      • Effective Dose of Peri-operative Oral Pregabalin as an Adjunct to Multimodal Analgesic Regimen in Lumbar Spinal Fusion Surgery

        STUDY DESIGN.: A prospective, randomized, controlled, and double-blind trial. OBJECTIVE.: To evaluate the effects of 2 different doses of perioperative pregabalin administration, twice on the day of surgery, on acute postoperative pain after spinal surgery. SUMMARY OF BACKGROUND DATA.: Besides its well-established role on neuropathic pain, pregabalin seems to be a promising adjunct to multimodal analgesic regimen following surgery. No comprehensive data exist regarding the optimal dosage of pregabalin on reducing postoperative pain and opioid consumption in spinal surgery. METHODS.: Patients were randomly assigned to 1 of 3 groups. The placebo group (n = 28) received placebo capsules 1 hour before the anesthetic induction and 12 hours after surgery. The pregabalin groups received pregabalin 75 mg (P75 group, n = 28) or 150 mg (P150 group, n = 28), respectively at the same points. Assessed variables were total amount of administered fentanyl-based intravenous patient-controlled analgesia, pain intensity, and the frequency of rescue analgesic administered during the first 48 hours after surgery, subdivided into the following 4 periods: on arrival of patient to the postanesthesia care unit, 1 to 6 hours, 6 to 24 hours, and 24 to 48 hours. RESULTS.: The amount of patient-controlled analgesia volume infused until 24 hours (P 5 0.025) and 48 hours (P 5 0.028) after surgery was significantly less in the P150 group compared with the control group. The frequency of additional anodynes administered until 6 hours (P 5 0.049) and 24 hours (P 5 0.045) after surgery was significantly less in the P150 group compared with the control group. CONCLUSION.: Perioperative administration of pregabalin 150 mg before and 12 hours after surgery, but not 75 mg, significantly reduced opioid consumption and the use of additional pain rescue for 48 hours after surgery without significant side effects in patients undergoing spinal fusion surgery.

      • Regulation of conceptus interferon-tau gene subtypes expressed in the uterus during the peri-implantation period of cattle

        <P><B>Abstract</B></P> <P>Conceptus interferon tau (IFNT), produced by the embryonic trophectoderm, is known as a major signaling protein essential for the process of maternal recognition of pregnancy in ruminants. Similar to other <I>IFN</I> gene families such as <I>IFNA</I> and <I>IFNB</I>, multiple <I>IFNT</I> genes exist. The number of <I>IFNT</I> genes actively transcribed and regulated in conceptuses of cattle has, however, not been well characterized. In this study, <I>IFNT</I> transcripts <I>in utero</I> were studied through the use of next generation sequencer. Among 38 <I>IFN</I> genes registered and eight annotated as <I>IFNT</I>, only two transcripts, <I>IFNT1</I> and <I>IFNTc1</I>, were found in conceptuses <I>in utero</I>. Relative abundance of transcription factor(s) involved in the regulation of <I>IFNT</I> genes were investigated by real-time PCR. Transcriptional activity of <I>IFNT1</I> and <I>IFNTc1</I> were investigated using bovine non-trophoblast ear fibroblast (EF) cells, which were co-transfected with luciferase reporter constructs with upstream (−631 to −51) promoter regions of <I>IFNT1</I> or <I>IFNTc1</I> and various transcription factor expression plasmids, CDX2, AP1 (JUN), ETS2 and/or CREBBP. CDX2 with AP1 and ETS2 was found to increase luciferase activity of <I>IFNT1</I> and <I>IFNTc1</I> approximately 14- and 11-fold, respectively, in EF cells, which do not express the CDX2 gene. These results indicated that two isoforms of conceptus <I>IFNT</I> genes of cattle could be regulated differently <I>in utero</I>. Furthermore, <I>IFNT1</I> and <I>IFNTc1</I> were found to have similar antiviral activity, suggesting that both <I>IFNT</I> genes could function to increase conceptus signaling to the uterine endometrium for the process of maternal recognition of pregnancy during the pre-implantation period.</P>

        Characterizations of the bovine subtype Interferon-tau Genes : Sequences of Genes and Biological Activity of Transcription Factors in JEG3 Cell

        Multiple interferon tau (IFNT) genes exist in bovine. An antiluteolytic substance secreted by the bovine conceptus and primarily responsible for maternal recognition of pregnancy is bovine trophoblast protein 1 (bIFNT1), a new type I interferon tau (IFNT) genes. The objectives of this research were to investigate whether multiple, distinct gene encode bIFNT1 and other type I bIFNT gene in the bovine genome and to examine expression of bIFNT1 and other bIFNTc1 mRNAs during conceptus development. These transcrips could be regulated through caudal-related homeobox-2 (CDX2) and ETS2 and/or AP1 (JUN) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. The presence of mRNAs encoded by bIFNT1 and type I bIFNTc1 genes were examined quantitatively via reverse transcription-polymerase chain reaction (RT-PCR) analysis of total cellular RNA (tcRNA) extracted from on day 17, 20 and 22 bovine conceptuses. The expression level of bIFNT1 was higher on day 17 transcripts were gradually weakly detectable on day 20 and 22. However, the other bIFNTc1 gene examined transcripts was highly expressed on day 20 and transcripts were weakly detectable on day 17 and 22 bovine conceptuses. Furthermore, human choriocarcinoma JEG3 was co-transfected with an -1kb-bIFNT1/c1-Luc constructs and several transcription factor expression plasmids. Compared to each -1kb-bIFNT1/c1-Luc increased when this constructs were co-transfected with, ETS2, AP1(JUN), CREBBP and/or CDX2. Also, bIFNTc1 gene was had very effect on activity by alone ETS2, and AP1 (JUN) expression factors in choriocarcinoma JEG3 cell. However, bIFNT1 gene expression of the upstream region was not identified. We demonstrated that the activities of bIFN genes are regulated by differential, tissue-specific and developmental competence during pregnancy.

        A Simple Confocal Microscopy-based Method for Assessing Sperm Movement

        In the field of reproductive medicine, assessment of sperm motility is a key factor for achieving successful artificial insemination, in vitro fertilization, or intracellular sperm injection. In this study, the motility of boar sperms was estimated using real-time imaging via confocal microscopy. To confirm this confocal imaging method, flagellar beats and whiplash-like movement angles were compared between fresh and low-temperature-preserved ($17^{\circ}C$ for 24 h) porcine sperms. Low-temperature preservation reduced the number of flagellar beats from $11.0{\pm}2.3beats/s$ (fresh sperm) to $5.7{\pm}1.8beats/s$ and increased the flagellar bending angle from $19.8^{\circ}{\pm}13.8^{\circ}$ (fresh) to $30.6^{\circ}{\pm}15.6^{\circ}$. These data suggest that sperm activity can be assessed using confocal microscopy. The observed motility patterns could be used to develop a sperm evaluation index and automated confocal microscopic sperm motility analysis techniques.

        The effect of inorganic and organic mineral premix in broiler diets on growth performance, and fecal mineral excretion

        본 연구는 육계사료에 무기태 또는 유기태 형태의 미네랄 프리믹스 첨가가 생산성 및 분변내 미네랄 배설에 미치는 영향을 구명하고자 수행하였다. 총 750수의 1일령 ROSS 308 (Initial BW = 42.32±0.32g)을 공시하여 3처리 5반복으로 반복당 50수씩 완전 임의 배치하여 자유 섭식케 하였으며 28일간 전기(0~14일), 후기(15~28일)로 나누어 실시하였다. 처리구들은 대조구(프리믹스 무첨가), 무기태 미네랄(대조구 +무기태 미네랄 프리믹스 1.0 g/kg), 유기태 미네랄(대조구 + 유기태 미네랄 프리믹스 1.8 g/kg) 총 3처리구 였다. 전기(0~14일) 및 후기(15~28일) 및 전기간(0~28일) 생산성에는 처리구간에 유의적 차이는 나타나지 않았지만, 폐사율은 무기태 및 유기태 미네랄 처리구가 대조구와 비교하여 유의적으로 (p<0.05) 낮았다. 분변 내 철, 구리, 아연, 망간, 마그네슘 함량은 처리구간에 차이가 유의적으로 나타났으며 (p<0.05), 무기태 처리 그룹에서 가장 높은 함량을 보여주었다. 비장과 간에서 철, 구리, 아연, 망간, 마그네슘의 함량은 유기태 처리그룹에서 유의적으로 (p<0.05) 가장 높은 함량을 보여주었다. 결론적으로 유기태 미네랄 프리믹스의 첨가는 육계의 생산성에 영향을 미치지 않으며, 환경오염을 유발하는 분변 내 미네랄 함량을 낮출 수 있는 것으로 확인되었다. This study examined the effect of diets supplemented with organic and inorganic mineral premixes on growth performance and mineral excretion. Seven hundred and fifty one-day-old ROSS 308 (initial BW= 42.32±0.32 g) were assigned randomly to one of three dietary treatments, repeated five times: mash-type basal diet supplemented with an inorganic mineral premix (1.0 g/kg) and a basal diet supplemented with an organic mineral premix (1.8 g/kg). In the starter period (0 to 14 days), grower period (15 to 28 days), and overall periods (0 to 28 day), no significant differences were observed between any of the performance parameters measured. On the other hand, the mortality rate was lower (p < 0.05) in the inorganic and organic mineral premix groups than in the basal diet group. The concentrations of iron, copper, zinc, manganese, and magnesium in the fecal were different (p < 0.05) among the treatment groups, with the highest values reported in the inorganic mineral premix group. The concentrations of iron, copper, zinc, manganese, and magnesium in the spleen and liver were different (p < 0.05) among the treatment groups, with the highest values reported in the organic mineral premix group. In conclusion, replacing inorganic mineral premix with organic mineral premix does not adversely affect the productivity of broilers and can reduce the concentration of minerals in feces that cause environmental pollution.

      • The Effects of Cummulus Oocyte Complex Quality on the Embryo Production Via OPU Procesures

        OPU (Ovum pick up) has been an important technique to increase genetic resources for early establishment of genetically important herd and in vitro embryo production system. This study compared relationship between OPU-derived COCs quality and embryo development. The recovery procedures of COCs were performed twice a week using ultrasound guided OPU tools. The 18 gauge disposable needle was connected into 50 mL conical tube by teflon tubing, The grade of COCs was evaluated by microscopic examination. Grade A had 3 or more layers of cumulus cell and compact cytoplasm. Grade B had 1~3 layers of cumulus cell and compact cytoplasm. Grade C had 1 layers cumulus cell and compact cytoplasm. Grade D was denuded oocyte and poor cytoplasm. The collected COCs were fertilized in vitro and developed for 7~8 day in mSOF media. The cleavage rate of grade A/B dominant pick-up group (above 60% of A/B grade rate from total recovered COCs) was 48.9% and that of poor quality was 51.9%. The rate of blastocyst rate in grade A/B dominant OPU procedures after IVF was 28.9%, but poor quality group (below 60% of A/B grade COCs) was 14.8%. The developed blastocysts were transferred into recipient or cryopreserved for further research. When total 11 blastocysts were transferred into surrogate cows with the pregnancy rate (4/11) of 36.3% and 4 calves are expected.

        <B>Purpose</B><P>This study was conducted to evaluate the effectiveness of a computerized surveillance system for adverse drug events (ADEs) reinforced with mandatory reporting of all past drug hypersensitivity reactions (DHSRs) and supervision of the processes by allergy specialists in a university hospital.</P><B>Methods</B><P>All information on both prior and newly developed DHSRs was collected via the surveillance system described above and compared with the data from previous system based on voluntary reporting of DHSRs by attending physicians.</P><B>Results</B><P>The report rate of past DHSRs was greatly increased and the estimated incidence of new events decreased under the new system. The occurrence rate of new DHSRs during hospitalization, which were caused by the repeated administration of the agents previously suspected as culprit drugs enormously, decreased from 15% of previous system to 1% of new system.</P><B>Conclusion</B><P>The mandatory reporting system for past DHSRs and the supervision by allergy specialists appear to be important in improving the management of patients with drug hypersensitivity and in preventing the occurrence of DHSRs in a general hospital.         Changes in major polyphenolic compounds of tea (Camellia sinensis) leaves during the production of black tea

        The objective of this study was to quantify the polyphenolic compounds present in tea samples during black tea processing, and to determine the correlation between the contents of individual catechins and theaflavins. Nine monomeric and four dimeric compounds were identified and quantified by HPLC. During black tea processing, the catechins content decreased, whereas the gallic acid content increased. The decrease in the catechins-in particular, the cis-catechins-was due to the formation of dimeric theaflavins. In the present study, we found a significant negative correlation between the changes in the catechins and theaflavins contents during black tea processing. Theaflavin-3-gallate showed the strongest correlations with the cis-catechins ((-)-epigallocatechin: r=0.713; (-)-epicatechin: r=0.755; (-)-epigallocatechin gallate: r=0.681; and (-)-epicatechin gallate: r=0.771).

      • Seasonal effect of Hanwoo oocyte recovery & embryo development by OPU technology

        The ovum pick up(OPU) technique can be used to produce embryos after in vitro culture of ovarian oocytes, can be used for early securement for effective herd early proliferation and excellent Hanwoo genetic resources, It is attracting attention as a very important technique for establishing technology. In addition to in vitro culture techniques, the number of oocytes retrieved depends on the environment and timing of the OPU. This study was conducted to compare and examine seasonal effect to the differences in the number of recovered oocytes, recovery rate and embryo development rate using Korean cattle kept in animal genetic resource research center by OPU technique. The grade of COCs was evaluated by microscopic examination. Grade A had 3 or more layers of cumulus cell and compact cytoplasm. Grade B had 1~3 layers of cumulus cell and compact cytoplasm. Grade C had 1 layers cumulus cell and compact cytoplasm. Grade D was denuded oocyte and poor cytoplasm. The recovery rate was 47.8±3.4% in summer (June to August) and 51.6±3.8% in autumn (September to October). The number of oocytes was 5.7±0.6 in summer and 5.2±0.7 in autumn. Oocyte grade A and B was 46.2%±6.3% in summer and 51.1±5.0% in autumn. The cleavage rate was 46.1±7.1% in summer and 65.0±11.3% in autumn. Blastocyst development rate was 19.9±9.4% in summer and 29.0±8.7% in autumn. There was no difference the recovery rate of oocytes and the number of embryos between summer and autumn. Cleavage rate and blastocyst rate of autumn was higher than summer. we will investigate to study the appropriate method for the production of Hanwoo embryos and the systematic comparison.
