      • Association of Matrix Metalloproteinase (MMP)-2 and -9 Expression with Extra-gastrointestinal Stromal Tumor Metastasis

        Matrix metalloproteinase (MMP)-2 and MMP-9 are important proteases involved in invasion and metastasis of various tumors. Extra-gastrointestinal stromal tumors (EGISTs) are rare neoplasms. This study was performed to assess MMP-2 and MMP-9 expression in EGIST tissue samples for association with clinicopathological data from the patients. Twenty-one surgical EGIST tissue specimens were collected for analysis of MMP-2 and MMP-9 expression using immunohistochemistry. MMP-2 and MMP-9 proteins were expressed in all of the epithelial cell types of EGISTs, whereas they were only expressed in 75% of the spindle cell type, although there was no statistically significant difference (p>0.05). Expression of MMP-2 and MMP-9 proteins was associated with tumor size, mitotic rate, tumor necrosis, and distant metastasis (p<0.05). MMP-2 expression was linked with MMP-9 levels (p<0.05). However, there was no correlation between MMP-9 expression and age, sex, primary site, or cell morphology in any of these 21 EGIST patients (p>0.05). Moreover, expression of MMP-2 and MMP-9 proteins increased with the degree of EGIST risk. This study provided evidence of an association of MMP-2 and MMP-9 expression with advanced EGIST behavior.

        Deuterium Clusters Fusion Induced by the Intense Femtosecond Laser Pulse

        <P>Neutrons (2.45 MeV) from deuterium cluster fusion induced by the intense femtosecond (30 fs) laser pulse are experimentally demonstrated. The average neutron yield 10<SUP>3</SUP> per shot is obtained. It is found that the yield slightly increases with the increasing laser spot size. No neutron can be observed when the laser intensity I < 4.3×10<SUP>15</SUP> W/cm<SUP>2</SUP>.</P>

        Robust Multi-mode Flight Control Design for an Unmanned Helicopter Based on Multi-loop Structure

        In this paper, a novel multi-mode flight control strategy for unmanned helicopter, in presence of model uncertainty, atmospheric disturbances and handling qualities specification requirements (as in ADS-33E), based on multi-loop control structure combining robust H-infinity and PI control is presented. In inner loop H-infinity optimal control technique is utilized ensuring the stability of flight control system in case of change of helicopter dynamics, model uncertainties and eliminates effect of gust disturbance on helicopter states and collective/cyclic inputs. PI control in outer loop is used to improve the dynamic and static operation characteristics. Attitude control and attitude holding flight mode with satisfactory command response and decoupling characteristics is designed using the proposed control strategy. Analysis and simulation results show that Level 1 handling requirements as defined in ADS-33E are accomplished even when helicopter is under constant wind circumstance.

      • Folding Graphene Film Yields High Areal Energy Storage in Lithium-Ion Batteries

        <P>We show that a high energy density can be achieved in a practical manner with freestanding electrodes without using conductive carbon, binders, and current collectors. We made and used a folded graphene composite electrode designed for a high areal capacity anode. The traditional thick graphene composite electrode, such as made by filtering graphene oxide to create a thin film and reducing it such as through chemical or thermal methods, has sluggish reaction kinetics. Instead, we have made and tested a thin composite film electrode that was folded several times using a water-assisted method; it provides a continuous electron transport path in the fold regions and introduces more channels between the folded layers, which significantly enhances the electron/ion transport kinetics. A fold electrode consisting of SnO<SUB>2</SUB>/graphene with high areal loading of 5 mg cm<SUP>–2</SUP> has a high areal capacity of 4.15 mAh cm<SUP>–2</SUP>, well above commercial graphite anodes (2.50–3.50 mAh cm<SUP>–2</SUP>), while the thickness is maintained as low as ∼20 μm. The fold electrode shows stable cycling over 500 cycles at 1.70 mA cm<SUP>–2</SUP> and improved rate capability compared to thick electrodes with the same mass loading but without folds. A full cell of fold electrode coupled with LiCoO<SUB>2</SUB> cathode was assembled and delivered an areal capacity of 2.84 mAh cm<SUP>–2</SUP> after 300 cycles.         Discovery of novel glycoside hydrolases from C-glycoside-degrading bacteria using sequence similarity network analysis

        C-Glycosides are an important type of natural product with significant bioactivities, and the C-glycosidic bonds of C-glycosides can be cleaved by several intestinal bacteria, as exemplified by the human faeces-derived puerarin-degrading bacterium Dorea strain PUE. However, glycoside hydrolases in these bacteria, which may be involved in the C-glycosidic bond cleavage of C-glycosides, remain largely unknown. In this study, the genomes of the closest phylogenetic neighbours of five puerarin-degrading intestinal bacteria (including Dorea strain PUE) were retrieved, and the protein-coding genes in the genomes were subjected to sequence similarity network (SSN) analysis. Only four clusters of genes were annotated as glycoside hydrolases and observed in the genome of D. longicatena DSM 13814T (the closest phylogenetic neighbour of Dorea strain PUE); therefore, genes from D. longicatena DSM 13814T belonging to these clusters were selected to overexpress recombinant proteins (CG1, CG2, CG3, and CG4) in Escherichia coli BL21(DE3). In vitro assays indicated that CG4 efficiently cleaved the O-glycosidic bond of daidzin and showed moderate β-D-glucosidase and β-D-xylosidase activity. CG2 showed weak activity in hydrolyzing daidzin and pNP- β-D-fucopyranoside, while CG3 was identified as a highly selective and efficient α-glycosidase. Interestingly, CG3 and CG4 could be selectively inhibited by daidzein, explaining their different performance in kinetic studies. Molecular docking studies predicted the molecular determinants of CG2, CG3, and CG4 in substrate selectivity and inhibition propensity. The present study identified three novel and distinctive glycoside hydrolases, highlighting the potential of SSN in the discovery of novel enzymes from genomic data.

        Domain of Unknown Function 143 is required for the functioning of PEP-associated protein DG238 in the chloroplast

        Plastid-encoded plastid RNA polymerase (PEP) isessential for chloroplast development and plastid geneexpression in Arabidopsis thaliana. However, PEP is a largecomplex, and many proteins in this complex remain to beidentified. We previously reported that Delayed Greening238 (DG238) interacts with PEP subunit protein FLN1 andmay function as a PEP-associated protein and participate inearly chloroplast development and PEP-dependent plastidgene expression. DG238 contains Domain of UnknownFunction 143 (DUF143), whose function is currently unknown. Here, we found that a deficiency of the DUF143 domain inDG238 affected its localization, which resulted in abnormalinteractions with PEP-associated proteins in the chloroplast. Furthermore, DG238 lacking the DUF143 domain or DG238with only this domain failed to function. Interestingly, thelack of conserved amino acids 193–217 of the DUF143domain in DG238 also affected its function. In addition toFLN1, DG238 also interacts with other PEP-associatedproteins, including FSD2, FSD3, MRL7-L, and MRL7, toregulate plastid gene expression. These results suggest thatthe DUF143 domain is necessary for the functioning of thePEP-associated protein DG238 in chloroplasts.

        <P><B>Abstract</B></P><P>High‐spin iron(III) iodosylarene complexes bearing an N‐methylated cyclam ligand are synthesized and characterized using various spectroscopic methods. The nonheme high‐spin iron(III) iodosylarene intermediates are highly reactive oxidants capable of activating strong CH bonds of alkanes; the reactivity of the iron(III) iodosylarene intermediates is much greater than that of the corresponding iron(IV) oxo complex. The electrophilic character of the iron(III) iodosylarene complexes is demonstrated in sulfoxidation reactions.</P>

        Expression and regulation of Angiopoietins and their receptor Tie-2 in sika deer antler

        The cartilage vascularization and chondrocyte survival are essential for endochondral ossification which occurs in the process of antler growth. Angiopoietins (Ang) is a family of major angiogenic growth factors and involved in regulating the vascularization. However, the expression and regulation of Angs in the antler are still unknown. The aim of this study is to localize the expression of Ang-1, Ang-2 and their receptor Tie-2 in sika deer antler using in situ hybridization and focused on analyzing the regulation of testosterone, estrogen, all-trans-retinoic acid (ATRA) and 9cRA on their expression in antler chondrocytes. The results showed that Ang-1, Ang-2 and Tie-2 were highly expressed in antler chondrocytes. Administration of testosterone to antler chondrocytes led to a notable increase in the expression of Ang-1 and Tie-2, and a reduction in the expression of Ang-2. The similar result was also observed after estrogen treatment. In contrast, ATRA and 9cRA could inhibit the expression of Ang-1 in antler chondrocytes and heighten the expression of Ang-2. Simultaneously, ATRA could downregulate the expression of Tie-2 in antler chondrocytes at 12 and 24 h, while 9cRA upregulate the expression of Tie-2 at 3 and 6 h. Collectively, Ang-1, Ang-2 and Tie-2 are expressed in antler chondrocytes and their expression can be affected by testosterone, estrogen, ATRA and 9cRA.

      • Effects of Ribosomal Protein L39-L on the Drug Resistance Mechanisms of Lung Cancer A549 Cells

        Background: Cancer is a major threat to the public health whether in developed or in developing countries. As the most common primary malignant tumor, the morbidity and mortality rate of lung cancer continues to rise in recent ten years worldwide. Chemotherapy is one of the main methods in the treatment of lung cancer, but this is hampered by chemotherapy drug resistance, especially MDR. As a component of the 60S large ribosomal subunit, ribosomal protein L39-L gene was reported to be expressed specifically in the human testis and human cancer samples of various tissue origins. Materials and Methods: Total RNA of cultured drug-resistant and susceptible A549 cells was isolated, and real time quantitative RT-PCR were used to indicate the transcribe difference between amycin resistant and susceptible strain of A549 cells. Viability assay were used to show the amycin resistance difference in RPL39-L transfected A549 cell line than control vector and null-transfected A549 cell line. Results: The ribosomal protein L39-L transcription level was 8.2 times higher in drug-resistant human lung cancer A549 cell line than in susceptible A549 cell line by quantitative RT-PCR analysis. The ribosomal protein L39-L transfected cells showed enhanced drug resistance compared to plasmid vector-transfected or null-transfected cells as determined by methyl tritiated thymidine (3H-TdR) incorporation. Conclusions and Implications for Practice: The ribosomal protein L39-L gene may have effects on the drug resistance mechanism of lung cancer A549 cells.

      • A Dependence Stability Bound based on the VC Dimension for Relational Classification

        Relational classification (RC) is concerned with the application of statistical learning to relational data. RC models do not have improved stability to smooth the perturbations generated by variations in the correlation between the relational data. Therefore, few studies have attempted to derive a bound and develop a stability learning framework for RC models. To solve this problem, we derive a learning bound with a new measure dependence stability and a limited Vapnik–Chervonenkis (VC) dimension. Based on the learning bound, we then design a stable learning framework that serves as a guideline for the development of new learning algorithms for a broad class of RC models. Applying a Markov logic network on synthesized and real-world datasets, our experimental results demonstrate that our bound can be tight if the RC model has appropriate dependence stability and limited VC dimension and our learning framework increases the stability of RC models while reducing the deviation between empirical risk and true risk.
