        Investigation of gastro protective activity of Xanthium strumarium L. by modulation of cellular and biochemical marker

        Peptic ulcer owes its pathological features to disregulated oxido-nitrosative and necrotic changes in gastrointestinal mucosa. Xanthium strumarium L. is a ubiquitous, abundant herb possessing an array of pharmacological properties. In view of anti-inflammatory and antioxidant activity of Xanthium strumarium L., the present investigation was designed to unravel its antiulcer potential in pylorus ligation induced gastric ulcer in laboratory animals. X. srtumarium L. leaves were extracted by refluxing with 95 % ethanol to obtained X. srtumarium L. ethanolic extract (XSEE). After 1 h pretreatment period of XSEE (100, 200 and 400 mg/kg) gastric ulcer was induced by ligation of pylorus portion of stomach. Various parameters including volume of gastric fluid, pH of gastric fluid, free as well as total acidity were measured in the gastric fluid whereas ulcer area, ulcer index, mucin content, superoxide dismutase, myeloperoxidase, nitric oxide, total calcium, histamine level, membrane bound inorganic phosphate (Na-K-ATPase), TNF-${\alpha}$ level, DNA fragmentation were determined in the gastric tissue. Pretreatment with XSEE (200 and 400 mg/kg) significantly and dose dependently (P<0.01 and P<0.001) decreased ulcer area, ulcer index, macroscopical gastric damage score, volume of gastric fluid, free acidity as well as total acidity and mucin content. It also significantly increased pH of gastric fluid. Rats treated with XSEE (200 and 400 mg/kg) significantly and dose dependently (P<0.01 and P<0.001) attenuated decreased level of superoxide dismutase, nitric oxide and Na-K-ATPase. It also significantly decreased level of myeloperoxidase, calcium and inflammatory cytokine i.e. TNF-${\alpha}$. XSEE treated rats showed significant reduction in the pylorus ligation induced apoptotic changes. The possible mechanism by which Xanthium strumarium exerts its gastro-protective action may be due to free radical quenching, DNA repair and down regulation of oxidative-nitrosative stress along with cytokines.

        Ameliorative effects of type-A procyanidins polyphenols from cinnamon bark in compound 48/80-induced mast cell degranulation

        Allergic diseases are a significant health concern in developing countries. Type-A procyanidin polyphenols from cinnamon (Cinnamomum zeylanicum Blume) bark (TAPP-CZ) possesses antiasthmatic and antiallergic potential. The present study was aimed at the possible anti-allergic mechanism of TAPP-CZ against the compound 48/80 (C48/80)-induced mast cell degranulation in isolated rat peritoneal mast cells (RPMCs). TAPP-CZ (1, 3, 10, and 30 μg/ml) was incubated for 3 hours with isolated, purified RPMCs. The C48/80 (1 μg/ml) was used to induce mast cell degranulation. The mast cell viability was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay whereas histamine, β-hexosaminidase (β-HEX), and interleukin-4 (IL-4) levels were determined in RPMCs. TAPP-CZ (3, 10, and 30 μg/ml) showed significant and dose-dependent decrease in a number of degranulated cells and levels of markers (histamine, β-HEX, and IL-4) as compared with C48/80 control. In conclusion, TAPP-CZ stabilizes mast cell and cause inhibition of the allergic markers such as histamine, IL-4, and β-HEX in IgE-mediated manner. The present study supports mast cell stabilization as a possible mechanism of action of TAPP-CZ against immune respiratory disorders such as asthma and allergic rhinitis.

        Alleviating exercise-induced muscular stress using neat and processed bee pollen: oxidative markers, mitochondrial enzymes, and myostatin expression in rats

        Background: The current study was designed to investigate the influence of monofloral Indian mustard bee pollen (MIMBP) and processed monofloral Indian mustard bee pollen (PMIMBP) supplementation on chronic swimming exercise-induced oxidative stress implications in the gastrocnemius muscle of Wistar rats. Methods: MIMBP was processed with an edible lipid-surfactant mixture (Captex 355:Tween 80) to increase the extraction of polyphenols and flavonoid aglycones as analyzed by UV spectroscopy and high performance liquid chromatography-photo diode array. Wistar rats in different groups were fed with MIMBP or PMIMBP supplements at a dose of 100 mg/kg, 200 mg/kg and 300 mg/kg individually, while being subjected to chronic swimming exercise for 4 weeks (5 d/wk). Various biochemical [superoxide dismutase (SOD), glutathione (GSH), malonaldehyde (MDA), nitric oxide (NO), and total protein content], mitochondrial (Complex I, II, III, and IV enzyme activity), and molecular (myostatin mRNA expression) parameters were monitored in the gastrocnemius muscle of each group. Results: Administration of both MIMBP (300 mg/kg) and PMIMBP (100 mg/kg, 200 mg/kg, and 300 mg/kg) wielded an antioxidant effect by significantly improving SOD, GSH, MDA, NO, and total protein levels. Further MIMBP (300 mg/kg) and PMIMBP (200 mg/kg and 300 mg/kg) significantly improved impaired mitochondrial Complex I, II, III, and IV enzyme activity. Significant down-regulation of myostatin mRNA expression by MIMBP (300 mg/kg) and PMIMBP (200 mg/kg and 300 mg/kg) indicates a muscle protectant role in oxidative stress conditions. Conclusion: The study establishes the antioxidant, mitochondrial upregulatory, and myostatin inhibitory effects of both MIMBP and PMIMBP in exercise-induced oxidative stress conditions, suggesting their usefulness in effective management of exercise-induced muscular stress. Further, processing of MIMBP with an edible lipid-surfactant mixture was found to improve the therapeutic efficiency of pollen.
