To investgate the causative Virus in aborbon of horse in Cheju, we examined the spleen of affected foal by immunohtochemistq and immunogold electron microscopy using EHV - specific antisera. Immunohistochemical staining revealed that a large number of EHV immunoreadive cells were deteded in the red pulp as well as in the sinudoidal cells in the affected spleen. By the immunogold electron microxopy(EM) of crude preparation of EHV culture, electron dense gold paricles were labelled along the envelope of amorphous virus parhcles with vanng sizes of 50-200nm. We suppose that immunohistochemical method is good for the diagnosis of tissues in EHV infections, and immunogold EM is recommended for the study of virus dia gnosis of fluid specimen as well as the ultrastustural study.
Synthesis and Optical Properties of Cycloolefin Copolymers for Plastic Substrates

<P>Cycloolefin copolymers comprise a new class of polymer materials showing properties, of high glass transition temperature, optical clarity, low shrinkage, low moisture absorption and low birefringence. There are several types of cyclic olefin copolymers based on different type of cyclic monomers and polymerization methods. In ethylene/norbornene (E/NB) copolymerization, <I>i</I>Pr(Cp)(Flu)ZrCl<SUB>2</SUB> catalyst exhibited more activity than Cp<SUB>2</SUB>ZrCl<SUB>2</SUB> and <I>rac</I>-Et(Ind)<SUB>2</SUB>ZrCl<SUB>2</SUB> catalysts. By the incorporation of Norbornene in copolymer, the glass transition temperature (Tg) of copolymer became higher. The Tg of copolymer increased up to 190°C. The transmission of copolymers was above 90% over 300 ∼ 900 nm. The RI of ethylene/tetracyclododecene (E/TCD) copolymer was higher than that of E/NB copolymer. The RI of E/NB copolymer could not changed increasing NB contents, however, the RI of E/TCD copolymer increased with increasing of TCD contents. The E/NB copolymer has very low birefringence and that film has very smooth surface.</P>
Streptomyces sp. T-256의 세포의 Adenosine Deaminase의 성질

Purine nucleotide의 대사계에 관련하는 aminohydrolase 가운데 adenosine deaminase는 동물 기원이 대부분이다. 미생물 기원의 효소에 대한 연구보고는 희소하며 특별히 세포외 효소로서의 adenosine deaminase 산생 미생물에 대한 보고는 극히 미진하다. 그러므로 본 실험은 미생물 기원 가운데서 Streptomyces T-256이 산생하는 세포의 adenosine deaminase의 효소학적인 성질에 대해서 검토하였다. Some properties of an extracellular adenosine deaminase produced from Streptomyces sp. Y-256 were examined after 30-80% of ammonium sulfate fractionation. The optimun pH and temperature for the stability of this enzyme were found to be near 7.0 and 40℃, respectively. The enzyme was stable in 0.05 M of potassium phosphate buffer. As results of examination for the enzyme activity to various pHs, the enzyme was generally highest in its activity near pH 7.0 but inactivated completely near pH 4.5 and pH 8.5. The enzyme activity to tempertures was highest near 37℃ andcompletely disappeared near 60℃. Of metal ions used 1 mM of Mg2+ increased slightly the enzyme activity, but 1 mM of α,α'-dipyridyl, NaCN and α-phenanthyoline, including 10mM of ethylendiaminetetraacetic acid, monoiodoacetate, pentachlorophenol and p-chloromercuric benzoate completely inhibited the enzyme activity.

Heat shock protein 70 (HSP70), a chaperone proteinassociated with tumorigenesis and chemoresistance, hasattracted significant attention as a potential therapeutic targetfor the development of anticancer drugs. Here, the effects ofpifithrin-μ, an effective dual inhibitor of HSP70 and p53, onanticancer activities and epithelial–mesenchymal transition(EMT) were investigated in malignant mesothelioma (MM)cells. MSTO-211HAcT cells, pre-incubated in a mediumcontaining lactic acid, showed more potent resistance to cisplatinand gemcitabine, compared with their acid-sensitiveparental MSTO-211H cells. Pifithrin-μ treatment inducedboth apoptosis and necroptosis, which were accompaniedby an EMT-like phenomenon, as evidenced by an elongatedcell morphology, decreased levels of epithelial cell markersincluding E-cadherin, claudin-1, and β-catenin, increasedlevels of mesenchymal markers including Snail, Slug, andvimentin, and increased cell migratory property. Moreover,pifithrin-μ increased intracellular ROS levels, which is associatedwith mitochondrial dysfunction and decreased cellularATP content. A series of changes caused by pifithrin-μ treatmentwere effectively restored by lowering the ROS levelthrough pretreatment with N-acetylcysteine. Collectively,our results suggest that pifithrin-μ may promote the metastaticbehavior of surviving cells by triggering the EMT, despite its effective cell-killing action against MM cells,possibly linked to oxidative mitochondrial dysfunction andATP depletion.

Apigenin, a naturally occurring flavonoid, is known to exhibit significant anticancer activity. This study was designed to determine the effects of apigenin on two malignant mesothelioma cell lines, MSTO-211H and H2452, and to explore the underlying mechanism(s). Apigenin significantly inhibited cell viability with a concomitant increase in intracellular reactive oxygen species (ROS) and caused the loss of mitochondrial membrane potential (ΔΨm), and ATP depletion, resulting in apoptosis and necroptosis in monolayer cell culture. Apigenin upregulated DNA damage response proteins, including the DNA double strand break marker phospho (p)- histone H2A.X. and caused a transition delay at the G₂/M phase of cell cycle. Western blot analysis showed that apigenin treatment upregulated protein levels of cleaved caspase-3, cleaved PARP, p-MLKL, and p-RIP3 along with an increased Bax/Bcl-2 ratio. ATP supplementation restored cell viability and levels of DNA damage-, apoptosisand necroptosis-related proteins that apigenin caused. In addition, N-acetylcysteine reduced ROS production and improved Δm loss and cell death that were caused by apigenin. In a 3D spheroid culture model, ROS-dependent necroptosis was found to be a mechanism involved in the anti-cancer activity of apigenin against malignant mesothelioma cells. Taken together, our findings suggest that apigenin can induce ROS-dependent necroptotic cell death due to ATP depletion through mitochondrial dysfunction. This study provides us a possible mechanism underlying why apigenin could be used as a therapeutic candidate for treating malignant mesothelioma.
Ring-resonator-integrated tunable external cavity laser employing EAM and SOA.

<P>We propose and demonstrate a tunable external cavity laser (ECL) composed of a polymer Bragg reflector (PBR) and integrated gain chip with gain, a ring resonator, an electro-absorption modulator (EAM), and a semiconductor optical amplifier (SOA). The cavity of the laser is composed of the PBR, gain, and ring resonator. The ring resonator reflects the predetermined wavelengths into the gain region and transmits the output signal into integrated devices such as the EAM and SOA. The output wavelength of the tunable laser is discretely tuned in steps of about 0.8 nm through the thermal-optic effect of the PBR and predetermined mode spacing of the ring resonator.</P>
방선균이 산생하는 세포외 Adenosine Deaminase의 특징

As results of examination for the enzyme activity to various pHs, the enzyme was generally highest in its activity near pH 7.0 but inactivated completely near pH 4.0 and pH 8.5 The enzyme activity to tempertures was highest near 37℃ and completely disappeared near 60℃. Of metal ions used, 1 mM of Ba2+ increased slightly the enzyme activity, but 1 mM of Hg2+ and Ag2+ markedly inactivated the enzyme activity. I mM of α, α'-dipyridyl, NaCN and o -phenanthroline, including 10mM of ethylenediaminetetraacetic acid, monoiodoacetate, pentachlorophenol and p-chloromercuric benzoate completely inhibited the enzyme activity, but 0. lmM of NaF, NaN3 and monoiodoacetate slightly increased the enzyme activity.
