number is larger than measured point number. Moreover, the slash profile and horizontal profile models may register characterized profile and point more precisely than grid point model. The slash profile model can produce the best registration accuracy and efficiency. It is confirmed that the micro-ground form errors are 23.8 μm and 7.9 μm, the characterized profile errors are 37.2 μm and 19.0 μm and the characterized peak errors are 51.2 μm and 34.1 μm for non-integrated surface and integrated surface, respectively. As a result,a #600 diamond grinding wheel may be used to perform a precision micro-grinding in CNC grinding system.

Expression of the Tobacco Mosaic Virus Movement Protein Alters Starch Accumulation in Nicotiana benthamiana

        Jin, Zheung Lu,Hong, Joon Ki,Yun, Dae Jin,Lee, Sang Yeol,Choi, Young Ju,Bahk, Jeong Dong,Beachy, Roger N.,Cho, Moo Je,Lim, Chae Oh 한국식물학회 2002 Journal of Plant Biology Vol.45 No.2

Nicotiana benthamiana plants were transformed with the movement protein (MP) gene of tobacco mosaic virus (TMV), using Agrobacterium-mediated transformation. Plants regenerated from the transformed cells accumulated 30-kDa MP and complemented the activity of TMV MP when infected with chimeric TMVs containing defective MP. These transgenic plants displayed stunting, pale-green leaves, and starch accumulations, indicating that TMV MP altered the carbon partitioning for leaves involved in TMV cell-to-cell movement.
