Conditional PTEN-deficient Mice as a Prostate Cancer Chemoprevention Model

Background: We generated a mouse model of prostate cancer based on the adult-prostate-specific inactivation of phosphatase and tensin homolog (PTEN) using the Cre-loxP system. The potential of our mice as a useful animal model was examined by evaluating the chemopreventive efficacy of the anti-androgen, chlormadinone acetate (CMA). Materials and Methods: Six-week-old mice were treated subcutaneously with $50{\mu}g/g$ of CMA three times a week for 9 or 14 weeks and sacrificed at weeks 15 and 20. Macroscopic change of the entire genitourinary tract (GUT) and histologically evident prostate gland tumor development were evaluated. Proliferation and apoptosis status in the prostate were examined by immunohistochemistry. Results: CMA triggered significant shrinkage of not only the GUT but also prostate glands at 15 weeks compared to the control (p=0.017 and p=0.010, respectively), and the trend became more marked after a further five-weeks of treatment. The onset of prostate adenocarcinoma was not prevented but the proliferation of cancer cells was inhibited by CMA, which suggested the androgen axis is critical for cancer growth in these mice. Conclusions: Conditional PTEN-deficient mice are useful as a preclinical model for chemoprevention studies and serve as a valuable tool for the future screening of potential chemopreventive agents.

In order to predict of accuracy of movement just before the onset of movement using biological signals, the relationships between MRCP measured as a physiological index and the accuracy of task performance were investigated. As experiments, subjects touched the target appearing 300 pixels away from a start point in a verticaldirection on a touch-sensitive screen with the forefinger. During experiments, EEG, EMG as trigger, high-speed camera images, and task efficiency were acquired. As a result, significant differences between the high- and low-performance groups were clearly confirmed on the slope of NS in MRCPs. Furthermore, a difference between these groups wasconfirmed in duration of ballistic movement.

The high plateaus in west China may provide suitable sites for astronomical observations with the institute's middle-range telescopes and possibly with larger telescopes. Under China-Japan collaborations for site survey in west China, we have been conducting searches for good sites and monitoring their characteristics over several years. As recent results of our site survey show, sites in west Tibet are revealed with a high possibility of good astronomical observations. Weather characteristics at Gar in Ali, Tibet, show its high clear-sky ratios, especially in winter, comparable to Mauna Kea, Hawaii. But it has some wind problem in winter, where stronger wind speeds, over 20m/sec, occur frequently even though the sky is clear. To find calmer sites, we have conducted numerical simulations for the Ali area using the Japan Meteorological Agency NonHydrostatic Model. We have found another site, named ZoZo Hill, near Gar. We will continue to monitor the Gar site to clarify weather characteristics over the whole year and hopefully start to negotiate for site monitoring at ZoZo Hill this year.
