      • The modernization approach to the regional processes monitoring of the social services provision in the context of the modern public policy

        The modernization approach to monitoring regional processes of providing social services is proposed. The methodological approach is developed in the context of the modern public policy, which includes the following stages: the formation of a system of indicators that characterize the level of the social services modernization; the determination of the levels of regions development by such components as the modernization level of the economic sphere, the modernization level of the demographic component and the modernization level of employment; the determination of weights for each group of indicators and calculation of integrated indicators, ranking of regions; the allocation of criteria for classification and grouping of regions according to the modernization level of the social services sphere; highlighting the most important problems of each region. The proposed method is tested on the example of the Ukraine's regions. According to the results of calculations of the modernization level of the processes of providing social services, the ranking and grouping of the regions was carried out. The rating allowed to distinguish four groups of regions: regions with a high modernization level of social services, regions with above-average levels, as well as regions with medium and low levels. The author's modernization approach to monitoring the processes of providing social services allows to investigate the real state of the main indicators influencing these processes and to identify problem regions in order to develop mechanisms to stimulate their development.

        Approaching Narrative Text with a Film Director’s Mindset: Producing Real and Imagined Scenes in the Secondary English Classroom

        This article presents a qualitative action research project that took place in a twelfth grade English class in the southeastern United States during a two week unit on narrative text and film. The purpose of this study was to examine how adopting a film director’s mindset could impact students’ reading of narrative text. The proposed director’s mindset tool, a pedagogical approach for examining filmic images and establishing basic cinematic understandings, provides a visual method to help students examine textual narratives, including formal elements of literature (e.g., characterization, point of view, setting, imagery, and mood), and describe their visualizations through the language and conventions of film. Data sources include a text pre/post-inventory, a film pre/post-inventory, a film-to-text translation exercise, a director’s mindset exercise, individual and group Instagram projects, and a director’s mindset evaluation. Findings suggest that the use of film can enhance students’ recognition of formal literary elements, visualization of narrative text, and methods for creative interpretation. To prepare students for the media-dominated world beyond their academic classrooms, the authors propose the use of the director’s mindset tool as an approach for helping students comprehend and evaluate the copious images that play a part in their everyday lives.

      • Formation of a Professional Communication Culture Among the Students Using Information Technologies

        Analyzing the psychological and pedagogical literature, we found the researchers' interest in the problem posed. The concept of "culture of professional communication» is considered, which is interpreted as the level of realization of creative abilities, exchange of messages, organization of mutual understanding, mutual knowledge in the process of professionally directed interaction between subjects, in which interpersonal relationships arise, manifest and form. The concept of "professional culture of communication of a teacher" is interpreted. The motives that are socially significant in the professional communication of the teacher are highlighted. The necessity of forming a culture of professional communication among students, in particular by means of information technologies in the present, is clarified. The interactive component of professional communication is considered. The types of interactions between people in everyday life (ritual and entertainment interaction, joint purposeful activity, no interaction, game and interpersonal interaction) are identified. Traditional and specific forms and methods of teaching are written out. All interactive technologies carried out by means of information technologies are conventionally divided into four groups, depending on the form of educational activity appropriate for their use (pair (work of the subject with the teacher or peers one on one by means of Information Technologies); frontal (the teacher simultaneously teaches a group of subjects by means of Information Technologies); group or cooperative (all subjects teach each other by means of Information Technologies); individual (independent work of the subject using Information Technologies)). In the higher education institution, future specialists should learn knowledge, acquire skills on the basic rules of the culture of professional communication and methods of interaction and their effective use, which is possible with the use of Information Technologies. Recommendations for optimal professional communication have been developed that help you express your thoughts easily and beautifully, and conduct a dialogue in a relaxed and harmonious way.

        The role of p53 in the efficiency of photodynamic therapy with hypericin and subsequent long-term survival of colon cancer cells

        Photodynamic therapy with hypericin (HY-PDT) is known as an efficient modality for treatment of various cancerous and non-cancerous diseases. Although the role of p53 protein in cell death signaling is well established, relatively little is known of its impact on the efficiency of HY-PDT. Comparison of sensitivity and long-term survival of p53-null versus wt-p53-expressing HCT-116 cells is reported here. The lack of p53 function did not affect cell proliferation or attenuate the initial phases of programmed cell death. However, analyses of apoptosis in the final stages revealed suppression of its incidence and delayed activation of caspase-3 in p53-null cells. Significantly higher clonogenic ability, especially in hypoxia, was identified in the case of p53-null cells. Induction of Mcl-1 and Bax levels were more prominent in wt-pt53 cells. Interestingly, the level of Bcl-2 did not react to HY-PDT at all, in both cell lines. Bringing the evidence together, we prove that despite insignificant impact on overall toxicity, expression of p53 affects the clonogenic efficiency of HCT-116 cells. Since destruction of tumor tissue and its vascular system by PDT tends to lead to hypoxia, superior survival of p53-deficient tumor cells under given conditions might result in recurrence of cancer diseases.

        The use of the shape memory alloy, Nitinol wire, is investigated as an actuator for enhancing the damping in structural vibration systems. The first-order mathematical model of the Nitinol wire is obtained from the experimental data for an actuator. Finite element method is utilized for the strain gage sensor model, which is installed at the root of cantilever beam. A simple system, cantilever beam, is built as a flexible structural system to implement a control law with the Nitinol wire actuator. The system model including sensor and actuator is derived, which agrees with the experimental results. The actuator dynamics is augmented with the system so as to design PI controller and the one of robust controllers, LQG/LTR controller, and the control laws are implemented experimentally. The experimental study shows the feasibility of utilizing the Nitinol wire as an actuator for the purpose of vibration control.

        Drug efflux transporters, MRP1 and BCRP, affect the outcome of hypericin-mediated photodynamic therapy in HT-29 adenocarcinoma cells

        Photodynamic therapy (PDT) is a flexible multi-target therapeutic approach. One of the main requirements of successful PDT is sufficient intracellular concentration of an applicable photosensitizer. Mechanisms of anticancer drug elimination by tumour cells are mostly linked to the elevated expression and activity of P-glycoprotein (P-gp), multidrug resistance-associated protein 1 (MRP1), breast cancer resistance protein (BCRP) and P450 monooxygenases. The interaction of hypericin with this cell drug-defence system is still unclear. We report here for the first time increased activity of MRP1 and BCRP in HT-29 colon cancer cells treated with hypericin per se. On the contrary, pre-treatment with proadifen (SKF525A) affected the function of MRP1 and BCRP leading to increased hypericin content, which might indicate a possible link between proadifen and these ABC transporter proteins. Subsequent enhanced intracellular oxidative stress was accompanied by loss of mitochondrial membrane potential, activation of caspase-9 and -3, PARP cleavage and onset of apoptosis. In conclusion, our study suggests that drug efflux transporters MRP1 and BCRP affect the pharmacokinetics of hypericin in HT-29 colon adenocarcinoma cells, and the action of hypericin-mediated PDT (HY-PDT) should be modulated by pre-treatment with their specific inhibitors.

        Analyzing the psychological and pedagogical literature, we showed the interest of researchers in the problem posed. The concept of competence is considered, which is interpreted as giving the key to solving a wide range of educational and life tasks. Research competence implies the ability to cooperate, enter into contacts, readiness for changes, for self-determination and is an integral quality of the individual, expressed in the readiness and ability to independently search for solutions to new problems and creative transformation of reality based on a set of personal and meaningful knowledge, skills, methods of activity and value attitudes.The article offers conditions that certify the improvement of forms and methods of training students in the formation of research competence of future specialists. The use of innovative technologies contributes to improving the level of training of future specialists: students are better prepared for classes, take an active part in the assimilation of program material in laboratory classes. It is noted that this creates a subject-subject relationship between the student and the teacher, and changes the attitude of students to classes. In the process of such organization of educational activities, students are convinced of the need for knowledge and its effectiveness, learn to compare, generalize, classify, establish cause-and-effect relationships, express opinions, defend their point of view, they ensure success in their studies, and develop research competence. It is proved that in order to apply the latest technologies, the teacher himself must know them well, that is, constantly improve himself, master new methods, techniques, ideas, which will help him create new pedagogical technologies and implement them in the educational process.

        A Comparative Field Study of Global Pose Estimation Algorithms in Subterranean Environments

        In this article, we perform a novel and extended field evaluation of the state-of-the-art algorithmic frameworks’ performance on global pose estimation. More specifically, we focus on relocalizing a mobile robot in a pre-built 3D point cloud map of a large subterranean environment. The evaluation is divided into two parts. The first part consists of multiple simulations performed in two different Gazebo SubT worlds, where one is flat with various types of features, while another has uneven structure and is more textured. The second part is an experimental evaluation and takes place in a real-world underground tunnel. In all evaluation tests, the robot’s pose is selected in such a way that we can test the robustness, as well as the feature extraction capability, of each algorithm. The evaluation is carried out using three ROS packages: a) hdl_global_localization using both BBS and FPFH+RANSAC, b) LIO-SAM_based_relocalization, and c) Fast-LIO-Localization. Our goal is to have a clear view of each algorithm’s efficiency in terms of CPU load, memory allocation and time to relocalize, while we increase the size of the map and transverse the robot in different parts of the map.
