        14-3-3ζ Regulates Immune Response through Stat3 Signaling in Oral Squamous Cell Carcinoma

        Ectopic expression of $14-3-3{\zeta}$ has been found in various malignancies, including lung cancer, liver cancer, head and neck squamous cell carcinoma (HNSCC), and so on. However, the effect of $14-3-3{\zeta}$ in the regulation of interactions between tumor cells and the immune system has not been previously reported. In this study, we aimed to investigate whether and how $14-3-3{\zeta}$ is implicated in tumor inflammation modulation and immune recognition evasion. In oral squamous cell carcinoma (OSCC) cell lines and cancer tissues, we found that $14-3-3{\zeta}$ is overexpressed. In OSCC cells, $14-3-3{\zeta}$ knockdown resulted in the up-regulated expression of inflammatory cytokines. In contrast, $14-3-3{\zeta}$ introduction attenuated cytokine expression in human normal keratinocytes and fibroblasts stimulated with interferon-${\gamma}$ (IFN-${\gamma}$) and lipopolysaccharide (LPS). Furthermore, supernatants from $14-3-3{\zeta}$ knockdown OSCC cells dramatically altered the response of peritoneal macrophages, dendritic cells and tumor-specific T cells. Interestingly, Stat3 was found to directly interact with $14-3-3{\zeta}$ and its disruption relieved the inhibition induced by $14-3-3{\zeta}$ in tumor inflammation. Taken together, our studies provide evidence that $14-3-3{\zeta}$ may regulate tumor inflammation and immune response through Stat3 signaling in OSCC.

        Effects of Lubiprostone on Pacemaker Activity of Interstitial Cells of Cajal from the Mouse Colon

        Lubiprostone is a chloride ($Cl^-$) channel activator derived from prostaglandin $E_1$ and used for managing constipation. In addition, lubiprostone affects the activity of gastrointestinal smooth muscles. Interstitial cells of Cajal (ICCs) are pacemaker cells that generate slow-wave activity in smooth muscles. We studied the effects of lubiprostone on the pacemaker potentials of colonic ICCs. We used the whole-cell patch-clamp technique to determine the pacemaker activity in cultured colonic ICCs obtained from mice. Lubiprostone hyperpolarized the membrane and inhibited the generation of pacemaker potentials. Prostanoid $EP_1$, $EP_2$, $EP_3$, and $EP_4$ antagonists (SC-19220, PF-04418948, 6-methoxypyridine-2-boronc acid N-phenyldiethanolamine ester, and GW627368, respectively) did not block the response to lubiprostone. L-NG-nitroarginine methyl ester (L-NAME, an inhibitor of nitric oxide synthase) and 1H-[1,2,4]oxadiazolo[4,3,-a]quinoxalin-1-one (ODQ, an inhibitor of guanylate cyclase) did not block the response to lubiprostone. In addition, tetraethylammonium (TEA, a voltage-dependent potassium [$K^+$] channel blocker) and apamin (a calcium [$Ca^{2+}$]-dependent $K^+$ channel blocker) did not block the response to lubiprostone. However, glibenclamide (an ATP-sensitive $K^+$ channel blocker) blocked the response to lubiprostone. Similar to lubiprostone, pinacidil (an opener of ATP-sensitive $K^+$ channel) hyperpolarized the membrane and inhibited the generation of pacemaker potentials, and these effects were inhibited by glibenclamide. These results suggest that lubiprostone can modulate the pacemaker potentials of colonic ICCs via activation of ATP-sensitive $K^+$ channel through a prostanoid EP receptor-independent mechanism.

        Synthesis of SrTiO3 Fibers and Their Effects on the Thermoelectric Properties of La0.1Dy0.1Sr0.75TiO3 Ceramics

        SrTiO 3 fi bers were fabricated by an in situ hydrothermal method using hydrated TiO 2 fi bers as both template and reactant. La 0.1 Dy 0.1 Sr 0.75 TiO 3 powders containing x wt.% SrTiO 3 fi bers ( x = 0, 1, 3, 5) were prepared by the sol–gel method andthen sintered at 1450 °C under a reducing atmosphere (N 2 /H 2 = 95/5). XRD analysis showed that the samples were mainlycomposed of SrTiO 3 phase and a few Dy 2 Ti 2 O 7 phase. TiO 2 phase was detected in the samples with x = 3 and x = 5, and itspeak intensity clearly reinforced with increasing x . With the addition of SrTiO 3 fi bers, the electrical conductivity increasedsignifi cantly and the Seebeck coeffi cient kept almost unchanged, resulting in a high power factor of 1015 μW m −1 K −2 at200 °C with a loading of 3 wt.% SrTiO 3 fi bers. Meanwhile, combined with low thermal conductivity, the sample with 3 wt.%SrTiO 3 fi bers showed the peak ZT value of 0.19 at 500 °C, which was 127% higher than that of La 0.1 Dy 0.1 Sr 0.75 TiO 3 .

      • Core–shell structured mesoporous magnetic nanoparticles and their magnetorheological response

        <P><B>Abstract</B></P> <P>As a new magneto-responsive smart material, core–shell structured mesoporous Fe<SUB>3</SUB>O<SUB>4</SUB>@mSiO<SUB>2</SUB> nanoparticles were fabricated by a surfactant-based sol–gel approach using Fe<SUB>3</SUB>O<SUB>4</SUB> nanoparticles, and then adopted as magnetorheological (MR) materials. The Fe<SUB>3</SUB>O<SUB>4</SUB> particles were initially synthesized by a solvo-thermal process. Scanning electron microscopy and transmission electron microscopy were employed to observe the morphology of the as-synthesized mesoporous silica-coated Fe<SUB>3</SUB>O<SUB>4</SUB> particles. Fourier transform infrared spectroscopy and X-ray diffraction results showed that the mesoporous silica shell is successfully coated on the Fe<SUB>3</SUB>O<SUB>4</SUB> nanoparticles. BET adsorption measurement also confirmed their mesopore formation. Rheological properties of the silica-coated Fe<SUB>3</SUB>O<SUB>4</SUB>-based MR fluid dispersed in silicone oil were investigated, and a change in the slope of the yield stress as a function of magnetic field strength from 2.0 to 1.5 was observed, with improved dispersion stability.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Core–shell structured mesoporous Fe<SUB>3</SUB>O<SUB>4</SUB>@mSiO<SUB>2</SUB> nanoparticles were fabricated. </LI> <LI> Both dynamic and elastic yield stresses of MR fluids were well correlated with universal yield stress equation. </LI> <LI> Dispersion stability of MR fluid was improved compared to the sedimentation rate of Fe<SUB>3</SUB>O<SUB>4</SUB>-based MR fluid. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Synthesis and Viscoelastic Behavior of Non-Stoichiometric Spinel Ferrite Particle Suspension

        <P>Magnetorheological (MR) fluid with tunable and reversible phase transition is one of the most promising smart materials for engineering applications. In this paper, to improve the MR performance of ferrites, non-stoichiometric Zn-doped spinel ferrite (Zn<SUB>0.417</SUB>Fe<SUB>2.583</SUB>O<SUB>4</SUB>) nanoparticles with a high saturation magnetization and a truncated octahedron-like shape were synthesized using a thermal decomposition process. The crystallographic structure was analyzed using X-ray diffraction and field emission transmission electron microscope image. The Zn<SUB>0.417</SUB>Fe<SUB>2.583</SUB>O<SUB>4</SUB> had a high saturation magnetization value of 101 <TEX>$\text {A}\cdot \text {m}^{2}$</TEX>/kg at 933 kA/m, showing higher saturation magnetization than other ferrites. The MR characteristics of the Zn<SUB>0.417</SUB>Fe<SUB>2.583</SUB>O<SUB>4</SUB> particles suspended in silicone oil were studied using a rotation rheometer under various magnetic field strengths. This MR fluid exhibited typical MR characteristics, and its flow curves were fit well with the Herschel–Bulkley model.</P>

      • Microwave Ablation Treatment of Liver Cancer with a 2,450-MHz Cooled-shaft Antenna: Pilot Study on Safety and Efficacy

        To evaluate efficacy of microwave ablation in a primary clinical study, sixty patients (44 men, 16 women; mean age 53 years) with 96, 1-8 cm (mean $3.20{\pm}0.17$ cm) liver cancers were treated with 2,450-MHz internally cooled-shaft antenna. Complete ablation (CA) and local tumor progression (LTP) rates as well as complications were determined. CA rates in small (< 3.0 cm), intermediate (3.1-5.0 cm) and large (5.1-8.0 cm) liver cancers were 96.4% (54/56), 92.3% (24/26) and 78.6% (11/14), respectively. During a mean follow-up period of $17.17{\pm}6.52$ months, LTP occurred in five (5.21%) treated cases. There was no significant difference in the CA and LTP rates between the HCC and liver metastasis patient subgroups (P<0.05). Microwave ablation provides a reliable, efficient, and safe technique to perform hepatic tumor ablation.

      • 3D CACT-assisted Radiofrequency Ablation Following Transarterial Chemoembolization for Hepatocellular Carcinoma: Early Experience

        Background: To explored the value of 3D C-arm CT (CACT) guidance system in performing radiofrequency ablation (RFA) following transarterial chemoembolizationon (TACE) for hepatocellular carcinomas. Materials and Methods: RFA of hepatocellular carcinomas (HCC) were performed on 15 patients (21 lesions) with the assistance of CACT guidance system. Technical success, procedure time, complications and patient radiation exposure were investigated. The puncture performance level was evaluated on a five-point scale (5-1: excellent-poor). Complete ablation rate was evaluated after two months follow-up using enhanced CT scans. Results: The technical success rate of RFA procedure under CACT navigation system was 100 %. Mean total procedure time was $24.24{\pm}6.53min$, resulting in a mean effective exposure dose of $15.4{\pm}5.1mSv$. The mean puncture performance level rated for CACT guided RFA procedure was $4.87{\pm}0.35$. Complete ablation (CA) was achieved in 20 (95.2%) of the treated 21 tumors after the first RFA session. None of patients developed intra-procedural complications. Conclusions: 3D CACT guidance system enables reliable and efficient needle positioning by providing real-time intraoperative guidance for performing RFA on HCCs.
