        방사선 조사 인삼이 랫드의 수태능 및 일반 생식독성에 미치는 영향에 관한 연구

        Korean ginseng products have been fumigated with ethylene oxide (EO) for sterilization and prolongation of storage periods. However, there had been controversies indicating that the consumption of food treated with EO might cause harmful effects in human. Since, in Korea the use of EO gas for food treatment was banned in 1991. Since then, irradiation technique has been developed as an alternative. This study was carried out to investigate the effects of irradiated ginseng on fertility, and reproductive and developmental toxicity. Either EO gas fumigated or gamma-irradiated ginseng was administered to male rats by oral gavage for 63 days during the premating period. Female rats were administered from 14 days before mating to day 20 of gestation or to day 21 of lactation. The exposure amount of irradiation used was 5, 10 and 30 kGy, respectively. There were no treatment related changes of dams in clinical signs, and parturition. No treatment related changes in food consumption, body/organ weights, male/female reproductive and fertility performances were observed. F1 fetuses showed no external abnormality. Reflex/sensory functions, physical/behavioral development, and reproductive performance of F1 rats were not adversary affected. The results of this study show that gamma-irradiated ginseng, up to 30 kGy, has no adverse effects on the fertility, reproduction and development in Wistar rats.

        Retinoic acid와 dibutyryl cyclic AMP가 F9 embryonic carcinoma cell 분화 중 G1 Phase 관련 분자에 미치는 영향

        Retinoic acid (RA) and dibutyryl cyclic AMP (dbcAMP) induce the differentiation of the multipotent embryonic carcinoma cell line, F9 cells, into parietal endoderm like cell. The F9 cells are highly proliferative doubling approximately 12 hours. S Phase is predominant, lasting 10 hours and G2/M phase occupies most of the remaining cycle (2 hours) and G1 phase is nearly non-existent. In this study, we showed the effect of RA and dbcAMP on the cell cycle associated molecules (especially around Gl phase) during F9 cell differentiation. Differentiation of F9 cells was induced by the combined addition of RA (10-7M) and dbcAMP (0.5mM), and cells were harvested daily up to 4 days. Flow cytometric analysis showed the prolongation of G1 phase around 30 hours after induction. Western blot analysis revealed that the amount of cyclin D1 and cdk2 were increased at day 4. However, histone H1 kinase activity of cdk2 was decreased. These data strongly suggest that RA and dbcAMP induce the growth arrest of F9 cells at G1 phase by decreasing the activity of cdk2, although they have increased the protein contents of cyclin Dl and cdk2. The reason for the discrepancy between the Hl kinase activity and protein contents are not clear yet.

      • In Vivo Animal Angiogenic Model and Gene Expression Patterns of Angiogenic Factors and Inhibitors in HCC Cell Lines by Hypoxia

        Angiogenesis, the formation of new blood vessels, is essential for the growth of tumor, and is also known to be tightly controlled by the various angiogenic factors and angiogenic inhibitors. As an in vivo method to quantitate angiogenesis, Matrigel with angiogenic factor was injected subcutaneously into C57BL/6 mice. As a result, Matrigel with bFGF markedly induced the many new blood vessels while no blood vessel was appeared in control mice. To investigate the role of hypoxia in balancing system of the angiogenic and antiangiogenic factors during hepatocarcinogeneis, the expression patterns of factors and inhibitors were examined using hepatocellular carcinoma cell lines, HepG2 and Hep3B. VEGF was highly induced by hypoxia in a time dependent manner. interestingly, IGF- II induced the expression of VEGF gene and synergistically increased that of VEGF when cotreated with hypoxia in HepG2 cells. In contrast, the expression of bFGF was not detected in HepG2 cells even in hypoxic condition by northern blot analysis. The tumor suppressor gene, p53 was decreased by the hypoxia in HepG2 cells. Taken together, VEGF and IGF- II are induced while p53 is down-regulated by hypoxia, suggesting that the expression changes of these factors coordinatively participate to form new blood vessels in early tumor mass.

      • Culture of Rat Embryos During Early Organogenesis

        포유동물의 최기형성을 연구하는데 가장 많이 사용되는 동물인 설치류중 랫트의 배자를 이용하여, egg-cylinder기를 지나 head-fold가 이루어지는 초기체절기인 9.5일경부터 뇌, 눈, 귀, 심혈관계가 형성되는 시기인 11.5일까지 2일(48시간)동안, 체외에서 배양하였다. 그 배양의 최적조건은 실험실마다 다르기 때문에 최적조건확립을 목적으로 이 실험을 행하였다. 온도는 37℃?.2℃로 유지시키고 가스는 배양초기인 9.5일~10.5일 동안을 50% O₂, 5% CO₂, 90% N₂가스를 공급하여 10.5일부터 6시간 동안은 20% O₂,5% CO₂, 55% N₂가스를 배양계에 공급 했을 때 가장 좋은 결과가 나왔다. 배양배지로서는 랫트 IC혈청을 분리하여 열비동화시켜서 사용하였다. 이종혈청이나 합성배지를 첨가해서 배양할 경우는 랫트 혈청에 비해 성장이 좋지 않은 것으로 관찰되었다. 동물실험법의 대체법으로서 배자양법의 기본조건을 확립하여 최기형성 평가 방법으로서의 가능성을 제시하였다. Rat embryos were cultured out of the mother from the head-fold stages, 9.5 days to 11.5 days during which they start to develop the brain, eyes, ears and cardiovascular system etc. We principally did the basic experiment in order to establish the best condition of the rat whole embryo culture in our laboratory. The temperature in the culture system was maintained 37℃ ±0.2℃ for 48 hrs. The culture was carried out in humidified atmosphere of the air, intially, the bottles were equilibrated with 5% O₂, 5% CO₂, and 90% N₂ gas mixture. After 22 or 24 and 29 or 30 hrs the cultures were reequilibrated with 20% O₂, 5% C0₂, 75% N₂ and 40% 0₂, 5% CO₂, 55% N₂ respectively. Various types of sera were tested and for the purpose of minimizing the quantity of rat serum, artificial medium was also tried and it was determined that rat serum supported normal growth over a period of 48 hrs, based on total growth analysis and structural comparisons with in vivo specimens.
