      • Comparative Proteomics Analysis of Colorectal Cancer

        Background and Objective: Protein expression in colon and rectal cancer (CRC) and paired normal tissues was examined by two-dimensional gel electrophoresis (2-DE) to identify differentially expressed proteins. Materials and Methods: Five fresh colorectal cancer and paired adjacent normal tissues were obtained and differentially expressed protein spots were determined using PDQuest software, with identification on the basis of MALDI-TOF mass spectra. Results: Compared with normal colorectal mucosa, protein abnormal expression of 65 spots varying more than 1.5 times were found in 2-DE gels from colorectal cancer samples (P<0.05); forty-two proteins were up-regulated and 23 were down-regulated; twelve protein spots were identified using mass spectrometry, of which 8 were up-regulated, includimng HSPB1and Annexin A4, while 4 were down-regulated, the results being consistent with Western blot findings. Conclusions: Two-dimensional electrophoresis reference maps for CRC tissues and adjacent normal mucosa (NMC) were established and 12 differentially expressed proteins were identified. Up-regulated HSPB1 and Annexin A4 may play many important roles in the pathogenesis of colorectal cancer.

        Myeloid-Derived Suppressor Cells Are Associated with Viral Persistence and Downregulation of TCRζ Chain Expression on CD8+ T Cells in Chronic Hepatitis C Patients

        Myeloid-derived suppressor cells (MDSCs) play an important role in impairing the function of T cells. We characterized MDSCs in two chronic hepatitis C (CHC) cohorts: a cross-sectional group that included 61 treatment-naive patients with CHC, 14 rapid virologic response (RVR) cases and 22 early virologic response (EVR) cases; and a longitudinal group of 13 cases of RVR and 10 cases of EVR after pegylated-interferon-α/ribavirin treatment for genotype 1b HCV infection. Liver samples from 32 CHC patients and six healthy controls were subjected to immunohistochemical analysis. MDSCs frequency in treatmentnaive CHC was significantly higher than in RVR, EVR, or healthy subjects and was positively correlated with HCV RNA. Patients infected with HCV genotype 2a had a significantly higher frequency of MDSCs than those infected with genotype 1b. Decreased T cell receptor (TCR) ζ expression on CD8+ T cells was significantly associated with an increased frequency of MDSCs in treatment-naive CHC patients and was restored by L-arginine treatment in vitro. Increased numbers of liver arginase-1+ cells were closely associated with the histological activity index in CHC. The TCR ζ chain was significantly downregulated on hepatic CD8+ T cells in CHC. During antiviral follow up, MDSCs frequency in peripheral blood mononuclear cells was directly correlated with the HCV RNA load in the plasma and inversely correlated with TCR ζ chain expression in CD8+ T cells in both RVR and EVR cases. Notably, the RVR group had a higher frequency of MDSCs at baseline than the EVR group. Collectively, this study provides evidence that MDSCs might be associated with HCV persistence and downregulation of CD8 ζ chain expression.

        Lung-Targeting Delivery of Dexamethasone Acetate Loaded Solid Lipid Nanoparticles

        The objective of the present study was to develop a novel solid lipid nanoparticle (SLN) for the lung-targeting delivery of dexamethasone acetate (DXM) by intravenous administration. DXM loaded SLN colloidal suspensions were prepared by the high pressure homogenization method. The mean particle size, drug loading capacity and drug entrapment efficiency (EE %) of SLNs were investigated. In vitro drug release was also determined. The biodistribution and lung-targeting efficiency of DXM-SLNs and DXM-solutions (DXM-sol) in mice after intravenous administration were studied using reversed-phase high-performance liquid chromatography (HPLC). The results (expressed as mean ± SD) showed that the DXM-SLNs had an average diameter of 552 ± 6.5 nm with a drug loading capacity of 8.79 ± 0.04% and an entrapment efficiency of 92.1 ± 0.41%. The in vitro drug release profile showed that the initial burst release of DXM from DXM-SLNs was about 68% during the first 2 h, and then the remaining drug was released gradually over the following 48 hours. The biodistribution of DXM-SLNs in mice was significantly different from that of DXM-sol. The concentration of DXM in the lung reached a maximum level at 0.5 h post DXM-SLNs injection. A 17.8-fold larger area under the curve of DXM-SLNs was achieved compared to that of DXM-sol. These results indicate that SLN may be promising lung-targeting drug carrier for lipophilic drugs such as DXM.

      • Screening Peptides Binding Specifically to Colorectal Cancer Cells from a Phage Random Peptide Library

        The aim of this study was to screen for polypeptides binding specifically to LoVo human colorectal cancer cells using a phage-displayed peptide library as a targeting vector for colorectal cancer therapy. Human normal colorectal mucous epithelial cells were applied as absorber cells for subtraction biopanning with a c7c phage display peptide library. Positive phage clones were identified by enzyme-linked immunosorbent assay and immunofluorescence detection; amino acid sequences were deduced by DNA sequencing. After 3 rounds of screening, 5 of 20 phage clones screened positive, showing specific binding to LoVo cells and a conserved RPM motif. Specific peptides against colorectal cancer cells could be obtained from a phage display peptide library and may be used as potential vectors for targeting therapy for colorectal cancer.

        Multi-Phase MPCA Modeling and Application Based on an Improved Phase Separation Method

        Regarding the multi-phase characteristic of batch process, a new phase separation method is developed in this paper. The method realizes a 3-step sub-phase separation of the batch process using the retained principal components number, loading matrixes and principal component matrixes, which can adequately reflect the features variation of the process. In line with the different features and clas-sification step, automatic identification of ‘burrs’ and transition phases has been expounded. The pro-posed method can directly separate the stable phases and transition phases in the batch process, and deduce high-precision transition phase models. Based on the proposed method, the MPCA modeling and online monitoring is applied in the injection molding process. The experimental results have veri-fied the effectiveness of the proposed method.

      • SYSTEM ANALYSIS OF DISASTER PREVENTION DESIGN CRITERIA FOR COASTAL AND ESTUARY CITIES

        In China, estuary and coastal cities are mostly regional economic development center. The disasters by combined effect of upper reaches flood, storm surge and Typhoon waves are the primary obstacle to economic development of cities. So how to make economic and risk system analysis for flood-storm surge-wave disaster, economic loss and flood-storm surge control measures plays a very important role in the sustainable development of coastal cities. There are three types of coastal cities for consideration. First type city like Tianjin, the most significant damage is from upper reaches flood of a river. The effect of storm surge is negligible, because in the estuaries of river Haihe catchment basin are built tidal barrages. This paper presents a Grey Markov Model (GMM) to forecast flood peak level. GMM combines Grey system and Markov theory to present a high-precision model. Results of forecasted flood peak levels are closed to the measured data. Establish a synthetical model for economic assessment, risk analysis and flood-control benefit. As new contribution, a stochastic simulation technique is used to compute risk probability. In the same time, take into account the effect of correlation between variables on risk probability. Second type city like Shanghai, where sometimes the combined effect of flood peak of river, storm surge from sea is most severe disaster for city damage. 1000 yrs. return period water level of Huangpu river is used as traditional design criteria. Simulated combined water level of flood peak, storm surge and maximum astronomical tide level from sea side with 400 yrs. return period is close to 1000 yrs. design water level by traditional method. For third type city, like Qingdao, where combined effect of maximum astronomical tide, storm surge and waves brought most significant damage to Qingdao. Using stochastic simulation technique, different combinations of storm surge, waves with 1%,2% joint probability level are simulated for disaster prevention.
