        Objectives : The purpose of this study was to investigate the equivalence between A Yakchim (AY) and V Yakchim (VY) through anti-oxidant and anti-inflammatory activity. Methods : In this study, we analyzed the amount of total sugar, reducing sugar, protein, flavonoid and polyphenol of VY and AY. The anti-oxidant activity was determined by assaying the level of radical scavenging activity against 2,2‘-diphenyl-1-picrylhydrazyl (DPPH) and we investigate the anti-inflammatory effect of VY and AY by measuring the production of nitric oxide (NO) in Raw 264.7 cells activated by lipopolysaccharide. Results : VY (10 mg/ml) and AY (10 mg/ml) showed 4.7±1.7% and 61.7±2.1% of DPPH radical scavenging activity, respectively. And VY (1 mg/ml) and AY (1 mg/ml) decreased 17% and 42% of the NO production, respectively. Conclusions : These results suggested the possibility that the anti-oxidant activity and anti-inflammatory activity of AY are better than VY.

      • CSⅡ 약침의 마우스를 이용한 소핵시험

        Objectives : This study was designed to determine the potential of CSⅡ-YAKCHIM, manufactured from Brassicae Semen, Raphani Semen, Persicae Semen, and Carthami Fructus extracts, to induce micronuclei in bone marrow cells of mice when the CSⅡ-YAKCHIM was dosed once to ICR mice by intramuscular. Methods : Each of the groups consisted of three doses of CSⅡ-YAKCHIM (0.05, 0.1, and 0.2 mL/animal), positive control (Mitomycin C), and negative control (normal saline). The time to harvest bone marrow cells for the main study was 24 hours. Results : The incidence of micronucleated polychromatic erythrocyte (MNPCE) among polychromatic erythrocyte (PCE) was not significantly increased in the CSⅡ-YAKCHIM-treated groups. Also, the ratio of PCE among total erythrocyte of the CSⅡ-YAKCHIM-treated groups was not significantly different when compared to that of the negative control group. Conclusions : Based on these results, CSⅡ-YAKCHIM did not have any potential to induce micronuclei formation in bone marrow cells of mice under the conditions of this study.

        우슬 추출물이 흰쥐 좌골신경 손상 후 좌골신경의 기능회복에 미치는 영향

        Objectives: Peripheral nerve injuries are commonly encountered clinical problem and often result in severe functional deficits. The aim of this study is to evaluate the effects of aqueous extract of Achyranthes japonica(AJ) on functional recovery in sciatic nerve after crushed sciatic nerve injury. Methods: In the present study, the animals in the AJ-treated groups received the aqueous extract of AJ at the respective doses orally for 13 consecutive days. In order to assess the effects of the aqueous extract of AJ on function recovery in crushed sciatic nerve injury, sciatic functional index(SFI) was performed. c-Fos expression in the paraventricular nucleus(PVN) and ventrolateral periaqueductal gray(vlPAG), and neurofilament, and the expressions of brain-derived neurotrophic factor(BDNF), nerve growth factor(NGF) following crushed sciatic nerve injury in rats were investigated. For this, immunohistochemistry and western blot were performed. Results: In the present study, crushed sciatic nerve injury showed characteristic gait changes showing decrease of SFI value and treatment with the aqueous extract of AJ significantly enhanced the SFI value. Neurofilament expression in the sciatic nerve was decreased by crushed sciatic nerve injury and treatment with the AJ increased neurofilament expression. The expressions of BDNF and NGF in the sciatic nerve were increased following crushed sciatic nerve injury and treatment with the AJ significantly controlled the sciatic nerve injury-induced increment of BDNF and NGF expressions. c-Fos expressions in the PVN and vlPAG were increased following crushed sciatic nerve injury and treatment with the AJ significantly suppressed the sciatic nerve injury-induced increment of c-Fos expressions. Conclusions: These results suggest that AJ treatment after crushed sciatic nerve injury is effective in the functional recovery by enhancing axonal regeneration and suppressing of pain.

      • CA약침의 Sprague-Dawley Rats에 대한 단회 근육투여 독성시험

        Objectives : The purpose of this study was to investigate the toxicity and to determine approximate lethal dose of the CA-YAKCHIM, KOREA IMMUNO-YAKCHIM SOCIETY, following a single intramuscular injection to 6-week old male and female Sprague-Dawley rats. Methods : Test groups consisted of one dosing group at a dose of 1.0 mL/animal and one control group (saline), and 5 males and 5 females were assigned to each group. All animals were monitored for clinical signs and body weight changes during the 14-day observation period after administration. They were subjected to gross necropsy at the end of the observation period. At necropsy, necropsy findings and local tolerance test at the injection site were conducted. Results : All animals in the 1.0 mL/animal group survived the duration of the study. No CA-YAKCHIM-related effects were evident in clinical signs and necropsy findings in any animals in the 1.0 mL/animal group. In local tolerance test at the injection site, no CA-YAKCHIM-related effects were evident in any animals in the 1.0 mL/animal group. Conclusions : In this study, the approximate lethal dose of the CA-YAKCHIM was considered to be greater than 1.0 mL/animal in male and female rats.
