The two spotted mite, Tetranychus urticae Koch, is a global pest, and has developed severe resistance to several types of acaricides. Acequinocyl-resistant strain and bifenazate-resistant strain of T. urticae showed higher relative activity level of glutathion-S-transferases (GSTs) than susceptible strain. Studies of resistance in T. urticae have focused on classical detoxifying enzyme system, such as GSTs. We examined the expression of six GSTs genes in T. urticae belong to five different cytosolic classes, including two in delta, one in omega, one in mu, one in zeta, and one mitochondrial GSTs class, kappa. RT-qPCR results showed that, among these six GSTs genes, the relative expression level of delta 1 was significantly higher in bifenazate-resistant strain than in other strains (susceptible, abamectin, acequinocyl, etoxazole and pyridaben strain). Expression level of the delta 2 was also higher in acequinocyl-resistant strain than in other strains. The elevated GSTs gene transcripts following exposure to acequinocyl, bifenazate might be one of the mechanisms involved in detoxification of these acaricide. These results may be caused by different mechanisms in the resistance development of T. urticae to these two different types of acaricides.

The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is one of the most important pest species devastating many horticultural and ornamental crops and fruit trees. In the present study, a field strain was collected in January 2001 and has been selected for sixteen years with acequinocyl. The resistance ratio calculated from the LC50 of eggs and adults was 191- and 4,237-fold, respectively. The laboratory-selected acequinocyl resistance (LSAR16) strain was screened with 11 acaricides for cross-resistance. The detoxifying enzyme activities and quantitative real-time PCR analysis were performed in the LSAR16 strain. Crossing experiments revealed that LSAR16 strain was inherited maternally, incompletely dominant and monogenic. Most importantly, we identified two new point-mutations at mitochondrial cytb from acequinocyl resistant T. urticae.

We performed a survey for flavivirus infection and distribution of Aedes albopictus that known as Zika and Dengue virus vector using black–light trap and BG-sentinel trap around urban area in Korea. Mosquitoes were collected in 27 cities during March to November (twice a month) year 2016. Total numbers of mosquitoes collected 102,102 including 19 species 8 genera during collecting period. Total 21,467 Ae. albopictus was collected that 20,961(24.3%) by BG-sentinel trap and 506 (3.2%) by Black-light trap in urban area. Trap index(trap/night) of Ae. albopictus was showed highest in Hamyang (TI:992.3) and lowest in Taebaek (TI:0.3) there was only collected by Black-light trap. A total of 894 pools from all collecting Ae. albopictus were performed a Flavivirus detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases (include Zika virus) and vectors in Korea.
Novel JAK1-selective benzimidazole inhibitors with enhanced membrane permeability

<P><B>Abstract</B></P> <P>The previously identified Janus kinase 1 (JAK1)-selective inhibitor, 1-(2-aminoethyl)-2-(piperidin-4-yl)-1<I>H</I>-benzo[<I>d</I>]imidazole-5-carboxamide (<B>2</B>), suffered from low cell permeability, which resulted in poor pharmacokinetic properties. In this study, by introducing less polar hydrogen bond donors at <I>N</I> <SUP>1</SUP> (a hydroxyalkyl or a methylaminoalkyl group) and C2 (a cyclohexanol group) positions, a series of novel benzimidazole derivatives were prepared, which exhibited selective JAK1 inhibitory activity (IC<SUB>50</SUB> against JAK1=0.08–0.15μM; JAK1-selectivity=26–40 fold vs JAK2, 12–23 fold vs JAK3, and 38–54 fold vs Tyk2) along with significantly increased lipophilicity (3.3–15.8 times) as well as membrane permeability (6.3–12 times).</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
Benzimidazole Derivatives as Potent JAK1-Selective Inhibitors

<P>The Janus kinase (JAK) family comprises four members (JAK1, JAK2, JAK3, and Tyk2) that play a key role in mediating cytokine receptor signaling. JAK inhibition thus modulates cytokine-mediated effects. In particular, selective inhibition of JAK1 or JAK3 may provide an efficient therapeutic agent for the treatment of inflammatory diseases, with minimized side effects. In this study, as part of our continued efforts to develop a selective JAK1 inhibitor, a series of 1,2-disubstituted benzimidazole-5-carboxamide derivatives was prepared and their inhibitory activities against all four JAK isozymes were evaluated. A clear structure–activity relationship was observed with respect to JAK1 selectivity; this highlighted the importance of hydrogen bond donors at both <I>N</I><SUP>1</SUP> and R<SUB>2</SUB> positions located within a specific distance from the benzimidazole core. One of the synthesized compounds, 1-(2-aminoethyl)-2-(piperidin-4-yl)-1<I>H</I>-benzo[<I>d</I>]imidazole-5-carboxamide (<B>5c</B>), showed remarkable JAK1 selectivity (63-fold vs JAK2, 25-fold vs JAK3, and 74-fold vs Tyk2). Molecular docking revealed that the 2-aminoethyl and piperidin-4-yl substituents of <B>5c</B> function as probes to differentiate the ATP-binding site of JAK1 from that of JAK2, resulting in preferential JAK1 binding. <P>Defects in crystals such as antisites generally lead-to the deterioration of the ionic conductivity of solid-state ionic conductors. Herein, using first principles calculations, we demonstrate that the Li diffusion in Li2MP2O7 (M = Fe or Mn), a promising battery material, is sensitively affected by the presence of Li/M antisites; however, unexpectedly, the antisites significantly promote Li diffusion. The calculations reveal that the presence of antisites reduces the barrier of Li hopping and opens new paths for Li diffusion in the Li2MP2O7 crystal. In our experimental verification, we succeeded in synthesizing crystalline Li2MnP2O7 with varying Li/Mn antisite contents and demonstrated that the inclusion of antisites results in improved power capability with faster Li diffusion for Li-ion battery electrodes. We believe that this unexpected finding of increasing the ionic conductivity by introducing antisite defects broadens our understanding of solid-state ionic conductors and provides a new strategy for improving Li diffusion in conventional electrode materials for Li rechargeable batteries.</P>
A SCORM-based e-Learning Process Control Model and Its Modeling System

In this paper, we propose an e-Learning process control model that aims to graphically describe and automatically generate the manifest of sequencing prerequisites in packaging SCORM`s content aggregation models. In specifying the e-Learning activity sequencing, SCORM<sup>1</sup> provides the concept of sequencing prerequisites to be manifested on each e-Learning activity of the corresponding tree-structured content organization model. However, the course developer is required to completely understand the SCORM`s complicated sequencing prerequisites and other extensions. So, it is necessary to achieve an efficient way of packaging for the e-Learning content organization models. The e-Learning process control model proposed in this paper ought to be an impeccable solution for this problem. Consequently, this paper aims to realize a new concept of process-driven e-Learning content aggregating approach supporting the e-Learning process control model and to implement its e-Learning process modeling system graphically describing and automatically generating the SCORM`s sequencing prerequisites. Eventually, the proposed model becomes a theoretical basis for implementing a SCORM-based e-Learning process management system satisfying the SCORM`s sequencing prerequisite specifications. We strongly believe that the e-Learning process control model and its modeling system achieve convenient packaging in SCORM`s content organization models and in implementing an e-Learning management system as well.
Physician-Directed Diabetes Education without a Medication Change and Associated Patient Outcomes

<P><B>Background</B></P><P>When patients with diabetes mellitus (DM) are first referred to a hospital from primary health care clinics, physicians have to decide whether to administer an oral hypoglycemic agent (OHA) immediately or postpone a medication change in favor of diabetes education regarding diet or exercise. The aim of this study was to determine the effect of diabetes education alone (without alterations in diabetes medication) on blood glucose levels.</P><P><B>Methods</B></P><P>The study was conducted between January 2009 and December 2013 and included patients with DM. The glycosylated hemoglobin (HbA1c) levels were evaluated at the first visit and after 3 months. During the first medical examination, a designated doctor also conducted a diabetes education session that mainly covered dietary management.</P><P><B>Results</B></P><P>Patients were divided into those who received no diabetic medications (<I>n</I>=66) and those who received an OHA (<I>n</I>=124). Education resulted in a marked decrease in HbA1c levels in the OHA group among patients who had DM for <1 year (from 7.0%±1.3% to 6.6%±0.9%, <I>P</I>=0.0092) and for 1 to 5 years (from 7.5%±1.8% to 6.9%±1.1%, <I>P</I>=0.0091). Those with DM >10 years showed a slightly lower HbA1c target achievement rate of <6.5% (odds ratio, 0.089; <I>P</I>=0.0024).</P><P><B>Conclusion</B></P><P>For patients who had DM for more than 5 years, higher doses or changes in medication were more effective than intensive active education. Therefore, individualized and customized education are needed for these patients. For patients with a shorter duration of DM, it may be more effective to provide initial intensive education for diabetes before prescribing medicines, such as OHAs.</P>
