        An histochemical study of endocrine cells in the alimentary tract of the red-eared slider, Trachemys scripta elegans

        붉은귀거북이(red-eared slider, Trachemys scripta elegans)의 소화관에 존재하는 위장관내분비세포의 부위별 분포 및 출현빈도를 Grimelius 및 Masson-Hamperi(M-H)도은 염색을 이용하여 조직화학적으로 관찰하였다. 소화관은 식도, 기저부(fundus), 유문부, 십이지장, 공장, 회장 및 대장의 7부위로 구분하였다. 소화관 전 부위에 걸쳐 은호성세포(argy-rophil, Grimelius-positive cell) 및 은친화성세포(argentaffin cell, M-H-positive cell) 들이 관찰되었다. 타원형에서 방추형 또는 난원형에서 원형의 은호성세포 및 은친화성세포들이 기저부와 유문부의 위샘(gastric gland), 장 및 식도상피의 기저부에서 관찰되었으며, 소화관 각 부위별로 다양한 출현빈도를 나타내었다. 은호성세포들은 식도를 포함한 소화관 전 부위에서 관찰되었으며, 중등도의 출현빈도를 나타낸 직장에서 가장 높은 빈도를 나타내었고, 식도, 기저부, 십이지장, 공장 및 회장에서 극소수, 유문부에서는 소수 관찰되었다. 은친화성세포들 역시 소화관 전 부위에서 관찰되었으며, 은호성세포와 유사하게 다수의 출현빈도를 나타낸 직장에서 가장 높은 빈도를 나타내었고, 직장 이외의 부위에서는 소수의 세포들이 관찰되었다. The regional distributions and relative frequencies of endocrine cells were studied histochemically (Grimelius and Masson-Hamperl (M-H) silver methods) in the alimentary tract of the red-eared slider, Trachemys scripta elegans. Samples were taken from the esophagus, fundus, pylorus, duodenum, jejunum, ileum and large intestine. Argyrophil (Grimelius-positive) cells and argentaffin (M-H-positive) cells were found in the whole alimentary tract in this study. Spherical to spindle and/or oval to round-shaped argyrophil or argentaffin cells were located in the gastric glands of the stomach regions, in the basal portion of the epithelium of intestinal tract or the esophagus with variable frequencies. Argentaffin cells were more numerously detected in the whole alimentary tract compared with those of argyrophil cells in this study. Argyrophil cells were observed in the whole alimentary tract including the esophagus and the most predominant region was the rectum with moderate frequency. The relative frequency of these cells was rare in the esophagus, fundus, duodenum, jejunum and ileum, respectively and a few frequency in the pylorus. Argentaffin cells were also observed in the whole alimentary tract including the esophagus and the most numerously demonstrated region was the rectum with numerous frequency. They were observed with a few frequencies in the remaining regions of the alimentary tract except for the rectum, respectively. However, to know the exact type of the argyrophil cells and argentaffin cells that were observed in this study, more developmental methods such as immunohistochemistry were needed.

        아무르장지뱀의 위장관 내분비세포에 관한 조직화학적 및 면역조직화학적 연구

        아무르장지뱀(Takydromus amurensis)의 위장관 점막에 분포하는 내분비세포의 분포와 출현빈도 및 세포의 종류를 알아보기 위하여 몇가지 도은법 및 면역조직화학적 방법으로 관찰하였다. Grimelius법에만 염색된 은호성세포(argyrophil cell)는 유문부에 다수로 십이지장에 중등도로 국한되어 출현하였다. 한편 bovine CG면역반응세포는 유문부에서 최고의 빈도로 전장관에 걸쳐 분포하였다. BPP면역반응세포는 소장부위에서만 동정되었다. 따라서 아무르장지뱀의 위장관 내분비세포에서 은호성세포와 bovine CG면역반응세포는 분포와 출현빈도에서 일치하지 않았다. The distribution and the frequency of endocrine cells in the GIT of the Amur lizard(Takydromus amurensis) were investigated using silver techniques and immunohistochemical method. Only argyrophil cells stained by the Grimelius technique were found numerous in the pyloric glands and moderate in the duodenum. Two types of immunoreactive endocrine cells were identified by immunohistochemical method. Bovine CG-immunoreactive cells were demonstrated the entire GIT. BPP-immunoreactive cells were restricted in the duodenum and the ileum. The results showed that: the number of argyrophil cells was lower than the number of cells stained with bovine CG antiserum. Therefore, bovine CG-immunostaining and the Grimelius silver technique did not correspond with various endocrine cells in the Amur lizard.

        An immunohistochemical study of endocrine cells in the alimentary tract and pancreas of the toad, Bufo bufo gargarizans Cantor

        The regional distribution and relative frequencies of endocrine cells were studied immunohistochemically (PAP methods) in the alimentary tract and pancreas of the toad, Bufo bufo gargarizans Cantor using specific antisera against bovine Sp-1/chromogranin (BCG), serotonin, bombesin, gastrin, substance P (SP), somatostatin, insulin, glucagon, pancreatic polypeptide (PP), vasoactive intestinal polypeptide (VIP) and secretin. Nine kinds of endocrine cells were identified in this study. Spherical or spindleshaped immunoreactive (IR) cells were located in the gastric glands of stomach regions, in the basal portion of the epithelium of intestinal tract or esophagus, and in the exocrine or pancreatic islets with variable frequencies. In the alimentary tract, BCG-IR cells were found in the fundus and pylorus with rare and a few frequencies, respectively. Serotonin-IR cells were demonstrated in the whole alimentary tract including the esophagus. Bombesin- and SP-IR cells were restricted to the stomach regions and gastrin-IR cells were restricted to the pylorus. Somatostatin-IR cells were detected throughout the whole alimentary tract except for the large intestine, However, insulin-, glucagon-, PP-, VIP- and secretin-IR cells were not detected in the alimentary tract. In the pancreas of toad, the distribution and relative frequency of endocrine cells were similar to those of other mammals. Insulin-IR cells were located in the central portion of the pancreatic islets and interspaces of exocrine portions, and glucagon-, somatostatin- and PP-IR cells were detected in the marginal regions of the pancreatic islets and interspaces of exocrine. However, other IR cells were not found in the pancreas. In conclusion, the regional distribution and relative frequency of the endocrine cells in the alimentary tract and pancreas of the toad were similar to other anuran species but some differences which might be caused by feeding habits and species specification were also observed.

        An immunohistochemical study of the gut hormones in Hynobius leechii

        도롱뇽(Hynobius leechii)의 위장관 내분비세포를 면역조직화학적으로 관찰하였던 바, BPP 면역반응세포는 유문부에서 중등도, 십이지장과 소장에서 소수로 관찰되었다. 중등도의 somatostatin 면역반응세포는 유문부에 국한되어 출현하였다. 5-HT 면역반응세포는 전장관에 걸쳐 출현하였으나 유문부에서 다수 그리고 직장에서 극소수로 분포하였다. 특히 BPP와 5-HT면역반응세포는 도롱뇽에서 최초로 동정하였다. Using immunohistochemical technique, three endocrine cell types were observed in the GIT of the Korean salamander, Hynobius leechii. BPP-immunoreactive cells were distributed from the pyloric region to the small intestine. A moderated number of somatostatin-immunoreactive cells were restricted to the pyloric region. 5-HT-immunoreactive cells were demonstrated throughout the GIT, reaching a peak in the pyloric region. The presence of BPP- and 5-HT-immunoreactive cells are identified for the first time in Hynobius leechii.

        An immunohistochemical study on the gastro-entero-pancreatic endocrine cells of the cat-shark, Scyliorhinus torazame

        두툽상어의 위장관 내분비세포의 부위별 분포, 출현빈도 및 세포의 종류를 밝히고자 면역조직화학적 방법으로 관찰하였던 바, 5종의 면역반응세포가 동정되었다. 5-HT 면역반응세포는 최고의 빈도로, somatostatin 면역반응세포는 소수의 위부위와 극소수의 장부위에 걸쳐 각각 전장관에서 관찰되었다. Glucagon과 BPP 면역반응세포는 십이지장과 직장을 제외하고 다양한 빈도로 소화관 전체에서 출현하였다. 한편 Gas/CCK 면역반응세포는 소장부위에서 국한하여 소수 분포하였다. 이상에서 두툽상어의 위장관 내분비세포는 부위별 분포에 있어서 다른 종과 유사하였으나, 출현빈도는 다소 낮게 나타났음을 알 수 있었다. The regional distribution and relative frequencies of gastrointestinal endocrine cells were studied immunohistochemically in the GIT of the cat-shark. Five kinds of endocrine cells were identified in this study. Mostly these cells were of open types, except for glucagon-, BPP- and somatostatin-immunoreactive cells in the stomach which seemed to be of closed type. 5-HT-immunoreactive cells were detected throughout the GIT, and were more freguent than of the other regions. Glucagon-and BPP-immunoreactive cells were demonstrated in the entire GIT except for the duodenum an the rectum, and occurred in various frequencies. A few numbers of Gas/CCK-immunoreactive cells were restricted to the duodenum. Somatostatin-immunoreactive cells were distributed in the whole GIT, and were a few in numbers in the stomach and rare in the intestine, respectly. These results suggest that the pattern of the regional distribution is rather similar to that reported fur previously, but relative number was less frequent than that of other species.

        생후 발생단계에 따른 한국재래산양 위장관에 있어서 somatostatin 면역반응세포에 대한 면역조직화학적 연구

        한국재래산양의 개체발생의 단계별 (1일령, 1개월령, 6개월령, 성체) 차이에 따른 위장관에 존재하는 somatostatin 면역반응세포의 부위벌 분포와 출현빈도률 알아보기 위하여 면역조직화학적 방법으로 관찰하였던 바, somatostatin 면역반응세포는 모든 실험군의 전 위장관에 분포하였으며, 위저부에서 가장 다수로 관찰되었으며, 각 실험군에서 소화관의 원위부로 갈수록 또 연령이 중가할수록 이들 세포는 감소하는 것으로 관찰되었다. 또한 이들 세포들의 형태는 유문부에서 주로 구형, 타원형, 난원형의 폐쇄형 세포가 주로 관찰되었으며, 장 부위에서는 방추형의 개방형 세포가 다수 관찰되었다. 이들 면역반응세포의 각 연령에서외의 정확한 의의는 알 수 없으나 분포 및 빈도의 차이는 연령에 따른 식이습성과 생리적 변화에 기인한 것으로 생각된다. The regional distribution and relative frequency of somatostatin-immunoreactive cells in the gastrointestinal tract were studied during the postnatal development(1 day old, 1 month old, 6-month-old and adult) of the Korean native goat by immunohistochemical methods. Somatostatin-immunoreactive cells were found in the entire gastrointestinal tract and were most predominant in the fundic gland region except for the pyloric gland region of the 1 day old. The number of these cells was observed to decrease along the gastrointestinal tract and to decrease according to the increase with age in all postnatal stages. In this study, immunoreactive cells revealed the differences of regional distribution and the relative frequency in the gastrointestinal tract during postnatal development. These results suggest that although the functional significance of fetal endocrine cells remains unknown, this was caused by changing of feeding habits and physiological conditions during different ages.

        Seasonal variations of the gastro-entero-pancreatic endocrine cells of the frog, Rana dybowskii

        활동기와 동면기에서 산개구리의 위상관내분비세포를 면역조직화학적으로 비교 관찰하였던 바, 5-HT, somatostatin, glucagon, Gas/CCK, Porcine CG 및 BPP 등 6종류의 내분비세포를 동정하였으며, 동면기의 산개구리 위장관에서는 활동기에 비해 내분비세포의 부위별 분포 및 출현빈도에 있어서 특징적인 소견을 관찰할 수 있었다. 즉 면역반응세포의 출현빈도는 동면기에서 현저하게 높았으며, 부위별 출현분포 특히, glucagon-, Gas/CCK-와 BBP-면역반응세포들은 활동기에 비해 동면기의 소화관에 보다 더 광범위하게 출현하였다. 항혈청에 대한 대부분의 내분비세포들은 동면기에서 보다 더 강한 면역반응을 나타내었으며, 특히 위저선부에서 내분비세포들은 표층점막상피의 바로 아래에 위치해 있었다. 이상의 결과에서 산개구리의 위장관내분비세포들은 동면기에서도 활발한 장호르몬을 합성하고 그들의 세포질내 축적되어지는 것으로 시사된다. Comparing hibernation with active periods, the regional distribution and relative frequencies of gastrointestinal endocrine cells were studied immunohist-ochemically in the GITs of the Rana dybowskii. Six kinds of endocrine cells were identified in this study. In this study, several novel findings were emerged: in hibernation period, the perdominant presence of immunoreactive cells; the widely distributional regions of glucagon-, Gas/CCK- and BPP-immunoreactive cells; the intense reaction of immunostaining against the antiserum; and the existence of 5-HT- and somatostat-in-immunoreactive cells in upper regions of the fundic glands. These results suggest that the gastro-entero-endocrine cells in hibernation period synthesize continuously its gut hormones and accumulate in their cytoplasms.

        An immunohistochemical study of endocrine cells in the gut of the Prussian carp, Carassius auratus

        붕어 (Carassius auratus Linnaeus)의 장에 존재하는 소화관 내분비세포의 부위별 분포 및 출현빈도를 포유류의 peptide에 대한 10종류의 항혈청을 사용하여 면역조직화학적 방법으로 관찰하였다. 붕어의 장은 근위부에서부터 원위부까지 5 등분했으며 (Segments I~V), 대부분의 면역반응세포들은 상피부분의 상피세포들 사이 공간에서 관찰되었으며, 장 내강까지 신장되어 있는 긴 세포질돌기를 함유한 방추형의 개방형 세포 (open type cell)들이 주로 관찰되었으나, 세포질돌기 없이 원형 또는 타원형의 형태를 나타내는 폐쇄형 세포 (close type cell)들 역시 소수 관찰되었다. 본 실험에서는 somatostatin, cholecystokinin (CCK)-8 및 pancreatic polypeptide (PP) 면역반응세포들이 관찰되었으나, serotonin, glucagon, chromogranin A, secretin, vasoactive intestinal peptide (VIP), substance P 및 bombesin 면역반응세포들은 관찰되지 않았다. 한편 somatostatin 면역반응세포들은 장의 가장 근위부인 Segment I에 국한되어 극소수 관찰되었으며, CCK-8 면역반응세포들은 장의 근위부인 Segment I과 II에서 소수 또는 극소수의 빈도로 관찰되었다. PP 면역반응세포들 역시 장의 근위부에서 중간 부위인 Segment I에서부터 III에 걸쳐 중등도 또는 극소수의 빈도로 출현하였다. 이상에서 붕어 위 내분비세포들의 부위별 분포 및 출현 빈도는 다른 위가 없는 경골어류 (stomachless teleost)에서의 보고들과 유사하였으나, 독특한 분포를 나타내는 면역반응세포들 역시 관찰되었다. The regional distribution and relative frequency of neurohormonal peptides-producing cells were demonstrated in the gut of the stomachless teleost, the Prussian carp, Carassius auratus Linnaeus, using 10 types of specific antisera raised against mammalian regulatory peptides. The gut of the Prussian carp was divided into five portions from proximal to distal (Segments I~V). Most of immunoreactive cells in the epithelial lining portion, between epithelial cells, were generally spherical or spindle shape having long cytoplasmic process that reached to the lumen (open typed cell) while cells showing round in shape (close typed cell) were found in the basal portions of epithelial lining occasionally. Somatostatin-, cholecystokinin (CCK)-8- and pancreatic polypeptide (PP)- immunoreactive cells were observed in this study. However, no serotonin-, glucagon-, chromogranin A-, secretin-, vasoactive intestinal peptide (VIP)-, substance P- and bombesin-immunoreactive cells were found. Somatostatin-immunoreactive cells were restricted to most proximal segments of the gut (Segment I) with rare frequency and CCK-8-immunoreactive cells were demonstrated in the proximal segments of the gut (Segments I and II) with a few to rare frequencies. In addition, pancreatic polypeptide-immunoreactive cells were demonstrated in the proximal to middle segments (Segments I~III) with moderated to rare frequencies. In conclusion, the distribution and relative frequency of these immunoreactive cells are well corresponded to the previous reports in stomachless teleost but somewhat peculiar patterns are also detected.

        An Immunohistochemical study of the gastro-entero-pancreatic endocrine cells of the insectivorous Korean mole, Talpa micrura coreana

        한국산 두더지의 위장관 점막과 췌장에 분포하는 내분비세포들의 분포와 출현빈도 및 종류를 알아보기 위하여 면역조직화학적으로 관찰하였던 바, 5-HT 면역반응세포는 극소수내지 중등도의 빈도로 전 장관에 고루 분포하였다. Glucagon 면역반응세포는 대장에 국한하여 출현하였다. 또 bovine CG, BPP 및 somatostatin 면역반응세포들은 각각 십이지장, 공장 그리고 위저부를 제외한 전 장관에 다양한 빈도로 고루 분포하였다. Gas/CCK 면역반응세포는 유문부와 소장에서만 다수 그리고 극소수로 관찰되었으나, insulin 면역반응세포는 전 장관에서 관찰할 수 없었다. 한편 췌도의 주변부에서는 glucagon, somatostatin과 BPP 면역반응세포들이, 중심부에서는 insulin 면역반응세포가 높은 빈도로 동정되었으며 또한 외분비에서는 이들 면역반응세포들이 단독 또는 소수의 집단으로 출현하였다. 이상의 결과로써 두더지의 소화관내분비세포의 분포패턴는 고슴도치의 그것과 유사하였으나 십이지장에서 세포의 종류와 출현빈도가 낮은 것을 알 수 있었다. The regional distribution and relative frequencies of the gastro-entero-pancreatic endocrine cells were studied immunohistochemically in the GIT and pancreas of the Korean mole. Seven kinds of endocrine cells were identified in this study. Although 5-HT- and somatostatin-immunoreactive cells were seen throughout the GIT, they were most predominant in the pyloric gland region. Glucagon-immunoreactive cells were restricted on the large intestine. Bovine CG-immunoreactive cells were more frequent in the stomach than in the intestines which were not detected in the duodenum. Numerous Gas/CCK-immunoreactive cells were found in the pyloric gland region, but rarely in the jejunum and ileum. BPP-immunoreactive cells were observed to be rare in the stomach and ileum but were a few in number in the intestines. In the pancreas, four types, namely, glucagon-, somatostatin-, BPP- and insulin-immunoreactive cells were identified in the pancreatic islets and exocrine portion. These results suggest that although endocrine cells of the Korean mole is less abundant in the duodenum, the distribution pattern of its gastro-entero-pancreatic endocrine cells is similar to that reported for the Korean hedgehog.

        Immunohistochemical study of the endocrine cells of the gastrointestinal mucosa of the African clawed toad, Xenopus laevis

        ABC법에 의해 면역조직화학적으로 아프리카발톱두꺼비(Xenopus laevis)의 위장관내분비세포를 관찰하였던 바, neurotensin 면역반응세포는 중등도로 소장에서 약하게 반응하였다. 구형의 gastrin releasing peptide(GRP) 면역반응세포는 위에서 위저부의 상피 세포 바로 아래와 유문부선에서 다수로, 소장에서는 가는 방추형으로 극소수 또는 중등도로 분포하였다. 한편 substance P 면역반응세포는 방추형으로 장상피에서 관찰되었다. 그러나 secretin, motilin, met-enkephalin-8 (M-Enk) 및 polypeptide YY(PYY) 면역반응세포는 전체 소화관에서 관찰할 수 없었다. The gastrointestinal endocrine cells of the African clawed toad, Xenopus laevis have been investigated immunohistochemically using the avidin-biotin method. Seven antisera were tested and three endocrine cell types immunoreacted with antisera to neurotensin, GRP and substance P. A moderate number of neurotensin-immunoreactive cells were weakly reacted in the small intestine. GRP-immunoreactive cells were mainly situated among the upper portion in the fundic glands, and the basal portion in the pyloric glands. These cells were oval and round in shape. On the other hand, in the intestine they were thin spindly cells with the epithelium. Substance P-immunoreactive cells were observed in among intestinal epithelium. However, no secretin-, motilin-, M-Enk- and PYY-immunoreactive cells were found in the GIT of the African clawed toads.
