Although its demand increased greatly due to the volatile strong flavor and bioactive molecules, little information has been about the cultural characteristics of Tricholoma matsutake. In this study, we investigated the optimal medium composition of liquid culture with the goal of shortening the culture period, and to maximize polysaccharide production and mycelial growth. From these experiments we found that the optimal medium contained 40 g/L, glucose; 30 g/L, yeast extract; 1.5 g/L, $KH_2PO_4$; and 1 g/L $MgSO_4{\cdot}7H_2O$. In flask culture, the maximum mycelial growth and polysaccharide production were 22.45 and 5.3 g/L, which were about 9 and 3 g/L higher than that at the basal medium, respectively.

<P><B>Abstract</B></P><P>In this study, the effects of agitation and aeration on mycelial growth and exo-polysaccharide production were examined in batch cultures of <I>Tricholoma matsutake</I>. Agitation was varied from 100 to 300?rpm and aeration was varied from 0.5 to 1.5?vvm. Mycelial growth was 21.87?g/l at 150?rpm, and exo-polysaccharide production was 8.79?g/l at 1.5?vvm. When we analyzed the polysaccharide extractions from the cultured mycelium and the culture broth of <I>T. matsutake</I>, 1.4?g of crude polysaccharide was found per 100?g of dried weight in the cultured mycelium, and 1.47?g/l of polysaccharides was found in the culture broth. In addition, the amounts of β-Glucan in the soluble polysaccharide fractions of the cultured mycelium and culture broth were 75.4% and 83.6%, respectively. The cultured mycelium and the culture broth contained a higher amount of β-Glucan than that of the fruiting body.</P>
Optimal design using genetic algorithm with nonlinear elastic analysis

An optimal design method with nonlinear elastic analysis is presented. The proposed nonlinear elastic method overcomes the drawback of the conventional LRFD method that accounts for nonlinear effect by using the moment amplification factors of $B_1$ and $B_2$. The genetic algorithm used is a procedure based on Darwinian notions of survival of the fittest, where selection, crossover, and mutation operators are employed to look for high performance ones among sections in the database. They are satisfied with the constraint functions and give the lightest weight to the structure. The objective function taken is the total weight of the steel structure and the constraint functions are strength, serviceability, and ductility requirement. Case studies of a planar portal frame, a space two-story frame, and a three-dimensional steel arch bridge are presented.

본 연구에서는 차량-교량간의 상호작용 효과가 교량의 동적거동에 미치는 영향을 분석하기 위하여 실험적 연구를 수행하였다. 이를 위하여 차량-교량간 상호작용이 가능하도록 윤하중 실험장치를 개발하여 단순교 형식의 교량을 대상으로 이동 질량 실험을 수행하였다. 이동질량 이론을 이용한 해석 결과와 이동질량 실험의 결과를 상호 분석하여 각각의 합리성을 검증하였다. 실험분석 결과, 개발된 윤하중 실험기는 차량-교량간 상호작용에 의한 교량의 동적 거동을 재현할 수 있음을 알았다. 아울러 차량-교량간 상호작용이 교량의 동적거동에 미치는 영향을 이동질량 실험을 통하여 모사할 수 있었다. In this paper, all analytical and experimental study is performed in order to determine the effects of interaction between vehicle and structure. For this purpose an improved wheel tracking machine and an adequate single span bridge are designed. Results presented in the paper show that wheel tracking machine including moving mass effects can demonstrate more accurate dynamic interaction between vehicle and structure.
