        Antibody-Mediated Resistance to Rhizomania Disease in Sugar Beet Hairy Roots

        Agrobacterium rhizogenes-mediated transformation of sugar beet hairy roots expressing single-chain variable fragment (scFv) was exploited to evaluate the efficacy of four antibody-based constructs for interfering with the Beet necrotic yellow vein virus infection. The scFv specific to a major coat protein of virus, p21, was targeted to various cellular compartments including the cytosol (pIC and pICC constructs), apoplast (pIA), and mitochondrion (pIM). After mechanical virus inoculation, most of the hairy root clones expressing scFv in the cytosol displayed low virus titers while the majority of transgenic hairy root clones accumulated antibody in outer membrane of mitochondria or apoplast were infected. This hairy root system provided an efficient and rapid approach to initially investigating root disease resistance like rhizomania prior to transform whole recalcitrant plants such as sugar beet.

        Influence of Culture Conditions and Medium Composition on the Production of Antibacterial Compounds by Marine Serratia sp. WPRA3

        This study was undertaken to investigate the influence of culture conditions and medium components on production of antibacterial compounds by Serratia sp. WPRA3 (JX020764) which was isolated from marine water of Port Dickson, Malaysia. Biochemical, morphological, and molecular characteristics suggested that the isolate is a new candidate of the Serratia sp. The isolate showed strong antimicrobial activity against fungi, Gram-negative and Gram-positive bacteria. This bacterium exhibited optimum antibacterial compounds production at 28°C, pH 7 and 200 rev/min aeration during 72 h of incubation period. Highest antibacterial activity was obtained when sodium chloride (2%), yeast extract (0.5%), and glucose concentration (0.75%) were used as salt, nitrogen, and carbon sources respectively. Different active fractions were obtained by Thin-Layer Chromatography (TLC) and Flash Column Chromatography (FCC) from ethyl acetate crude extracts namely OCE and RCE in different culture conditions, OCE (pH 5, 200 rev/min) and RCE (pH 7/without aeration). In conclusion, the results suggested different culture conditions have a significant impact on the types of secondary metabolites produced by the bacterium.

        Liquid+liquid equilibria of ternary water+carboxylic acid+solvent systems at 288.15K

        The experimental liquid-liquid equilibrium (LLE) data for six ternary systems containing (chloroform+propionic acid+water), (chloroform+acetic acid+water), (diethyl ether+propionic acid+water), (diethyl ether+acetic acid+water), (trichloroethylene+propionic acid+water) and (trichloroethylene+acetic acid+water) were measured at 288.15 K and at atmospheric pressure. An accurate and simple titration method was used for determining of the concentration of carboxylic acid in the both liquid phases at equilibrium. The reliability of the experimental tie-line data was confirmed by using the Othmer-Tobias correlation. The distribution coefficients and selectivity factors were presented to evaluate the efficiency of the solvents for extraction of carboxylic acid from water. The results show that chloroform and diethyl ether are satisfactory solvents for extraction of carboxylic acids from water. Trichloroethylene separates propionic acid from water; however, it cannot be used as a solvent for separation of acetic acid.
