        LRG1 expression reduced inflammation of sepsis-renal injury via activation of NLRP3 inflammasome by HIF-1 alpha

        Background Sepsis is a commonly seen severe case in clinical practice and the main cause of health loss worldwide. Objective We investigated the biological function of LRG1 and its underlying molecular mechanisms in sepsis-renal injury. Mice of sepsis model group were subjected to cecal ligation and puncture (CLP) surgery after anesthetization. THP-1 cell was induced by LPS for vitro model. Results The mRNA and protein levels of LRG1 in renal tissue of sepsis model were reduced. LRG1 could reduce the inflammation reaction in model of sepsis-renal injury. In addition, LRG1 up-regulation reduced inflammation reaction and ROS production levels in vitro model via the activation of HIF-1α and suppression of NLRP3 inflammasome. Knockout of LRG1 increased inflammation reaction and ROS production levels in vitro model via the inactivation of HIF-1α and induction of NLRP3 inflammasome. Furthermore, the inhibition of HIF-1α reduced the effects of LRG1 on NLRP3 inflammasome in vitro model. Conclusions The present study suggests that LRG1 expression reduced inflammation of sepsis-renal injury via activation of NLRP3 inflammasome by HIF-1α, thereby may be a promising therapeutic target of sepsis-renal injury.

        Occurrence assessment of earth fissure based on genetic algorithms and artificial neural networks in Su-Xi-Chang land subsidence area, China

        Earth fissures in Su-Xi-Chang land subsidence areahave induced massive damages to the area. The non-linear characteristicassociated with the process of earth fissure formation requiresnon-linear method for evaluating the occurrence of the hazard. Based on quantification of influence factors on breeding the hazard,GA-ANN method, which integrates artificial neural networks(ANN) with genetic algorithms (GA), is developed for evaluating theoccurrence of earth fissure hazard. Six indicators, that include thedepth of bedrock burial (DBB), the degree of bedrock relief (DBR),water level (WL) (the II confined aquifer), the gradient of landsubsidence (GLS), transmissivity (T) (the II confined aquifer) andthe thickness of clay soil (TCS), are selected as the input patternsof the integrated approach, and danger index (DI) as the outputpattern. A multilayer back-propagation neural network is trainedwith 30 sets of data samples including 15 sets of earth fissure samplesand 15 sets of safety samples for defining the architecture ofANN. Subsequently, GA is employed by optimizing the initial weightsof trained ANN by minimizing the deviation of output. The efficacy ofthe integrated approach is demonstrated by comparing the deviationof output from ANN and GA-ANN for 5 testing samples andthe result shows that the GA-ANN method is more accurate thanANN in identifying the occurrence of earth fissure. The integratedmethod is applied to the assessment of earth fissure hazard in typicalregions of earth fissure. According to the classification of DI, theregions are divided into four zones ‒ danger zone, sub-danger zone,sub-safe zone and safe zone.

        Helicobacter pylori-Induced Progranulin Promotes the Progression of the Gastric Epithelial Cell Cycle by Regulating CDK4

        Helicobacter pylori, a group 1 carcinogen, colonizes the stomach and affects the development of stomach diseases. Progranulin (PGRN) is an autocrine growth factor that regulates multiple cellular processes and plays a tumorigenic role in many tissues. Nevertheless, the mechanism of action of PGRN in gastric cancer caused by H. pylori infection remains unclear. Here, we investigated the role of PGRN in cell cycle progression and the cell proliferation induced by H. pylori infection. We found that the increased PGRN was positively associated with CDK4 expression in gastric cancer tissue. PGRN was upregulated by H. pylori infection, thereby promoting cell proliferation, and that enhanced level of proliferation was reduced by PGRN inhibitor. CDK4, a target gene of PGRN, is a cyclin-dependent kinase that binds to cyclin D to promote cell cycle progression, which was upregulated by H. pylori infection. We also showed that knockdown of CDK4 reduced the higher cell cycle progression caused by upregulated PGRN. Moreover, when the PI3K/Akt signaling pathway (which is promoted by PGRN) was blocked, the upregulation of CDK4 mediated by PGRN was reduced. These results reveal the potential mechanism by which PGRN plays a major role through CDK4 in the pathological mechanism of H. pylori infection.

        Kraft Lignin Biodegradation by Dysgonomonas sp. WJDL-Y1, a New Anaerobic Bacterial Strain Isolated from Sludge of a Pulp and Paper Mill

        Wastewater containing kraft lignin (KL) discharged from pulp and paper industries could cause serious environmental contamination. Appropriate effluent treatment is required to reduce the pollution. Investigations on anaerobic bacteria capable of degrading KL are beneficial to both lignin removal and biofuel regeneration from the effluent. In this paper, an anaerobic strain capable of degrading KL was isolated from the sludge of a pulp and paper mill and identified as Dysgonomonas sp. WJDL-Y1 by 16S rRNA analysis. Optimum conditions for KL degradation by strain WJDL-Y1 were obtained at initial pH of 6.8, C:N ratio of 6 and temperature of 33°C, based on statistical analyses by response surface methodology. For a 1.2 g/l KL solution, a COD removal rate of 20.7% concomitant with biomass increase of 17.6% was achieved after 4 days of incubation under the optimum conditions. After the treatment by strain WJDL-Y1, KL was modified and degraded.

      • Model test method for dynamic responses of bridge towers subjected to waves

        In order to establish a dynamic model test method of bridge pylons subjected to ocean waves, the similarity method of hydroelastic model test for bridge pylons were analyzed systematically, and a model design and production method was proposed. Using this method, a dynamic test model of a bridge pylon was made, and then a free vibration test on the model structure and a dynamic response test of the model structure under wave actions were conducted in a wave flume. The results of the free vibration test show that the primary natural frequencies of the structure by the model test are close to the design frequencies of the prototype structure, indicating that the dynamic characteristics of the bridge pylon are well simulated by the model structure. The results of the dynamic response test show that wave induced base shear forces and motion responses on the model structure are consistent with the numerical results of the prototype structure. The model test results confirm that the proposed model test design method is feasible and applicable. It has application and reference significances for model testing studies of such marine bridge structures.
