      • Reversion of Multidrug Resistance by SKI-II in SGC7901/DDP Cells and Exploration of Underlying Mechanisms

        In order to investigate whether SKI-II could reverse drug resistance and its possible mechanisms, we treated SGC7901/DDP cells with SKI-II or SKI-II in combination with DDP. Then cell growth, apoptosis, micromorphological changes, and expression of SphK1, P-gp, NF-${\kappa}B$, Bcl-2 and Bax were assessed by MTT assay, flow cytometry, electron microscopy, immunocytochemistry and Western blot assay respectively. SGC7901/DDP cells were insensitive to cisplatin 2.5mg/L, but when pretreated with SKI-II, their proliferation was inhibited by cisplatin 2.5mg/L significantly, the inhibition rate increasing with time and dose. The apoptosis rate was also significantly elevated. Expression of SphK1 and P-gp was decreased significantly, Pearson correlation analysis showing significant correlation between the two (r=0.595, P<0.01). Expression of NF-${\kappa}B$ and Bcl-2 was decreased significantly,while that of Bax was increased, compared to the control group. There were significant correlations between SphK1 and NF-${\kappa}B$(r=0.723, P<0.01), NF-${\kappa}B$ and Bcl-2(r=0.768, P<0.01). All these data indicated that SKI-II could reverse drug resistance of SGC7901/DDP to cisplatin by down-regulating expression of P-gp and up-regulating apoptosis through down-regulation of SphK1. The increased apoptotic sensitivity of SGC7901/DDP to cisplatin was due to the decreasing proportion of Bcl-2/Bax via down-regulating NF-${\kappa}B$.

        Manufacture of Precheese Powder by Use of Low-temperature Renneting Made from Raw Milk Using Spray Dryer

        Among the food constituents, proteins differ in coagulation properties as compared to other constituents in food system. Especially milk protein coagulate through different pathways thus this coagulability can be used for manufacture of various dairy products or as a determinant of dairy product analysis. These milk coagulation methods include organic solvent, isoelectric point, trichloroacetic acid, Ca-sensitive casein, heavy metal ion and rennet coagulation. The coagulation experiment was performed using above parameters at $0^{\circ}C$ and $25^{\circ}C$ to find the dehydration conditions before coagulating for precheese powder making. After different chemical treatments, there was no coagulation at $0^{\circ}C$ rather at $25^{\circ}C$ whatever the mode of coagulation methods was. The appearance of precipitate with coagulation methods was quite different from above mentioned methods of coagulation illustrated by scanning electron microscope. These powders were used for fabrication of camembert cheese by renneting coagulation at $0^{\circ}C$, showing the possibility of cheese materials and of food additives for fabrication of products.

        Perforation optimization of hydraulic fracturing of oil and gas well

        Considering the influences of fluid penetration, casing, excavation processes of wellbore and perforation tunnels, the seepage-deformation finite element model of oil and gas well coupled with perforating technique is established using the tensile strength failure criterion, in which the user-defined subroutine is developed to investigate the dynamic evolvement of the reservoir porosity and permeability. The results show that the increases of perforation angle and decreases of perforation density lead to a higher fracture initiation pressure, while the changes of the perforation diameter and length have no evident influences on the fracture initiation pressure. As for initiation location for the fracture in wellbore, it is on the wellbore face while considering the presence of the casing. By contrast, the fractures firstly initiate on the root of the tunnels without considering casing. Besides, the initial fracture position is also related with the perforation angle. The fracture initiation position is located in the point far away from the wellbore face, when the perforation angle is around $30^{\circ}$; however, when the perforation angle is increased to $45^{\circ}$, a plane fracture is initiated from the wellbore face in the maximum horizontal stress direction; no fractures was found around perforation tunnels, when the angel is close to $90^{\circ}$. The results have been successfully applied in an oilfield, with the error of only 1.1% comparing the fracture initiation pressure simulated with the one from on-site experiment.

      • Tumor Markers in Serum and Ascites in the Diagnosis of Benign and Malignant Ascites

        Objective: To evaluate the values of 4 tumor markers in serum and ascites and their ascites/serum ratios in the identification and diagnosis of benign and malignant ascites. Materials and Methods: A total of 76 patients were selected as subjects and divided into malignant ascites group (45 cases) and benign ascites group (31 cases). Samples of ascites and serum of all hospitalized patients were collected before treatment. The levels of carcinoembryonic antigen (CEA), alpha fetoprotein (AFP), cancer antigen 125 (CA125) and carbohydrate antigen 19-9 (CA19-9) were detected by chemiluminescence (CLIA). Results: CEA, AFP and CA19-9 in both serum and ascites as well as CA125 in ascites were evidently higher in the malignant ascites group than in the benign ascites group (P<0.01). Malignant ascites was associated with elevated ascites/serum ratios for AFP and CA125 (P<0.01). The areas under receiver operating characteristic (AUROCs) of CEA and CA125 in ascites and the ratios of ascites/serum of AFP, CEA, CA125 and CA19-9 were all >0.7, suggesting certain values, while those of ascites CA19-9 and serum CEA were 0.697 and 0.629 respectively, indicating low accuracy in the identification and diagnosis of benign and malignant ascites. However, the AUROCs of the remaining indexes were <0.5, with no value for identification and diagnosis. Compared with single index, the sensitivity of combined detection increased significantly (P<0.05), in which the combined detection of CEA, CA19-9 and CA125 in ascites as well as the ratio of ascites/serum of CEA, CA19-9, CA125 and AFP had the highest sensitivity (98.4%) but with relevantly low specificity. Both sensitivity and specificity of combined detection should be comprehensively considered so as to choose the most appropriate index. Conclusions: Compared with single index, combined detection of tumor markers in serum and ascites can significantly improve the diagnostic sensitivity and specificity.

        An improved SCGM(1,m) model for multi-point deformation analysis

        Considering the deformation of discrete monitoringpoints within the same deformable body usually have similar physicalproperties and tend to undergoing identical dynamic process, jointmodelling of the deformation processes of these points in time domainare expected to generate better results. Yin et al. (1997) first extendedthe multi-variable grey model-system cloud grey model SCGM(1,m),with obviously superior modelling mechanism than single-variablegrey model, to multi-point deformation modelling. However, thismodel is still not widely recognized and its applications remain verylimited in the field of deformation analysis. The objective of this studyis to demonstrate the capability of the SCGM(1,m) model, to presenttwo revisions to further improve the performance of the model andto draw more attention to the community of deformation analysis. We first introduce the principles of the SCGM(1,m) model in theanalysis and prediction of deformation surveys. Two practicaltechniques, namely residuals re-modelling and linear regressionadjustment, are then presented to improve the SCGM(1,m) model. Combined with slope monitoring data, the modelling with the originaland the improved SCGM(1,m) models by residuals re-modellingand linear regression adjustment are illustrated. The mean relativeprediction errors decrease from 5.89% to 3.54% and 2.69%, whenthe two refining techniques are applied, respectively, indicating relativeimprovements of 39.9% and 54.3%.

        Four new compounds from the bulbs of Lycoris aurea with neuroprotective effects against CoCl2 and H2O2-induced SH-SY5Y cell injuries

        Three new alkaloids, 2a-hydroxy-6-O-n-butyloduline,O-n-butyllycorenine, (-)-N-(chloromethyl)lycoramine(1–3), and a new phenolic compound, ((7S)-7-(4-hydroxyphenyl)-7-hydroxypropyl)-20-methylbenzene-30,60-diol (14), along with ten known alkaloids (4–13), wereisolated from the bulbs of Lycoris aurea collected fromHuaihua County of Hunan Province, China. Their structureswere elucidated by spectroscopic methods includingHRESIMS, UV, IR, and NMR. All the isolated compoundswere tested for their neuroprotective effects against CoCl2and H2O2-induced SH-SY5Y cell death. Compounds 1–7and 10 exhibited significant neuroprotective effects againstCoCl2-induced SH-SY5Y cell injury, while compounds1–5, 7, 10 and 12 showed obvious neuroprotective effectsagainst H2O2-induced SH-SY5Y cell death.

        Data-driven Multiplayer Mixed-zero-sum Game Control of Modular Robot Manipulators with Uncertain Disturbance

        This paper develops a data-driven multiplayer mixed-zero-sum game control approach of modular robot manipulators (MRMs) with uncertain disturbance via adaptive dynamic programming (ADP). The dynamic model of MRMs is formulated via joint torque feedback. We deem n modules and uncertain disturbance as players in the game theory structure. The uncertainties in the MRM system such as joint friction and interconnected dynamic couplings (IDCs), we employ data-driven model based on recurrent neural networks (RNNs) to built. According to ADP, the Hamilton-Jacobi (HJ) equation can be solved by using critic neural networks and derivate the optimal control law. The closed-loop robotic system is proved to be asymptotic stable via mixed-zero-sum game control. Experiments are conducted to verify the effectiveness.

        6-Alkylsalicylic Acid Analogues Inhibit In Vitro ATPase Activity of Heat Shock Protein 90

        The molecular chaperone heat shock protein 90 (Hsp90) is responsible for maintaining the correct folding and stability of many signaling proteins. It is a promising target of cancer therapeutics and several other diseases, including neurodegenerative disease, nerve injuries, inflammation, and infection. In an effort to identify new Hsp90 inhibitors from natural sources using an in vitro ATPase inhibition assay, two 6-alkylsalicylic acid analogues, salaceyin A and B were identified from the culture extract of Streptomyces. Salaceyin A and B exhibited moderate ATPase inhibitory activities with IC50 values of 68.3 and 65.2 μM, respectively. Binding of salaceyins to human Hsp90α was examined by competition binding experiments with ATPSepharose beads. However, the compounds exhibited no degradation activity of Hsp90 client proteins, Her2, c-Raf, or Akt.

      • Oridonin Suppresses Proliferation of Human Ovarian Cancer Cells via Blockage of mTOR Signaling

        Oridonin, an ent-kaurane diterpenoid compound isolated from the traditional Chinese herb Rabdosia rubescens, has shown various pharmacological and physiological effects such as anti-tumor, anti-bacterial, and anti-inflammatory properties. However, the effect of oridonin on human ovarian cancer cell lines has not been determined. In this study, we demonstrated that oridonin inhibited ovarian cancer cell proliferation, migration and invasion in a dose-dependent manner. Furthermore, we showed oridonin inhibited tumor growth of ovarian cancer cells (SKOV3) in vivo. We then assessed mechanisms and found that oridonin specifically abrogated the phosphorylation/activation of mTOR signaling. In summary, our results indicate that oridonin is a potential inhibitor of ovarian cancer by blocking the mTOR signaling pathway.
