        Isolation, Characterization, and Molecular Cloning of the cDNA Encoding a Novel Phytase from Aspergillus niger 113 and High Expression in Pichia pastoris

        Phytases catalyze the release of phosphate from phytic acid. Phytase-producing microorganisms were selected by culturing the soil extracts on agar plates containing phytic acid. Two hundred colonies that exhibited potential phytase activity were selected for further study. The colony showing the highest phytase activity was identified as Aspergillus niger and designated strain 113. The phytase gene from A. niger 113 (phyII) was isolated, cloned, and characterized. The nucleotide and deduced amino acid sequence identity between phyII and phyA from NRRL3135 were 90% and 98%, respectively. The identity between phyII and phyA from SK-57 was 89% and 96%. A synthetic phytase gene, phyIIs, was synthesized by successive PCR and transformed into the yeast expression vector carrying a signal peptide that was designed and synthesized using P. pustoris biased codon. For the phytase expression and secretion, the construct was integrated into the genome of I? pustoris by homologous recombination. Over-expressing strains were selected and fermented. It was discovered that -4.2 g phytase could be purified from one liter of culture fluid. The activity of the resulting phytase was 9.5 U/mg. Due to the heavy glycosylation, the expressed phytase varied in size (120, 95, 85, and 64 kDa), but could be deglycosylated to a homogeneous 64 kDa species. An enzymatic kinetics analysis showed that the phytase had two pH optima (pH 2.0 and pH 5.0) and anoptimum temperature of 60℃.

        국문요약: 상담은 인간을 다루는 분야로서 상담의 중심에 서 있는 인간에 대한 이해가 상담의 성격을 결정한다 해도 과언이 아니다. 인간은 그가 처해 있는 사회와 문화의 영향을 받기 때문에 한 인간을 이해하려면 그가 속해 있는 문화권을 함께 이해해야 하고 이런 인간을 다루는 상담 역시 그 문화에 부합되어야 한다. 중국의 상담은 초창기(1949~1965), 침체기(1966~1977), 소생기(1978~1986), 상승기(1987~현재) 등 네 단계의 발전과정을 거치면서 현재까지 왔다. 이 과정에서 중국의 문화와 전통이 상담에 줄곧 영향을 끼쳐왔는데 그 영향은 주로 유가, 도가 등 전통사상의 영향, 중국이라는 특수한 사회체제의 영향이다. 이 영향으로 인해 중국의 상담은 심리치료와 상담의 미분화, 상담과 사상정치교육의 결부, 지시적 상담에 대한 선호 등 특징을 지니고 있다. 특히 중국이라는 특정사회에서 이 사회에 부합하는 본토화 상담이론을 정립하려는 노력이 상담의 발전 초기부터 시작되었다는 것이 중국 상담의 또 다른 특징이라 할 수 있겠다. 중국 본토화 상담의 연구는 주로 중국의 사회 문화에 부합하는 상담이론 연구, 중국 전통사상에 나타난 상담원리 연구 등 두가지 면에서 진행되어 왔으며 앞으로도 지속될 것이다. Like any other human activity, counseling is inevitably influenced by the socio-cultural context and such influence is substantial. Throughout the four periods that counseling has undergone in China . the beginning period (1949-1965), the recession period (1966-1977), the revival period (1978-1986), and the rising period (1987 to present) . the socio-cultural influence on counseling primarily from the traditional philosophies such as Confucianism and Taoism, and the current socio-cultural context, has never ceased. It is this unique influence that has made Chinese counseling characteristic of a mixture of counseling and psychotherapy, a combination of ideological and political education, and the preference of directive counseling. Meanwhile, another feature of Chinese counseling, emerging at the beginning period, is the localized study of counseling, which mainly focuses on the areas: developing counseling theory of Chinese characteristics based upon the Chinese socio-cultural context, and applying Chinese traditional thoughts into counseling practice.

      • Directed Evolution of Beta-galactosidase from Escherichia coli into Beta-glucuronidase

        In vitro directed evolution through DNA shuffling is a powerful molecular tool for creation of new biological phenotypes. E. coli $\beta$-galactosidase and $\beta$-glucuronidase are widely used, and their biological function, catalytic mechanism, and molecular structures are well characterized. We applied an in vitro directed evolution strategy through DNA shuffling and obtained five mutants named YG6764, YG6768, YG6769, YG6770 and YG6771 after two rounds of DNA shuffling and screening, which exhibited more $\beta$-glucuronidase activity than wild-type $\beta$-galactosidase. These variants had mutations at fourteen nucleic acid sites, resulting in changes in ten amino acids: S193N, T266A, Q267R, V411A, D448G, G466A, L527I, M543I, Q626R and Q951R. We expressed and purified those mutant proteins. Compared to the wild-type protein, five mutant proteins exhibited high $\beta$-glucuronidase activity. The comparison of molecular models of the mutated and wildtype enzymes revealed the relationship between protein function and structural modification.

        Revelations of Lao Tzu’s Views on Human Nature to Counseling

        상담은 인간을 다루는 분야로서 이 과정의 중심이 되는 인간 이해는 상담에 결정적인 영향을 미친다. 한 인간을 이해하려면 그가 속해 있는 문화권을 함께 이해해야 하고, 이런 인간을 다루고 있는 상담 역시 도움을 받고자 하는 내담자가 속해 있는 문화에 부합되어야 한다. 한국과 중국을 포함한 동양 사회를 살펴보면 수십세기 동안 주로 유교와 불교 그리고 노자 사상의 영향을 받아왔다. 이 중 노자의 철학은 중국 철학사상 최초로 완전한 철학 사상 이론을 정립한 사상으로서 동양 철학과 전통문화에 깊은 영향을 미쳤을 뿐만 아니라 사람들의 세계관, 사고방식, 행동양식 등에도 많은 영향을 미쳤다. 따라서 노자 철학 사상에 입각한 상담 이론의 고찰은 동양 사회의 부응하는 상담이론의 정립에 도움이 된다. Tao Te Ching, the Taoist classic written by Lao Tzu, not only expounds the way (Tao) of the universe, but also well presents Lao Tzu’s views on human nature, psychological problems and the causes, propositions of a mature person, etc. These profound thoughts can bring valuable revelations to counselors, especially those who serve in East Asia.

        Strength and Deformation Characteristics of Silty Sand Improved by Gravel

        Silty sand is a poor filler of highway and railway subgrade, and various physical and chemical improving methods have been applied to increase the strength and stability of silty sand. Adding gravel to silty sand is a routine physical improving method. In this paper, large-scale triaxial tests were carried out on silty sand and improved soil which is obtained by adding 4 different proportions of gravel into the silty sand in order to analyze the strength and deformation characteristics of the improved soil. The stress-strain relations obtained from tiaxial tests were analyzed and the effect of coarse particles and fine particles on the strength and deformation of the specimen were also analyzed. The test results show that with more coarse particles and under a higher confining pressure, a greater deviator stress is required to produce the same axial strain. The increase of coarse particles helps to enlarge the angle of internal friction and cohesion, while the increase of fine particles decreases the angle of internal friction and cohesion. Under low confining pressure, the soil specimens with more coarse particles exhibit evident shear dilation. Under high confining pressure, the soil specimens produce a greater volume strain. Adding appropriate amount of gravel into the silt soil can increase its strength and restrain its deformation, but excessive amount of gravel may produce too much volume deformation.

        Radionuclide identification method for NaI low-count gamma-ray spectra using artificial neural network

        An artificial neural network (ANN) that identifies radionuclides from low-count gamma spectra of a NaIscintillator is proposed. The ANN was trained and tested using simulated spectra. 14 target nuclides wereconsidered corresponding to the requisite radionuclide library of a radionuclide identification devicementioned in IEC 62327-2017. The network shows an average identification accuracy of 98.63% on thevalidation dataset, with the gross counts in each spectrum Nc ¼ 100~10000 and the signal to noise ratioSNR ¼ 0.05e1. Most of the false predictions come from nuclides with low branching ratio and/or similardecay energies. If the Nc>1000 and SNR>0.3, which is defined as the minimum identifiable condition, theaveraged identification accuracy is 99.87%. Even when the source and the detector are covered with leadbricks and the response function of the detector thus varies, the ANN which was trained using nonshieldingspectra still shows high accuracy as long as the minimum identifiable condition is satisfied. Among all the considered nuclides, only the identification accuracy of 235U is seriously affected by theshielding. Identification of other nuclides shows high accuracy even the shielding condition is changed,which indicates that the ANN has good generalization performance.
