황칠(黃漆)이 만성 스트레스 유발 백서의 스트레스 및 수면 호르몬에 미치는 영향

Objectives: To evaluate effects of Resina Dendropanacis Morbiferus (RDM) on stress and sleep hormones when administered to Chronic Mild Stressed (CMS) rats. Methods: Twenty-five healthy rats were randomly divided into 5 groups; Normal, CMS (Control), Resina Dendropanacis Mobiferus 50 mg/kg (RDM 50), 100 mg/kg (RDM 100), and 200 mg/kg (RDM 200). All rats except the normal group were exposed to unpredicted stress conditions such as water deprivation, empty bottles, forced thread mill, etc. according to timetable of CMS for 3 weeks. After a week starting experiments, rats in RDM 50, RDM 100, and RDM 200 groups were fed orally once a day for 2 weeks. Afterward, blood samples were taken from rats to analyze complete blood count, AST, ALT, and glucose. Noradrenalin, GABA and Melatonin were measured by ELISA kit. BDNF, CREB and TrkB were measured by RT-PCT. Results: 1. In Noradrenalin content, RDM 100 and RDM 200 groups revealed significant decrease compared to the control group. 2. In GABA content, RDM 50 and RDM 100 groups revealed significant decrease compared to the control group. 3. In Melatonin content, RDM 100 and 200 groups revealed significant decrease compared to the control group. 4. In Activity of BDNF, RDM 100 and 200 groups revealed significant increase compared to the control group. 5. In Activity of CREB and TrkB, RDM 100 group revealed significant increase compared to the control group. 6. In Erythrocyte and Thrombocyte changes, red blood cells and hematocrit significantly increased in RDM 50 and RDM 200 groups than the control group. Hemoglobin and platelet significantly increased in all experimental groups and the control group. 7. In Weight gain content, all RDM groups revealed insignificant increase compared to the control group. 8. In Glucose content, RDM 50, RDM 100, and RDM 200 groups revealed significant decrease compared to the control group. Conclusions: Results suggest that RDM have effects on stress and sleep hormones when administered to Chronic Mild Stressed (CMS) rats.

Background: Numerous reports suggest the role of oxygen in melanogenesis. However, little has been reported on the effect of a hypoxic environment on cellular melanogenesis. Objective: The effect of low oxygen tension on cellular melanogenesis was investigated in B16 murine melanoma cells. Methods: Using cells cultured under an ambient (21% O2) or hypoxic (5% O2) condition, melanin content and tyrosinase activity were measured spectrophotometrically. The expression of tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 were analyzed by RT-PCR and Western blot. Results: Culture of cells under hypoxic conditions caused significant inhibition of isobutylmethylxanthine (IBMX)-induced increase of melanin content. No cytotoxicity was observed during the hypoxic culture periods. Decreased melanin content occurred through the decrease of tyrosinase protein and activity (p<0.01). The mRNA levels of tyrosinase and TRP-2 were also decreased by hypoxia, while that of TRP-1 was unchanged. Similar inhibitions of melanin content and tyrosinase activity were observed in the cells stimulated with dibutyryl-cAMP. Conclusion: IBMX-induced melanogenesis in B16 cells was significantly inhibited under hypoxic culture conditions, suggesting the important role of oxygen tension in cellular melanogenesis. 반하후박탕(半夏厚朴湯)의 사회, 심리적 스트레스에 대한 실험적 연구(實驗的 硏究)

Objectives: This study is designed to investigate the effects of Banhahubak-Tang Extract (BHTe) on stress. Methos: BHTe was fed to ICR male mice (20±2 g) orally with the dose of 100 mg/kg/day for five days. Mice were then exposed to sociopsychological stress by observing foot shock stressed mice for one hour for five days while restrained. Results: 1) The BHTe-administered group showed a tendency of decreasing of serum corticosterone secretion compared with the control group. 2) The BHTe-administered group showed an increase in noradrenalin secretions in the dorsal cortex of the brain, but it was not significant. 3) BHTe administration had no effect on the brain level of lipid peroxidation. 4) BHTe administration decreased the serum level of lipid peroxidation. 5) BHTe administration decreased the Cu,Zn-SOD in the brain. 6) BHTe administration had no effect on catalase activity in the brain. 7) BHTe administration had no effect on the brain level of GSH. 8) BHTe administration increased the serum level of DPPH free radical scavenging activity, but not significantly enough to make a comparison with BHT. 9) The elevated plus-maze test is designed to detect the effect of anxiolytic drugs. The BHTe group showed a significant increase in latency time. Conclusions: These results suggest that BHTe can effectively rid the subject of the effects of sociopsychological stress.
반하(半夏)가 스트레스로 인한 생쥐의 뇌조직 유전자변화에 미치는 영향 연구

Objectives: Pinelliae Rhizoma has traditionally been used as an anti-depressant in oriental medicine. This study is to investigate the effect of Pinelliae Rhizoma extract (PRe) on psychological stress in genome wild expression of mice. Methods: After giving physical stress to mice, PRe was orally administered with 100 mg/kg/day for five days. After extracting whole brain tissue from the mice, their genome changes were observed by micorarray analysis method. The genome changes were analyzed by IMAGENE 4.0, TREEVIEW, FatiGo algorithems, BOND database, cytoscape program, etc. Results: 1. PRe administered group were remained at normal level; 60% of increase was shown in expressed genes by physical stress, and 65% of decrease was shown in expressed genes by psychological stress. 2. Genes with increased expression in control group that remained at a normal state in PRe administered group were involved with the gene of a cellular metabolic process on biological process, protein binding on molecular function, and cell part on cell composition. The pathway was found to be cytokin-cytokin receptor interaction. 3. Genes with decreased expression in control group that remained at a normal state in PRe administered group were involved with the gene of a cellular metabolic process on biologiacl detail and coupled ATPaes activity on molecular function. This gene related path was Ubiquintin mediated proteolysis etc. 4. Core node genes analyzed by protein interaction network were Vinculin, Cell sdivision cycle 42 homolog (S. cerevisiae) etc. They played an important role in maintaining cytoskeleton and controlling cell cycle. Conclusions: Several genes were up-regulated and down-regulated in response to psychological stress. The expression of most of the genes that were altered in response to psychological stress was restored to normal levels in PRe treated mice. When the interaction network information was analyzed, the recovery of the core node genes in PRe treated mice indicates that this final set of genes may be the effective target of PRe.

A study was conducted to design automotive thermal energy storage for reduction in exhaust emission and enhancing the passenger's comfortability. Ba(OH)2 8H2O was chosen, based on its thermodynamic characteristics and a series of test, as a base material for the energy storage system. Sr(OH)2 8H2O and H2O were added to the base material to protect degradation that is appeared during the cycling test of the pure base material. Thermal storage were, then designed to provide 2,000 kJ of energy within a few minutes of engine ignition with the computer simulation results.
