      • Arg 1+ TAM Contributes Tuberculous Fibrosis- Induced Lung Cancer Progression via Autophagy Signaling

        Background The association between previous tuberculous fibrosis and lung cancer has been reported in some epidemiologic and experimental studies, but the detailed mechanisms remain unclear. Tumor associated macrophages (TAMs) are known to play key roles in tumor spread and efficacy of anticancer drugs. Methods We investigated the role of M2 Arg 1+ macrophage in tuberculous pleurisy assisted tumorigenicity both in vitro and in vivo. Bone marrow-derived macrophages (BMMs) from WT C57BL/6 mice were stimulated with tuberculous pleural effusion (TPE) for 24 hour and cellular interactions between macrophage and cancer cell were evaluated. A murine model injected with cancer at 4 weeks after Mycobacterium bovis bacillus Calmette-Guerin (BCG) pleural infection were used to validate the contribution of tuberculous fibrosis to tumor invasion. Results CXCL 9 and CXCL 10 in TPE induced M2 Arg 1+ polarization of murine BMMs. TPE educated macrophages facilitated lung cancer proliferation and migration in vitro and lung cancer after treatment of conditioned medium of TPE-educated macrophage exhibited elevated autophagy signaling, increased E-cadherin and decreased caspase-3. Pharmacological inhibition of arginase 1 decreased autophagy signaling and E-cadherin. In accordance with our earlier study, BCG pleural infection enhanced the tumor growth in lung metastasis mouse model. Arginase-1 inhibitor suppressed tuberculous fibrosispromoted metastatic potential of lung cancer, decreased autophagy signaling and E-cadherin expression. Conclusions In conclusion, our study indicates that tuberculosis pleural fibrosis induces M2 Arg1+ polarization and M2 Arg1+ TAM contributes lung cancer metastasis via autophagy signaling and E-cadherin signaling. These Results suggest that M2 Arg 1+ TAM may be a novel therapeutic target for tuberculous fibrosis- induced lung cancer.

        <P>Conclusions: Our results suggest that the overproduction of calpain-1 and SBDP is associated with pleural fibrosis in tuberculous pleurisy. While ADA is a conventional marker for diagnostic TPE, the simultaneous measurement of calpain-1 and SBDP 1 in pleural fluid may improve the diagnostic efficacy.</P>

        Study of the Primo-Vascular System and Location-Dependent Oxygen Levels for a Mouse Embryo

        <P>The two major circulatory systems, the lymph system and the blood vessel system, play significant roles in controlling embryonic development. The primo-vascular system (PVS) was recently reported as an additional circulatory system in various animals. In this paper, the PVS in a mouse embryo was investigated. The structural characterization of the PVS in the mouse placenta and umbilical cord, which was visualized with the trypan blue staining technique, was focused on. The PVS was well_developed in the mouse placenta area. Using a nanopore-based amperometric oxygen sensor, the oxygen levels at four different areas of the embryonic brain, placenta, blood vessel, and primo-vessel of the PVS were measured. The relatively higher oxygen levels that were measured at the primo-vessels than at the brain and the placenta, while still lower than the oxygen level that was measured at the blood vessels, may suggest a role of PVS in oxygen transport.</P>

        <P>The distribution of partial oxygen pressure (pO<SUB>2</SUB>) is analyzed for the anterior aspect of the left wrist with an amperometric oxygen microsensor composed of a small planar Pt disk-sensing area (diameter = 25 <I><I>μ</I></I>m). The pO<SUB>2</SUB> levels vary depending on the measurement location over the wrist skin, and they are systematically monitored in the analysis for both one-dimensional single line (along the wrist transverse crease) and two-dimensional square area of the wrist region. Relatively higher pO<SUB>2</SUB> values are observed at certain area in close proximity to the position of acupuncture points with statistical significance, indicating strong relationship between oxygen and acupuncture point. The used oxygen microsensor is sensitive enough to detect the pO<SUB>2</SUB> variation depending on the location. This study may provide information helpful to understand possible physiological roles of the acupuncture points.</P>

        A Multilevel Examination of the Influence of Content Domain Choice on Attitudes toward Convergence and Science Teaching Efficacy Beliefs in a TPACK Training Program for Elementary Teacher Candidates

        이 연구는 초등학교 예비교사를 대상으로 TPACK 훈련 프로그램을 적용하는 과정에서 융합수업 교수-학습 계획을 위해 생명 영역 또는 지구 영역을 선택하도록 하고, 그 선택의 여부와 종류가 예비교사의 태도 및 교수효능감에 어떤 영향을 미치는가를 알아보았다. 이 연구의 결과를 통하여 TPACK 프로그램에 대한 전반적인 영향, 내용 영역의 선택 유무 따른 TPACK 프로그램의 효과의 차이, 내용 영역의 종류에 따른 효과 차이를 확인하였다. 이를 위해 초등학교에서 과학과 내용을 중심으로 테크놀로지를 융합한 수업을 할 수 있는 교사를 양성하기 위한 목적의 TPACK 프로그램을 194명의 교육대학생을 대상으로 2개학년도 2회에 걸쳐 각각 8주간 적용하였다. 이 연구에서 TPACK 프로그램의 적용은 전체 연구참여자 집단의 융합에 대한 태도과 과학 교수 효능감에 유의한 영향을 주었다. 그러나 내용 영역 선택 여부에 따른 실험 집단과 비교집단을 대조한 결과는 상승 정도에 따른 집단 간의 유의한 차이가 없었다. 뿐만 아니라 실험 집단의 하위 집단을 생명 영역 및 지구와 우주 영역으로 구분한 내용 영역의 종류에 따라 비교한 결과에서도 사후 검사에서 융합에 대한 태도 및 과학 교수 효능감 점수에 차이를 보이지 않았다. This study investigated the effect of a technological, pedagogical, and content knowledge (TPACK) program on teacher candidates’ attitudes and self-efficacy and the influence of an opportunity to choose a content domain between either life science or earth science. The research questions were designed to ascertain the overall effect of the TPACK program, the different consequences of the choice of the content domain, and the distinct influences of the two content domain types. With this objective, we implemented the TPACK program with 194 elementary teacher candidates for a total of 16 weeks during two academic years. The training program embodied teaching methods to conduct science content-based lessons in technology-integrated context. This study showed that the TPACK program impacted participating elementary school teacher candidates in terms of their attitudes toward convergence and science teaching efficacy beliefs. However, no significant differences were observed between the intervention group 2, in which teachers could choose their content domain, and the intervention group 1 in the improvement. Furthermore, there were no significant differences between the subgroups of the intervention group 2 based on the selected content domain type, i.e., life science or earth science, in the posttest scores for attitudes toward convergence or science teaching efficacy beliefs.

        작은 생물 서식 환경에 대한 그리기와 연상 단어 분석을 통한 초등학생의 인식 연구

        The purpose of this study was to investigate elementary students conceptions of the environments that small organisms can inhabit, through picture drawing and associated words about the organisms. The participants were 449 elementary students from grades 5th and 6th living in J province. The results showed that the largest proportion of the students answered small-sized animals and/or plants as associated words about small organisms. Students conceptions of the habitats in which small organisms live were related to four categories, the ground, the hydrosphere, the living environment, and everywhere, although the ground environment was described most often. The negative perceptions of the influence of microorganisms on humans were revealed in aspects such as disease and health, decay, and positive perceptions were related to technological applications regarding food production and medical and environmental uses. The group of students also had beneficial and detrimental perceptions about the influence of small sized organisms without a one-sided inclination. It is inferred that elementary school students who associate with words about microorganisms tend to perceive a greater variety of places as inhabitable environments, because there was a positive correlation between microorganisms and the entire surrounding environment. The analysis of this study showed that elementary school students depend on their personal experiences regarding the habitats of small organisms. These results could contribute to developing a curriculum concerning small organisms and the environments in which they live within the ecosystem.

        본 연구는 초등학생 5∼6학년을 대상으로 그림 그리기 및 연상 단어를 통해서 작은 생물의 서식 환경에 대한 이미지를 조사하였다. 연구 결과, 초등학생들은 작은 생물에 대한 연상 단어로서 작은 규모의 동식물을 가장 높은 비율로 답하였으며, 작은 생물의 서식 환경에 대한 이미지로 ‘땅’을 가장 빈번하게 묘사하였다. 또한 작은 생물과 우리 생활과의 관계를 질병과 건강, 부패, 기술적 활용의 범주로 나누어 분석한 결과, 학생들은 미생물의 긍정적인 측면과 부정적인 측면을 모두 인식하고 있었고, 작은 동식물에 대해서도 이로움과 해로움을 비슷한 비율로 인식하고 있는 것으로 나타났다. 작은 생물에 대하여 미생물을 연상하는 학생들이 서식 환경을 ‘어디에서나’ 살고 있다고 인식하는 상대적인 경향이 있는 것으로 보아, 작은 생물을 동식물에 국한하기보다 미생물까지 연상하는 경우에 서식 환경도 다양한 사례로 인식한다고 생각된다. 본 연구의 결과는 앞으로 초등학교의 작은 생물 교육 및 환경 교육을 어떻게 접근해야 하는지에 대한 시사점을 제공할 수 있을 것이다.       • 토양미생물을 활용한 탐구 학습이 초등과학 영재학생에게 미치는 영향

        본 연구는 토양미생물에 대한 탐구 프로그램을 개발하고 적용하여 융합인재소양, 창의적 문제해결력, 정의적 영역에 어떤 영향을 미치는지 알아보았다. 개발한 프로그램의 적용은 J대학교 과학영재교육원 2개 영재학급 34명 및 사사과정 5명의 과학영재를 대상으로 하였다. 토양미생물의 탐구 프로그램 수업은 융합인재소양, 창의적 문제해결력, 정의적 영역에 대하여 모두 긍정적인 영향을 미쳤다 (p<.001). 학생들이 실험 과정 동안에 탐구 학습이 흥미로웠고 도움이 되었다는 면담을 통해, 본 연구는 토양미생물이 과학 교육의 학습 소재로서 활용될 수 있음을 입증하였다. The purpose of the present study was to examine effects of an inquiry program regarding soil microorganisms on STEAM literacy, creative problem solving ability and affective domain. The program was applied to 34 gifted students from J institute of science education and 5 gifted students from R&E activity for 9 months. The developed soil microorganisms inquiry lessons affected on STEAM literacy, creative problem solving ability and affective domain to gifted students (p<.001). The results of this study suggest that soil microorganisms could be used for learning matter in science education because the students involved this study answered the inquiry learning was interesting and helpful through their experiments process.
