      • The role of diffusion in NMR proton relaxation enhancement by ferritin

        By using binary solutions of water and glycerol, we controlled diffusion so as to better understand its role in the relaxation rate enhancement of protons in the vicinity of ferritin at 7 tesla. The slower diffusion rates and higher external magnetic field used in these experiments are more consistent with the conditions expected in MRI experiments. New data was obtained on the diffusion coefficients of water and glycerol in binary solutions with relatively dilute amounts of glycerol. The effects of chemical exchange in such systems was also quantified. Two main relaxation mechanisms have been proposed for protons in the vicinity of ferritin: an outer-sphere mechanism (OS) in which spins diffuse past ferritin and experience a changing Larmor frequency by moving through the ferritin's magnetic field, and a proton exchange dephasing mechanism (PE), where protons temporarily reside on the surface of the ferritin core and sample a single, enhanced, Larmor frequency. At high-field, the OS mechanism becomes increasingly important because of a quadratic dependence on field strength, versus linear for the competing mechanism involving proton exchange. It was found that the relaxation enhancement of protons of both water and glycerol in the presence of ferritin was inversely proportional to their diffusion coefficients, in agreement with the OS model of relaxation enhancement. The strength of the relaxation enhancement on inverse diffusion coefficient was weaker for slow-diffusing glycerol than for water: glycerol molecules spent more time in a weaker magnetic field, indicating that glycerol did not approach the ferritin core as closely as water, potentially answering questions about molecular intake into the ferritin structure. The results of these experiments have important implications for the quantification of brain iron in vivo.

      • Simulations of microdomain lattice defects in block copolymer thin films

        In this dissertation we discuss three studies of microdomain ordering in block copolymer thin films. In the first study we report on two new numerical techniques for removing metastable microdomain defects that can be used in field-theoretic computer simulations of block copolymer thin films: (1) a spectral amplitude filter (SF) which encourages the simulation to relax into high symmetry states (representing zero defect states), and (2) different variants of forced-biased, partial saddle point Monte Carlo algorithms that allow for barrier crossing towards lower energy defect-free states. Beyond their use for removing defects, the force-biased Monte Carlo algorithms will be shown to provide a promising tool for studying equilibrium defect populations. In the second study, we use self-consistent field theory (SCFT) simulations to examine the effects of small-scale, hexagonal, lateral confinement on microdomain ordering in block copolymer thin films. Specifically, we examine a hexagonal confinement wall with side length approximately equal to five cylindrical microdomain lattice spacings. The commensurability constraints of the small-scale lateral confinement, coupled with various surface-directed phase separation effects, allows the confining wall to have a significant effect on microdomain ordering. Finally, in the third study, we adopt a self-consistent field theory based model to study the self-assembly behavior of a thin block copolymer melt on the surface of a sphere. A sphere is an example of a curved surface with constant positive Gaussian curvature, and therefore, topological constraints require the presence of microdomain defects. In this model, we assume that the composition of the thin block copolymer film only varies in longitude and colatitude and is constant in the radial direction. The modified diffusion equation is efficiently solved using an extension of the Rasmussen-Kalosakas pseudo-spectral splitting algorithm [J. Polym. Sci. Part B: Polym. Phys. 40 (2002), 1777]. Using this approach, we present a detailed examination of defect formation in the lamellar and cylindrical phases on a sphere. A strong segregation limit approximate free energy is also presented, along with simple microdomain packing arguments, to shed light on the observed SCFT simulation results.

        My dissertation examines the definition of manhood in American literature, culture, and society of the post-1945 era. I emphasize the way that the therapeutic value of personal well-being provides the conceptual apparatus as well as the vocabulary for codes of manhood invoked by conflicts between individual identity and institutional identity. Within a society in which manhood is defined by team membership (i.e. the "organization man") and cool confidence, manhood is alternatively defined as an inward struggle for emotional and intellectual. self-sufficiency. From this conflict, we see the way that institutions create identity, thus affecting the way men move in the world. At the same time, the institutions aren't the real barrier to well-being. The obstacles to individual well-being are other people within groups and institutions. Individuality is achieved and retained through demonstrations of difference from other people. Writers and social critics share an inquiry into how one can remain self-directed, in control of one's own direction in life as well as one's own thoughts and feelings, while participating with the group. At stake is whether or not one acts and thinks according to one's own independent mind or whether one's thoughts and actions are directed from without. It is within this framework that I read masculine identity in the middle decades of the twentieth century. Therapeutic masculinity is coded by inward separation between the individual and others with whom he interacts.       • An educator's perception of STARS from selected Nebraska assessment coordinators

        This study was part of a larger four-part study that investigated the problem of implementing a state accountability system through locally developed assessment and reporting of student performance. The purpose of this quantitative, descriptive study was to survey and describe the perceptions of Nebraska assessment coordinators regarding the implementation of the School-based, Teacher-led Assessment Reporting System (STARS). Seven hundred eighteen educators from across the state of Nebraska participated in this four-part research study. Upon conclusion of the four studies, the researchers completed a comparative analysis of the four groups. The major findings of the four-part study was that educators were generally positive in their perceptions of STARS. ESU staff developers gave the STARS model the most positive responses of the four educator groups surveyed; conversely, teachers gave STARS consistently lower marks relative to the other groups. Scores from assessment coordinators and principals were generally similar and usually placed between the scores of ESU staff developers and teachers. All groups perceived that public education in Nebraska improved due to STARS with results aligning to educators' general perceptions of STARS. Other major findings of the four-part study were that curricular and assessment knowledge improved due to STARS as did teachers as leaders of learning in this teacher-led assessment system.

      • Characteristics and analysis of major U.S. air carrier accidents between 1991 and 2010

        Scope and Method of Study: This study described the characteristics of major U.S. air carrier accidents between 1991 and 2010. A case control methodology was used to compare the characteristics of air carrier accidents citing pilot performance as a causal or contributing factor with air carrier accidents not citing pilot performance as a causal or contributing factor in order to determine whether any significant differences existed between groups. The researcher was particularly interested in determining whether a statistical difference existed between groups with regard to flight experience and level of certification. Findings and Conclusions: The major findings of this study indicate that between 1991 and 2010, 96% of the first officers involved in a major U.S. air carrier accident possessed at least 2,000 hours of total flight time. Of the two first officers (4%) with less than 1,500 hours of total flight time, neither were involved in an accident which cited pilot performance as a causal or contributing factor. This finding did not support the notion that a 1,500 hour total flight time requirement will contribute to the safety of air carrier operations conducted under 14 CFR 121. However, the findings of this study also indicate there was a significant increase between 1991-2000 and 2001-2010 with regard to the distribution of accidents involving a first officer who's highest certificate was a commercial pilot certificate, irrespective of the citing/not citing of pilot performance. While an ATP certification requirement for first officers will certainly eliminate the possibility of any future accidents involving commercially certificated first officers, it was not possible to predict whether such a change will contribute to the enhancement of safety for 14 CFR 121 air carrier operations. It is possible there will simply be a redistribution of the number of accidents involving ATP certificated first officers.

      • Becoming: Female subjectivity, cultures of consumption, and embodiment in the contemporary Spanish novel

        This investigation examines the novels of three contemporary Spanish female writers---Silvia Grijalba's Alivio rapido (2002), Gabriela Bustelo's Veo, veo (1996), and Lucia Etxebararia's Amor, curiosidad, prozac y Judas (1997)---to explore the relationships between globalization, consumption., and economies of experiences that produce new meanings and practices of culture, and as a result, yield innovative manners in which to re-think and re-create female subjectivity. The premise is that Woman, as the nomadic female subject dislocated within the masculine centered power structure, shifts and seeks alternative spaces for the process of becoming (or constructing identity) vis-a-vis her interaction with cultures of consumption. While the three authors maintain distinct personal narratives, which have informed and served as contingencies in the articulation of their identity, there exists a commonality among their work. By way of their novels, each author depicts interconnected vignettes of female subjectivity manifest through myriad cultures of consumption---technology, dance, drugs, fashion---that transpires in distinct proxemics: bar, multinational, underground 'movida' nightlife, music festivals, various neighborhoods of Madrid, and the confines of the home. Through consumption, the protagonists negotiate female subjectivity based upon a nomadic, molecular journey of becoming. This process of becoming is hinged on the continuous interrogation of the hegemonic system and conventional gender roles, intimately linked to maneuverings through the consumer realm. Each protagonist, through divergent means, constructs her subjectivity via specific accoutrements and questions her identity and that of Woman within the masculine economy. The mode in which each cultivates subjectivity is distinct; yet she etches a self who negotiates meaning via the appropriation of goods (tangible and abstract), associates with 'imagined communities' (Anderson), and constructs an active subjectivity, while transmitting codes legible to the masses. Via consumption, the protagonists navigate a multiplicity of images and codes in their journey of becoming an active female subject---an agent with potential to enact change and evolve. Furthermore, for some of the protagonists, there exists liberatory potential stemming from autonomous self-creation by way of the consumer realm. In this sense, in accord with Rosi Braidotti and her theory of the nomad, the protagonists arrive to embody the feminist female subject who has abandoned longing for salvation. Rather, these women focus on regeneration either via subversion of the power structure or working within it to their advantage, evidenced by the dynamic multiplicity of female subjectivity that is molecular, fluid, and evolving.

      • The organization and performance implications of vertical interfirm exchanges at small and entrepreneurial firms

        This dissertation aims to further advance our theoretical and empirical understandings of interfirm governance decisions among entrepreneurial or small firms and their large firm exchange partners. Entrepreneurial firms often establish relationships with large firm partners to gain access to critical resources. While these relationships can support the growth and even survival of the entrepreneurial firm, they can also present great risk if the large firm partner behaves in an opportunistic manner. Entrepreneurial firm managers must decide how to govern these relationships so the potential benefits can be realized and the risks minimized. Consisting of three related essays, this dissertation applies resource-based theory (RBT) and transaction cost economics (TCE) to empirically investigate the antecedents to and performance implications of exchange governance choice among entrepreneurial and small firms in exchanges with large firm partners. The first essay develops and tests a model to provide simultaneous consideration of the benefits and costs associated with how entrepreneurial firms govern alliances with large partners. The empirical setting is alliances between entrepreneurial biotechnology firms and their large downstream partners. Primary and secondary data for this study was collected on 59 entrepreneurial firm-large firm dyads in a three-phase process. The second essay presents a similar model and tests it using a sample of 365 relationships between small firms and their primary financial services supplier. Data for this study is taken from the Federal Reserve Board's 1998 Survey of Small Business Finances (SSBF). Whereas the first two essays analyze the antecedents to and performance consequences of one governance device in each interfirm relationship, the third essay examines the tradeoffs among multiple governance devices that firms bundle together. A total of 796 small firm-financial institution relationships from the SSBF are used in this study. The study examines the relationships and tradeoffs among five different governance devices to determine how they tend to be bundled into effective and efficient governance mechanisms. The performance implications and possible prioritization schemes of different governance device combinations are compared and discussed.
