        구강편평세포암종에서 미슬토 추출물의 Apoptosis 유도 효과에 대한 실험적 연구

        This study was performed to investigate mistletoe extract-induced apoptosis in oral squamous cell carcinoma. In vivo study, HN22 cells were xenografted in nude mice. After tumor was experimentally induced, mistletoe extract was directly injected on the tumor mass. The specimens were evaluated using light and transmission electron microscopes. In vitro study, HN22 cells were cultured and exposed to mistletoe extract. The cells were evaluated using transmissin electron microscope. To evaluate apoptotic cells, flow cytometric analysis was done. The results were obtained as follows: 1. Light microscopic view of tumor mass showed necrosis at 2-4 weeks. 2. Transmission electron micrographs of tumor mass showed apoptosis and necrosis. 3. In TEM view of cell lines, necrosis and apoptosis were shown with mistletoe extract at 300μg/ml, apoptosis was shown with mistletoe extract at 100μg/ml. 4. In flow cytometric analysis, early and late apoptosis was shown when using caspase-3Ab and annexin-V, but no significant change was noted when using mebstain and Apo2.7 Ab. In this study, mistletoe extract induced necrosis and apoptosis in the tumor mass was induced by HN22 cells, early and late apoptosis in vitro study. Mistletoe extract was likely to induce cell death in oral squamous cell carcinoma through apoptosis.

      • 에스트로겐 투여가 난소절제로 유도된 백서의 골다공증에 미치는 효과에 관한 실험적 연구

        Osteoporosis is a state of reduced bone mass per unit volume with a normal ratio of mineral to matrix. There were many methods for osteoporosis, and estrogen is one of them. In this study, 30 female rats were ovariectomized to make a state of estrogen deficiency and induce osteoporosis. After 13 weeks, 15 ovariectomized rats were injected 6 times with 60㎍/㎏ of estradiol by 1 week interval (experimental group). The remaining 15 ovariectomized rats were used as control group. According to the time schedule, both of them were sacrified and femur were harvested. Paraffin sections were stained with HematoxylinEosin staining and modified Gomori's Trichrome staining, and observed under light microscope. The results were as follows : 1. There were no significant bony changes between the experimental and control group of cancellous bone. 2. The density of cortical bone showed increasing with time in experimental group. 3. The resorption of cortical bone in experimental group was less than that of control group.

      • 가토 편측 악관절원판 제거후 악관절 과두에서 S-100 단백 변화에 관한 실험적 연구

        S-100 protein is chiefly in neuroectodermal origin tissue of human body. For example it exists in odontogenic epithelial cell, melanocyte in skin, chondrocyte in cartilage, and it is involved in calcium metabolism. The purpose of this study was to observe the change of cell proliferation of condylar head after unilateral diskectomized by anti S-100 protein antibody method. 20 rabbits of 3.0 Kg body weight(control;5, experimental;15) were used and diskectomized in left side of experimental group. According to the sacrification schedule, excised TMJ of control and experimental group was fixed, decalcified and immunohistochemically stained by LSAB method. S-100 protein positive cells were observed under the microscope according to stainability. The results were as follows, 1. Histopatholigic features showed increased hyperplasia of hypertrophic zone, but decreased narrowness of errosive zone in surgical site. 2. S-100 protein positive cells could be seen in the hypertrophic zone of control group. In experimental group, they were seen in hyalinized chondrocyte at 1 week, from 3 to 6 weeks they were found at the borderline of proliferative and hypertrophic zone. 3. In 8 weeks of experimental group, positive reaction was appeared in upper portion of hypertrophic and erosive zone. In 10 weeks of experimental group, slight positive reaction in hypertrophic zone was similar to control group. From the above findings, we can observe the change of undifferentiated cell into chondrocyte at early stage of diskectomized condyle head. After 10 weeks, narrowing of proliferative zone and widening of hypertrophic zone and cartilagenous cell megration into subchondral bone could be seen.
