        Impact of White Matter Changes on Activities of Daily Living in Mild to Moderate Dementia

        <P>The association between white matter changes and activities of daily living (ADL) in a large, well-defined cohort of patients with mild-to-moderate dementia (either Alzheimer’s disease or subcortical vascular dementia) were investigated. A total of 289 patients were divided into three groups (140 mild, 99 moderate, and 50 severe) depending on the degree of white matter changes as indicated on brain magnetic resonance image scans. Further, we analyzed the three groups’ performances on basic and instrumental ADL. The degree of white matter changes was associated with greater age, hypertension, previous history of stroke, higher Hachinski Ischemic Score, worse global cognitive and functional status, and an increased impairment of basic ADL and instrumental ADL. The increased impairment with regard to the severe group’s performance on both the basic and instrumental ADL remained significant after adjustment for age and hypertension. Tasks involving physical activities were most significant. This was the first study investigating the association between white matter changes and ADL in a large, well-defined dementia cohort. The present study suggests that severe white matter changes may be associated with higher impairment on both basic and instrumental ADL.</P><P>Copyright © 2010 S. Karger AG, Basel</P>

        Changes in S-allyl cysteine contents and physicochemical properties of black garlic during heat treatment

        This study aimed to investigate the effects of temperature on the black garlic manufacturing process. The moisture content, pH, browning intensity, S-allyl cysteine (SAC) content and antioxidant activity, including DPPH radical scavenging activity and reducing power, were determined. The moisture content of garlic gradually decreased throughout the heating process. The rate of moisture removal was higher at high temperatures compared with low temperatures. The pH also decreased more significantly in garlic heated at high temperatures. The browning intensity increased with increasing temperature. The SAC contents of black garlic were significantly different according to heating temperature; the garlic samples heated at a low temperature had a higher SAC contents. Antioxidant activity, as determined by the DPPH radical scavenging activity and reducing power, increased when the garlic was exposed to higher temperatures.

        A comparative study of the different analytical methods for analysis of S-allyl cysteine in black garlic by HPLC

        A sensitive and reproducible analytical method to measure S-allyl cysteine (SAC) in black garlic was developed using the HPLC-FLD (fluorescence detection) with prior derivatization with AccQ-Fluor Reagent. The results were then compared with those from obtained using the HPLD-UVD (ultraviolet detection) that has been conventionally used for measuring SAC in raw garlic. The SAC content in raw garlic measured by the HPLC-FLD and the HPLD-UVD were 21.52 and 22.73μg/g, respectively. And these results were satisfactory with %CV (coefficient of variation) less than 2%. However, the application of the HPLC-UVD for the determination of SAC in black garlic was limited due to the high %CV that arose from the unstable baseline. However, the HPLC-FLD could be applied to the measurement of SAC in black garlic with satisfactory %CV. The HPLC-FLD method was validated by determining the linearity, limit of detection, limit of quantification, accuracy and precision. This method showed excellent linearity with a coefficient of determination (R<SUP>2</SUP>) greater than 0.998. The limits of detection (LOD) and the limit of quantification (LOQ) were 6.28 and 19.02μg/mL, respectively. The accuracy ranged from 98.51 to 102.08%, and the precision of the method was found to be less than 2%.

      • The effects of topical mesenchymal stem cell transplantation in canine experimental cutaneous wounds

        <P><B>Background</B></P><P>Adult stem cells have been widely investigated in bioengineering approaches for tissue repair therapy. We evaluated the clinical value and safety of the application of cultured bone marrow-derived allogenic mesenchymal stem cells (MSCs) for treating skin wounds in a canine model.</P><P><B>Hypothesis</B></P><P>Topical allogenic MSC transplantation can accelerate the closure of experimental full-thickness cutaneous wounds and attenuate local inflammation.</P><P><B>Animals</B></P><P>Adult healthy beagle dogs (<I>n</I> = 10; 3–6 years old; 7.2–13.1 kg) were studied.</P><P><B>Methods</B></P><P>Full-thickness skin wounds were created on the dorsum of healthy beagles, and allogenic MSCs were injected intradermally. The rate of wound closure and the degree of collagen production were analysed histologically using haematoxylin and eosin staining and trichrome staining. The degree of cellular proliferation and angiogenesis was evaluated by immunocytochemistry using proliferating cell nuclear antigen-, vimentin- and α-smooth muscle actin-specific antibodies. Local mRNA expression levels of interleukin-2, interferon-γ, basic fibroblast growth factor and matrix metalloproteinase-2 were evaluated by RT-PCR.</P><P><B>Results</B></P><P>Compared with the vehicle-treated wounds, MSC-treated wounds showed more rapid wound closure and increased collagen synthesis, cellular proliferation and angiogenesis. Moreover, MSC-treated wounds showed decreased expression of pro-inflammatory cytokines (interleukin-2 and interferon-γ) and wound healing-related factors (basic fibroblast growth factor and matrix metalloproteinase-2).</P><P><B>Conclusion and clinical importance</B></P><P>Topical transplantation of MSCs results in paracrine effects on cellular proliferation and angiogenesis, as well as modulation of local mRNA expression of several factors related to cutaneous wound healing.</P><P><B>Résumé</B></P><P><B>Contexte</B></P><P>Les cellules souches adultes ont été largement étudiées dans les approches de bio-ingénierie pour la thérapie de réparation tissulaire.         Phenotypic characteristics of<i>Streptococcus iniae</i>and<i>Streptococcus parauberis</i>isolated from olive flounder (<i>Paralichthys olivaceus</i>)

        <P>The etiological agents of streptococcosis were isolated from diseased olive flounder collected on the Jeju island of Korea. A total of 151 bacterial isolates were collected between 2003 and 2006. The isolates were examined using various phenotypic and proteomic analyses, including sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), immunoblotting, and glycoprotein assays. In addition, isolates were grown on blood agar to assess hemolytic activity, and biochemical assays were performed using the API20 Strep kit. Our results revealed that all isolates were nonmotile, Gram-positive cocci that displayed negative catalase and oxidase activities. Multiplex PCR assays revealed that 43% and 57% of the isolates were Streptococcus iniae and Streptococcus parauberis, respectively. These results were consistent with those of the SDS-PAGE and immunoblot analyses using whole-cell lysates of bacterial isolates. Significant differences were observed with respect to the Voges-Proskauer, pyrrodonyl arylamidase, alkaline phosphatase, and hemolytic activities of the S. iniae and S. parauberis isolates. Isolates of S. iniae displayed uniform profiles in the immunoblot and glycoprotein assays; however, immunoblot assays of S. parauberis isolates (using a chicken IgY antibody raised against a homologous isolate) revealed three distinct antigenic profiles. Our findings suggest that S. parauberis and S. iniae are endemic pathogens responsible for the development of streptococcosis in olive flounder.</P>

        A study on the status of disease in a Korean rural Community, Su-Dong Myun, Yong Ju Gun, Kyung-Gi Do was carried out from December, 28 1978 to January, 19 1979 through analizing the meclical records of Su-Dong Health Subcenter during the period september, 1977 to August, 1978 and following results were obtained. 1. New cases of out patients were 5,695 composed of 49.1% male and 50.9% female during the study period. 2. Cumulative cases of out patients were 9,956 composed of 56.4% male and 43.6% female during the study period. 3. Annual morbidity rate was 908.6 per 1,000 for male and 986.4 for female and average annual morbidity rate was 946.6 per 1,00 population. 4. The highest morbidity rate was 3,660.0 as composed of aged 0∼4, the lowest was 498.4 as composed of aged 10∼14 per 1m000 population per year. 5. Following was the order of five major disease which were classified with I.C.D. ① Diseases of the respiratory system : 33.4% ② Diseases of the digestive system : 21.5% ③ Diseases of the skin and subcutaneous tissue : 15.4% ④ Diseases of the nervous system and sense organs : 7.9% ⑤ Accidents, poisonings and violence : 9.4% 6. The visiting rate in the health subcenter were 2.01 for male, 1.44 for female and the average 1.75 per each out patient. 7. Average medical fee per new case was 653±510 won and average cumulatived fee per case was 923±833 won. 8. The distribution of out patients according to the cost of medical fee, 48.9% of total new patients was treated with less than 500 won and 93.3% of new patients was treated with less than 1,000 won.

        저장벽 양자우물구조와 비태칭 패브리 - 페로 공명기 구조에 의한 고성능 2×4 S-SEED Array 구현

        GaAs/Al_(0.04)Ga_(0.96)As 다중양자우물 구조를 이용한 반사형 PIN 다이오우드 S-SEED의 설계에 있어 낮은 동작전압, 높은 포화에너지 및 높은 반사율 on/off 강도비를 얻고자 저장벽 양자우물구조와 비대칭 페브리-페로 공명구조를 결합하였다. 2×4 array를 구성하는 S-SEED들은 역방향 동작전압 5V에서 평균적으로 13 이상의 반사율 on/off 강도비 (CR)와 약 24%의 반사율차 (ΔR) 및 91% 이상의 광쌍안정폭 (Δ)을 나타내었다. 공명구조를 이용함으로서 PIN 다이오우드 진성영역내의 양자우물의 주기수를 줄일 수 있어 외부동작전압 없이도 CR~4.7, R~9.2%, ~22%의 향상된 무전압 광쌍안정 동작특성을 얻었다. We designed and fabricated a 2×4 symmetric self electro-optic effect device array using GaAs/Al_(0.04)Ga_(0.96)As extremely shallow quantum wells grown by a molecular beam epitaxy. By employing ESQW and asymmetric Fabry-Perot cavity structure simultaneously, we improved the performances of S-SEED such as on/off contrast ratio (CR), reflectivity change (ΔR), and optical bistability loop width (Δ). The average values of the elements of the 2×4 S-SEED array were CR~13.1, R~24%, and Δ~91%. It was found that the AFP cavity structure enhances the self-biased optical bistability in ESQW-SEED under no external bias. That is due to the decreased intrisic region thickness in AFP-SEED structures, and which increases the built-in electric fields. The zero-biased S-SEED showed CR of ~4.7, R~9%, and Δ~22%.
