        The Effect of Solvents on Solid Dispersion of Ipriflavone with Polyvinylpyrrolidone In Vivo

        Ipriflavone is a synthetic flavonoid derivate that improves osteoblast cell activity inhibiting bone resorption. In order to improve the bioavailability, solid dispersions of ipriflavone with PVP (poly-N-vinylpyrrolidone, MW=40,000 g/ mole) were prepared by a spray-drying method. During the manufacturing of solid dispersion, various solvents [ethanol (EtOH), acetonitrile, methylene chloride and cosolvent-EtOH:acetone=1:1] were used to dissolve the ipriflavone and PVP. Scanning electron microscopy (SEM) and differential scanning calorimetry (DSC) were used to evaluate the physicochemical interaction between ipriflavone and PVP. Particle size, crystallinity and the area of the endotherm (ΔH) of solid dispersed ipriflavone using the acetonitrile as solvent were much smaller than those of the other preparation types. Bioavailability of ipriflavone in vivo was changed by solvents. When considering the result of in vivo test, solid dispersion of ipriflavone using the acetonitrile as solvent showed the best choice.

        Preparation and Characterization of Solid Dispersion of Ipriflavone with Polyvinylpyrrolidone

        Solid dispersions of ipriflavone with PVP were prepared by a spray-drying method in order to improve the bioavailability. They were measured with scanning electron microscopy, differential scanning calorimetry, x-ray powder diffraction, and Fourier transform infrared spectroscopy to evaluate the physicochemical interaction between ipriflavone and PVP and study the correlation between these physicochemical characteristics and bioavailability. Ipriflavone exhibited crystallinity, whereas PVP was almost amorphous. The area of the endotherm(△H) of freezer milled ipriflavone, freezer milled ipriflavone physically mixed with freezer milled PVP, and physically mixed ipriflavone with PVP was almost the same, whereas △H of the solid dispersed ipriflavone with PVP was much smaller than that of the other preparation types. Also, the crystallinity and the crystal size of ipriflavone in the solid dispersed ipriflavone with PVP were much smaller than those of the other preparation types. Form the in vivo test, the AUC of the solid dispersed ipriflavone with PVP was approximately 10 times higher than that of the physically mixed ipriflavone with PVP. The solid dispersion using the spray-drying method with a water-soluble polymer, PVP, may be effective for the improvement of the bioavailability.

        제제형태에 따른 이프리플라본의 생체이용률 비교

        Bioavailability of ipriflavone (3-phenyl-7-isopropoxy-4H-1-benzopyran-4-one, IP), an antiosteoporotic drug with poor water-solubility, was studied for various types of pharmaceutical preparation in SD rats. The IP preparation types included ① intact IP, ② freezer milled IP (FIP), ③ freezer milled IP physically mixed with freezer milled poly-N-vinylpyrrolidone (PVP) (FIP+FPVP) and ④ spray-dried IP with PVP (SIP). Upon oral administration, SIP showed significantly higher absorption and elimination half-lives and the lag time (t_(lag)) than those of FIP+FPVP (approximately 2-fold). These results may be due to a sustained releasing effect of IP in the gastrointestinal tract by spray-drying with PVP. The C_(max) of SIP was about 2 and 10 times higher than those of FIP+FPVP and FIP, respectively. The AUC of SIP was about 6 times higher than that of FIP+FPVP and 60 times that of FIP. Scanning electron microscopy (SEM) showed that SIP consisted of the finest particle size and minimal aggregation than other IP preparations. It is concluded that the IP formula prepared by the spray-drying method with PVP is the most effective approach to the improvement of bioavailability of IP.

        이프리플라본 고체분산체의 생체이용률에 미치는 폴리비닐피롤리돈의 분자량 및 혼합비율의 영향

        Ipriflavone (3-phenyl-7-isopropoxy-4H-1-benzopyran-4-one, IP) is a well-known antiosteoporotic drug with poor bioavailability. In the previous study, we reported that the IP formulation prepared by spray-drying method with polyvinylpymolidone (PVP) (SIP) was very effective in improving the bioavailability of IP. In this study, we examined the effects of molecular weight and mixture ratios of PVP to IP on the systemic absorption of IP following oral administration of SIP at a dose of 50 ㎎/㎏ to rats. In the effect of molecular weight, the Cmax of spray-dried IP with PVP K30 (SIP-K30) was significantly higher than those of spray-dried IP with PVP 360 (SIP-360), spray-dried IP with PVP K90 (SIP-K90), and spray-dried IP with PVP K17 (SIP-K17) (p<0.05). The AUC of SIP-K30 was about 2, 3, and 5.5 times higher than those of SIP-360, SIP-K90, and SIP-K17, respectively. The AUC value of SIP-K30 was significantly greater than those of SIP-K17 and SIP-K90 (p<0.05) except for SIP-360. In the ratio of PVP K30 to drug, the C_(max) and the AUC value of 3 : 7 IP-PVP solid dispersion were similar to those of 5 : 5 IP-PVP and significantly higher than those of the other solid dispersions (p<0.05). It was concluded that the spray-dried IP with PVP K30 at the ratio of 3:7 (w/w) was the best formulation for improving the bioavailablity of IP.

        Korean Medicine Treatments in Threepatients with Bell’s Palsy after Coronavirus Disease 2019 Infection: A Retrospective Case Series

        Bell’s palsy is an acute facial paralysis caused by peripheral facial nerve dysfunction. The aim of this study is to assess the efficacy of Korean medicine in the treatment of three patients who were hospitalized and diagnosed with Bell’s palsy within 2 weeks of coronavirus disease 2019 (COVID-19) infection. The patients were administered with Korean medicine treatments, steroids, and antiviral drugs. Moreover, the Korean medicine treatments include acupuncture, pharmacopuncture, moxibustion, physical therapy, and herbal medicine. Symptom improvement was evaluated daily using the Yanagihara facial nerve grading system, a facial function evaluation tool. Furthermore, it was suggested that the patients affected by Bell’s palsy after COVID-19 infection may have a slower improvement in their treatment progress compared with those without COVID-19 infection in the acute stage.

        Byproducts from Piggery Wastewater Treatment for the Sustainable Soil amendment and Crop Production

        Livestock manure is generally beneficial to soil and crop production when appropriate amount is applied, but excessive application may be detrimental to soil and water environments. A proper protocol of livestock waste treatment is required to manage the quality of soil and water. A trickling filter system using rice straw media was employed to treat piggery wastewater from small-scaled livestock farms as an alternative to the currently available methods. Batches of piggery wastewater were treated with this system, and the byproducts of rice straw media and trickling filtrate were applied to the soil with cultivating rye (Secale cereale L.). Objective of this research was to characterize these byproducts for the sustainable soil amendments and rye production. Both the treated straw medium and filtrate were proven to be effective organic fertilizers for rye plant development, with the enhanced but balanced absorption of nutrients. The synergistic effects of filtrate in addition to straw application did not show, but the filtrate appeared to lead to a higher water content of the plant. No specific nutrient deficiency or toxicity symptom was shown due to the salts derived from the byproducts applied. Chemical parameters of the soil quality were significantly improved with the application of straw medium either with or without the filtrate. Judging from parameters relating to the salt accumulations, such as sodium adsorption ratio (SAR), electrical conductivity (EC), exchangeable sodium percentage (ESP), potassium adsorption ratio (KAR), and residual P concentrations, the byproducts from piggery wastewater exhibited no detrimental effects on soil quality within the ranges of treatments used. In addition to the effectiveness of the rice straw trickling filter system for the small-scaled swine farms, both rice straw medium and filtrate could be recycled for the sustainable soil amendment and plant nutrition.

        Cultivation of Laminaria japonica (Laminariales, Phaeophyta) in Udo Coast, Jeju, Korea

        In order to produce Laminaria japonica in Jeju as feed for abalone, a transplant experiment was performed with the Baekryungdo and the Wando cultivar stocks at the Udo aquafarm from January to July 2003. Eight water conditions at the aquafarm and eleven traits of the two cultivar stocks were measured once a month. The water temperature ranged from 12.9$^{\circ}C$ to 23.5$^{\circ}C$. The salinity was 31.3-36.8‰ the DO was 5.40-9.86 mg ${\cdot}$ $l^{-1}$ the pH was 7.82-9.61. Concentrations of $NO_2-N,\;NO_3-N,\;NH_4-N\;and\;PO_4-P$ were 0.02-0.15 $\mu$M, 2.27-3.49 $\mu$M, 0.16-0.56 $\mu$M and 0.07-0.99 $\mu$M, respectively. The whole frond length of the Baekryungdo and the Wando specimens were 173.84 and 153.67 cm. The blade width, stipe length, blade length, fascia length, blade thickness, total weight and substantiality of the Baekryungdo and the Wando specimens were 14.61 and 13.05 cm, 3.94 and 3.02 cm, 169.88 and 150.65 cm, 155.81 and 137.10 cm, 1.01 and 0.96 cm, 258.04 and 200.96 cm, and 101.56 and 94.62, respectively at the conclusion of the experiment in July. The measurements of the Baekryungdo specimens were slightly higher than those of Wando specimens. The fascia width, stipe thickness and fascia thickness of the Wando specimens were slightly higher than those of the Baekryungdo specimens. The relative growth rates of these traits of two cultivar stocks during the growth test were very similar. Generally, the performance of the Baekryungdo cultivar stock seems to be better than that of the Wando samples. Further study of the physiological ecology for cultivation and breeding is needed in the future.

        Fertility Assessment of the Piggery Wastewater Trickling Filtrate for Orchard Grass(Dactylis glomerata L.) and Soil

        In search of a method to achieve sustainable agricultural practices, a trickling filter was employed for the piggery wastewater treatment, where rice straw was the support medium in place of more commonly used materials. The filtrate from the trickling filter were applied to a soil, on which orchard grass (Dactylic glomerata L.) was grown followed by amendment of the treated straw medium. Orchard grass was cut twice, and growth parameters and yields were measured. Soil chemical properties before and after harvesting orchard grass were analyzed. Development of the forage crop was greatly enhanced by the application of the filtrate in terms of fresh weight, dry weight, and the absorption of nutrients. Better growth and higher nutrient uptake were found in the second cutting of the orchard grass. Organic matter content, ionic intensity, exchangeable cations, and phosphate of the soil increased with the application of the filtrate. The relative high concentrations of salts in the piggery trickling filtrate, expressed in electrical conductivity and content of sodium, exerted no detrimental effect on the crop and soil.
