The karyotypic analyses of Crinum asiaticum var. japonicum and C moorei var. album were carried out using conventional staining and Giemsa C-banding techniques, and distribution of B-chromosome was investigated. Chromosome compositions were diploid of 2n=22 in both species, of which five pairs including sat-chromosomes were submetacentrics, five pairs metacentrics and one pair subtelocentrics in C. asiaticum var. japonicum, but five pairs including sat-chromosomes were submetacentrics and the rest six pairs metacentrics in C. moorei var. album. Chromosome length was in a range of 5.6±0.4μm and 17.5±1.4 ㎛ in C. asiaticum var. japonicum, and 6.7±0.6㎛ and 15.8±4.7㎛ in C. moorei var. album. Consequently a little more variation in chromosome length was observed in C. asiaticum var. japonicum. Giemsa C-banding distribution of two species was very low, and thick C-bands were observed only near secondary constriction part of chromosome g. But there was a little difference in appearence of minor C-bands between two species, and this considered to be a characteristic of both species. B-chromosomes were observed only in C. asiaticum in a number of one to seven. But distinguishable morphological differences according to the different B-chromosome number were not observed.

C-banding karyotypes of Pinus rigida, P. densiflora and P. thunbergii were investigated. Somatic chromosome number of three species was found to be 2n=24, and twelve pairs of homologous chromosome were classified on the basis of the number and size of bands, secondary constriction, total length and arm ratio. The results obtained under this study are summarized as follows; chromosome complements in three species were all metacentric, and the interspecific difference was difficult to identify in the chromosome morphology. The bands on chromosomes were generally located in the centromere, secondary constriction, and intercalary regions of the long and short arms. The bands were revealed in the all centromeric regions except for a pair of E chromosome in P. rigida. The number of bands revealed on each chromosome was one or two.

Some genetic variants of Amphihaploid, Hypoamphidiploid, Amphidiploid are regenerated from tissue culture in Allium wakegi. They were applied to investigate the effects of the chromosomal differences on the gene expression of some isozymes. The electrophoretic migration patterns of total proteins were different at the position of MW5×10^5 Dalton among the genetic variants. Three different isozymes were characterized from the migration patterns of isozymes in the discontinuous polyacrylamide gel electrophoresis. Alcohol dehydrogenase(ADH) appeared 3 bands in the gel electrophoresis corresponding to 3 isozymes, that is allozymes distributed as single isozyme in each individual. Isocitric acid dehydrogenase(ICDH) appeared 5 bands in the gel and corresponds to 5 isozymes as allozymes which distributed single isozyme in each individual. Phosphoglucoisomerase(PGI) appeared 8 bands in electrophoresis and to 8 isozymes which originated from 2 genes and distributed more than two isozumes in each individual. According to the isozume expression in the genetic variants of Allium wagegi, the amplification of chromosomal numbers can affect neither to increase the isozyme numbers and concentration nor to express heterogeneous types in the genetic variants.

Trypsin 處理에 依한 고양이 染色體의 Giemsa分染을 얻었으며, 各染色體上에 나타난 band의 分布와 크기를 詳細히 記述하였으며, 特히 ZW 型인 性染色體를 band 分布로 區分하였다. On the basis of karyotypic analysis performed by conventional staining and Giemsa C-banding technique, cytological relationship was inferred for 21 lines of Paeonia lactiflora Pal. cultivated in Korea. it was very difficult to infer their organized karyotypic classification system using the composition of somatic chromosomes involving sat-chromosomes, relative length of chromosomes, arm ratio and karyotypic formulae by conventional staining. From the distribution and number of Giemsa C-bands on the chromosomes b and c, 21 lines can be subclassified into 5 groups. It seems that the karyotypic polymorphism is observed in 21 lines of cultivated P. lactiflora because peony mainly propagates by outbreeding.
CALLUS FORMATION FROM PROTOPLAST CULTURES OF ALLIUM WAKEGI

Protoplasts were enzymatically isolated from leaves obtained from bulb cultures of amphidiploid A. wakegi, and protoplasts were inoculated in KM8P liquid medium containing 0.1 M Mannitol, 0.4m glucose. 0.4 ㎎/ℓ 2,4-D, 2 ㎎/ℓ NAA and 1 ㎎/ℓ BAP, and incubated in dark. Colony formation was observed within two weeks of inoculation, and developed into mini-callus after one month. The minicallus obtained were transferred to MS medium containing 3% sucrose, 0.4 ㎎/ℓ 2,4-D, 2 ㎎/ℓ NAA. 1 ㎎/ℓ BAP, and 0.8% agarose, and developed into friable callus.

한국에서 재배되는 벼의 품종중 낙동벼, 신선찰벼, 삼강벼에 대하여 핵형분석을 수행하였다.벼의 체세포 염색체가 너무 작아서 기존의 염색방법들로는 정확하게 구별할 수 없었기 때문에 spirit-flame drying과 Giemsa염색법을 이용한 새로운 기술로서 전중기상의 각 염색체를 확인하였다. 3품종의 염색체수는 2n=24이며 각 상동염색체들은 염색양상, 2차협착, 염색체의 길이, 그리고 완의 비로서 확인하였다. 낙동벼의 염색체는 7쌍의 중부동원체, 3쌍의 차중부동원체, 2쌍의 차단부동원체로 구성되어 있고, 신선찰벼의 염색체는 6쌍의 중부동원체, 4쌍의 차중부동원체, 2쌍의 차단부동원체로 구성되어 있다. 그리고 삼강벼의 염색체는 4쌍의 중부동원체, 6쌍의 차중부동원체, 2쌍의 차단부동원체로 구성되어 있다.
Cytological Studies on Two Species of Hosta Ⅰ. Karyotypes of H.minor and H. plantaginea

Hosta 屬 植物의 種分化硏究의 一還으로 좀비비추와 둥근옥잠화의 核型을 分析하였다. 두 種에서 染色體수는 2n=60으로서 大型, 中型, 小型의 3群으로 染色體를 分類하였다. 두 種間에 中型群에서 核型上의 差異가 있었으며, 染色體形態上 中部에서 次中部로 移行한다고 추정된다. The somatic chromosome numbers and chromosomal composition of two species were ascertained. The karyotype of H. minor were as follows: the 60 chromosomes were in 30 pairs, which could be classified into four pairs of large-sized chromosomes, two pairs of middle-sized ones, and 24 pairs of small-sized ones. In H. Plantaginea, the 60 chromosomes could be divided into 4 pairs of large-sized chromosomes, three pairs of middle-sized ones, and 23 pairs of small-sized ones.
