ATP Binding Cassette Transporter A1 is Involved in Extracellular Secretion of Acetylated APE1/Ref-1
Lee, Yu Ran,Joo, Hee Kyoung,Lee, Eun Ok,Cho, Hyun Sil,Choi, Sunga,Kim, Cuk-Seong,Jeon, Byeong Hwa MDPI AG 2019 INTERNATIONAL JOURNAL OF MOLECULAR SCIENCES Vol.20 No.13

Acetylation of nuclear apurinic/apyrimidinic endonuclease-1/redox factor-1 (APE1/Ref-1) is associated with its extracellular secretion, despite the lack of an N-terminal protein secretion signal. In this study, we investigated plasma membrane targeting and translocation of APE1/Ref-1 in HEK293T cells with enhanced acetylation. While APE1/Ref-1 targeting was not affected by inhibition of the endoplasmic reticulum/Golgi-dependent secretion, its secretion was reduced by inhibitors of ATP-binding cassette (ABC) transporters, and siRNA-mediated down-regulation of ABC transporter A1. The association between APE1/Ref-1 and ABCA1 transporter was confirmed by proximal ligation assay and immunoprecipitation experiments. An APE1/Ref-1 construct with mutated acetylation sites (K6/K7R) showed reduced co-localization with ABC transporter A1. Exposure of trichostatin A (TSA) induced the acetylation of APE1/Ref-1, which translocated into membrane fraction. Taken together, acetylation of APE1/Ref-1 is considered to be necessary for its extracellular targeting via non-classical secretory pathway using the ABCA1 transporter.

Mitogen-activated protein kinases (MAPK) signalling cascades are activated by extracellular stimuli such as environmental stresses and pathogens in higher eukaryotic plants. To know more about MAPK signalling in plants, aMAPK cDNA clone, OsMAPK33, was isolated from rice. The gene is mainly induced by drought stress. In phylogenetic analysis, OsMAPK33 (Os02g0148100) showed approximately 47-93% identity at the amino acid level with other plant MAPKs. It was found to exhibit organ-specific expression with relatively higher expression in leaves as compared with roots or stems, and to exist as a single copy in the rice genome. To investigate the biological functions of OsMAPK33 in rice MAPK signalling, transgenic rice plants that either overexpressed or suppressed OsMAPK33 were made. Under dehydration conditions, the suppressed lines showed lower osmotic potential compared with that of wild-type plants, suggesting a role of OsMAPK33 in osmotic homeostasis. Nonetheless, the suppressed lines did not display any significant difference in drought tolerance compared with their wild-type plants. With increased salinity, there was still no difference in salt tolerance between OsMAPK33-suppressed lines and their wild-type plants. However, the overexpressing lines showed greater reduction in biomass accumulation and higher sodium uptake into cells, resulting in a lower K+/Na+ ratio inside the cell than that in the wild-type plants and OsMAPK33-suppressed lines. These results suggest that OsMAPK33 could play a negative role in salt tolerance through unfavourable ion homeostasis. Gene expression profiling of OsMAPK33 transgenic lines through rice DNA chip analysis showed that OsMAPK33 altered expression of genes involved in ion transport. Further characterization of downstream components will elucidate various biological functions of this novel rice MAPK.

Lee, Jina,Shin, Na-Ri,Jung, Mi-Ja,Roh, Seong Woon,Kim, Min-Soo,Lee, Jung-Sook,Lee, Keun Chul,Kim, Young-Ok,Bae, Jin-Woo Society for General Microbiology 2013 International journal of systematic and evolutiona Vol.63 No.2

A Gram-stain-negative, non-motile, aerobic, endospore forming and rod-shaped bacterium, designated strain L10(T), was isolated from marine sediment collected from the South Korean coast. The organism grew optimally under conditions of 30 C, 1?% (w/v) NaCl and pH 6.0. It was oxidase-negative and catalase-positive. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain L10(T) was associated with the genus Paenibacillus and most closely related to Paenibacillus barcinonensis BP-23(T) (98.2?% similarity). The major fatty acids of strain L10(T) were iso-C(14?:?0), anteiso-C(15?:?0) and iso-C(16?:?0). The cell-wall peptidoglycan was the A1γ type, and the predominant isoprenoid quinone was menaquinone-7. Strain L10(T) contained two unidentified lipids, an unidentified amino-phospholipid, phosphatidylethanolamine, phosphatidylglycerol, and diphosphatidylglycerol. The G+C content of the genomic DNA was 44 mol% and the DNA-DNA hybridization values with closely related strains were below 142?%. Based on phenotypic, genotypic, and phylogenetic data, strain L10(T) should be classified as a novel species within the genus Paenibacillus. The name Paenibacillus oceanisediminis sp. nov. is proposed. The type strain is L10(T) (?=?KACC 16203(T)?=?JCM 17814(T)).

A study on nanoparticle filtration characteristics of multilayer meltblown depth filters
Lee, Kang-San,Hasolli, Naim,Jeon, Seong-Min,Lee, Jae-Rang,Kim, Kwang-Deuk,Park, Young-Ok,Hwang, Jungho Korean Association for Particle and Aerosol Resear 2016 Particle and Aerosol Research Vol.12 No.3
Due to recent development in nanotechnology and increasing usage and production of nanomaterials, numerous studies related to environment, sanitation and safety handling of nanoparticle are being conducted. Since nanoparticles can be easily absorbed into human bodies through breathing process, based on their toxic substances and their large specific surface, these particles can cause serious health damage. Therefore, to reduce nanoparticle emissions, nanofiltration technology is becoming a serious issue. Filtration is a separation process during which a fluid passes through a barrier by removing the particles from the stream. Barrier filters can be made of various materials and shapes. One of the most common type of barrier filter is the fibrous filter. Fibrous filters are divided in two types: nonwoven and woven fabrics. Polypropylene is a thermoplastic material, used as a base material for melt blown nonwoven fabric. In this study, we examined filtration property of KCl nanoparticles with a mean particle diameter of 75 nm using multilayer meltblown filter samples. These experiments verify that the penetration of nanoparticle in the filter correlate with pressure drop; the meltblown layer MB1 has the greatest effect on dust collection efficiency of the filter. Among all tested samples, dust collection efficiency of 2-layer filter was best. However, when considering the overall pressure drop and dust collection efficiency, the 4-layer filter has the highest quality factor for particles smaller than 70 nm.
