        Piping structural design for the ITER thermal shield manifold

        <P><B>Abstract</B></P> <P>The thermal shield (TS) provides the thermal barrier in the ITER tokamak to minimize heat load transferred by thermal radiation from the hot components to the superconducting magnets operating at 4.2K. The TS is actively cooled by 80K pressurized helium gas which flows from the cold valve box to the cooling tubes on the TS panels via manifold piping. This paper describes the manifold piping design and analysis for the ITER thermal shield. First, maximum allowable span for the manifold support is calculated based on the simple beam theory. In order to accommodate the thermal contraction in the manifold feeder, a contraction loop is designed and applied. Sequential Quadratic Programming (SQP) method is used to determine the optimized dimensions of the contraction loop to ensure adequate flexibility of manifold pipe. Global structural behavior of the manifold is investigated when the thermal movement of the redundant (un-cooled) pipe is large.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We finalized piping design of ITER thermal shield manifold for procurement. </LI> <LI> Support span is determined by stress and deflection limitation. </LI> <LI> SQP, which is design optimization method, is used for the pipe design. </LI> <LI> Benchmark analysis is performed to verify the analysis software. </LI> <LI> Pipe design is verified by structural analyses. </LI> </UL> </P>

        Effect of Producing Different Phenazines on Bacterial Fitness and Biological Control in Pseudomonas chlororaphis 30-84

        Pseudomonas chlororaphis 30-84 is a biological control agent selected for its ability to suppress diseases caused by fungal pathogens. P. chlororaphis 30-84 produces three phenazines: phenazine-1-carboxylic acid (PCA), 2-hydroxy-phenazine-1-carboxylic acid (2OHPCA) and a small amount of 2-hydroxy-phenazine (2OHPHZ), and these are required for fungal pathogen inhibition and wheat rhizosphere competence. The two, 2-hydroxy derivatives are produced from PCA via the activity of a phenazine-modifying enzyme encoded by phzO. In addition to the seven biosynthetic genes responsible for the production of PCA, many other Pseudomonas strains possess one or more modifying genes, which encode enzymes that act independently or together to convert PCA into other phenazine derivatives. In order to understand the fitness effects of producing different phenazines, we constructed isogenic derivatives of P. chlororaphis 30-84 that differed only in the type of phenazines produced. Altering the type of phenazines produced by P. chlororaphis 30-84 enhanced the spectrum of fungal pathogens inhibited and altered the degree of take-all disease suppression. These strains also differed in their ability to promote extracellular DNA release, which may contribute to the observed differences in the amount of biofilm produced. All derivatives were equally important for survival over repeated plant/harvest cycles, indicating that the type of phenazines produced is less important for persistence in the wheat rhizosphere than whether or not cells produce phenazines. These findings provide a better understanding of the effects of different phenazines on functions important for biological control activity with implications for applications that rely on introduced or native phenazine producing populations.

        DNA-Stable Isotope Probing Integrated with Metagenomics for Retrieval of Biphenyl Dioxygenase Genes from Polychlorinated Biphenyl-Contaminated River Sediment

        <B>ABSTRACT</B><P>Stable isotope probing with [<SUP>13</SUP>C]biphenyl was used to explore the genetic properties of indigenous bacteria able to grow on biphenyl in PCB-contaminated River Raisin sediment. A bacterial 16S rRNA gene clone library generated from [<SUP>13</SUP>C]DNA after a 14-day incubation with [<SUP>13</SUP>C]biphenyl revealed the dominant organisms to be members of the genera <I>Achromobacter</I> and <I>Pseudomonas</I>. A library built from PCR amplification of genes for aromatic-ring-hydroxylating dioxygenases from the [<SUP>13</SUP>C]DNA fraction revealed two sequence groups similar to <I>bphA</I> (encoding biphenyl dioxygenase) of <I>Comamonas testosteroni</I> strain B-356 and of <I>Rhodococcus</I> sp. RHA1. A library of 1,568 cosmid clones was produced from the [<SUP>13</SUP>C]DNA fraction. A 31.8-kb cosmid clone, detected by aromatic dioxygenase primers, contained genes of biphenyl dioxygenase subunits <I>bphAE</I>, while the rest of the clone's sequence was similar to that of an unknown member of the <I>Gammaproteobacteria</I>. A discrepancy in G+C content near the <I>bphAE</I> genes implies their recent acquisition, possibly by horizontal transfer. The biphenyl dioxygenase from the cosmid clone oxidized biphenyl and unsubstituted and <I>para</I>-only-substituted rings of polychlorinated biphenyl (PCB) congeners. A DNA-stable isotope probing-based cosmid library enabled the retrieval of functional genes from an uncultivated organism capable of PCB metabolism and suggest dispersed dioxygenase gene organization in nature.</P>

        Pheochromocytoma 적출술의 마취관리 증례보고

        Pheoehromocytoma is a kind of very rare endocrine disease and which is completely recovered by surgical resection of the tumor, but rapidly changing cardiovascular response to the cathecholamine during the anesthetic course and after removal of tumor make the aestheic management difficulty. Various anesthetic agent can be used with their good and bad characteristics sucessfully if the pathophysiology of the disease and pharmacology of the drugs are kept in mind. We have experienced sucessful anesthetic management for pheocromocytoma removal using halothane-N₂O-O₂, and muscle relaxant and cardiovascular change was supported by pentolamine (Regitine), Propranolol(Inderal), and Levophed.
