        NIH3T3 세포에서 UVB에 의한 세포고사에 미치는 옻 추출물과 fisetin의 효과

        For a long time Rhus vemiciflua Stokes (RVS) has been traditionally used as a herbal plant in Asia. In this study, we have investigated the effect of acetone extract of Rhus verniciflua Stokes (RVSE) and fisetin, a component of RVSE, on DNA damage response in NIH3T3 cells. Exposure of cells to DVB light $(200 J/m^2)$ and postincubation in growth medium for 48 hr resulted in a decrease of cell viability to about $10-20\%$ of nontreated control. Addition of various concentrations of RVSE in the postincubation medium, however, significantly increased the cell viability as compared with the values expected. The genotoxicity-decreasing effect was also demonstrated in cells exposed to UVB light and incubated in medium containing fisetin. The genotoxicity-decreasing effect of RVSE and fisetin was further demonstrated by various analyses including cell morphology studies, trypan blue exclusion assay and DAPI staining. By Annexin V binding analysis, RVSE and fisetin were shown to decrease the early apoptosis induced by UVB exposure. These results suggest the RVSE contain components that either increase the DNA repair or decrease the apoptosis in UVB-exposed cells.

      • Azide/alkyne 고리화 반응에 의한 Polydiacetylene Vesicle의 색 전이

        Polydiacetylenes (PDA) have been widely investigated as smart sensing materials because of their unique optical properties. PDA showed the color change from blue to red by some external stimulus. Due to this property, PDAs have been widely studied as a sensing material in chemistry and biological industry. In this study, in order to investigate the sensing mechanism of PDA, the color change of PDA vesicle system by azide/alkyne cycloaddition reaction was conducted. 4-Methoxyphenyl azide, benzyl azide, and 1-pyrene azide were used as the azide compounds, and PDA vesicle system was fabricated by sonication using 10,12-pentacosadiyoic acid (PCDA) as a monomer. The results showed that the color change showed clearly. And the strength of color change depending on the adding amount of azide and the reaction time was detected. The chemical structure of the azide had a strong effect to the color change. Because 1-pyrene azide has the large aromatic groups, it showed very slow reaction rate. Conclusively speaking this reaction could be the diffusion controlled reaction.

        NIH3T3 세포에서 UVB에 의한 세포고사와 DNA 단사절단에 미치는 fisetin의 효과

        본 연구에서는 UVB에 조사된 NIH3T3 세포에서 세포고사와 DNA 단사절단에 미치는 fisetin후처리의 효과에 대해서 연구하였다. 세포에 UVB $(200J/m^2)$를 조사하고 정상배지에서 48시간 배양한 세포의 세포고사에 수반되는 핵분절은 50% 정도의 세포에서 관찰되었다. 흥미롭게도 배양배지에 fisetin이 첨가될 경우 핵분절을 보이는 세포의 빈도는 상당한 감소를 보였다. 알칼리 아가로스 겔에 의한 DNA 단사절단 분석에서 자외선 조사 후 fisetin처리는 정상배지 배양시보다 단사절단의 빈도를 감소시켜 DNA크기의 증가를 유도하였는데 이는 fisetin이 UVB에 의한 DNA 상해의 회복에 긍정적 효과를 나타냄을 시사한다 Western blot 분석에 의해 fisetin은 자외선 조사에 의해 활성화되는 p53의 수준을 유의한 수준으로 감소시키며 자외선 상해의 결과 세포주기의 정지에 수반되는 PCNA의 감소 경향을 다소 완화시켰다. 이러한 결과들은 fisetin이 DNA 회복의 활성을 통해 세포고사의 감소에 기여하며 이 과정에서 p53 및 PCNA의 수준변화와 관련하여 행동함을 시사한다. In the present study, we have investigated the effect of fisetin on the apoptosis and DNA single strand breaks in ultraviolet light B (UVB)-exposed NIH3T3 cells. Exposure of cells to UVB light $(200J/m^2)$ and post-incubation in growth medium for 48 hr resulted in about 50% of cells with apoptotic nuclear fragmentation. Addition of various concentrations of fisetin in the postincubation medium, however, significantly reduced the apoptotic nuclear fragmentation as compared with the values expected when the effects are additive and independent. DNA single strand breaks induced by UVB exposure were also significantly decreased by postincubation with fisetin. By Western blot analysis, fisetin post-incubation was shown to attenuate the p53 upregulation upon UVB exposure. Furthermore, the decrease of proliferating cell nuclear antigen (PCNA) level upon UVB exposure was alleviated by fisetin postincubation. These results suggest that fisetin decrease the apoptosis and increae DNA repair in a possible association with alteration of p53 and PCNA levels in UVB-exposed cells.

        전기방사에 의한 폴리아미드/다중벽 탄소나노튜브 나노복합체섬유의 제조

        This study used a new and simple processing route to incorporate multiwalled carbon nanotubes (MWCNTs) into a polymer matrix via in-situ interfacial polymerization and electrospinning. Poly(hexamethylene sebacamide) (nylon 610)/MWCNT composites were prepared using two liquid phases containing the reactants and MWCNT. One phase contained a solution of sebacoyl chloride in carbon tetrachloride and the other contained a solution of hexamethylenediamine and MWCNT in water, which was prepared by sonication with a surfactant. Electrospinning of the composite solution produced fibers with well embedded MWCNT. We investigated morphology of individual MWCNT incorporating in nylon 610 fibers by scanning electron microscopy and transmssion electron microscopy and the electrical conductivity of the electro spun nylon 610 fibers with MWCNT.
