        Eco RI Restriction Fragment Length Polymorphism of L - myc gene has arisen by a single base change

        Human genomic DNA shows an EcoRI restriction fragment length polymorphism (RFLP) of L-myc. defined by 10 and 6 kilobase EcoRI fragments. Recently, it has been found that the RFLP of L-myc gene was closely correlated with the progression of human lung cancer to the metastasis. To investigate the differences of the genetic information between two ECoRI fragments, we have isolated clones of these genes from a genomic library. The genomic library was prepared using the DNA isolated from the tumor tissue of a lung cancer patient. The 10Kb and 6Kb L-myc genes were mapped using several restriction enzymes. Comparison of the restriction map of 10Kb L-myc gene to that of 6Kb gene showed that the 6Kb gene may have arisen by the mutation of the 10Kb gene leading to the creation of a EcoRI site. Nucleotide sequence analysis of both genes in this region revealed that a single base change from C to A in 10Kb gene resulted in a creation of a ECoRI site.
