A Novel Method for Continuous Synthesis of Bicelles Using Microfluidic Chip

Bio-membranes consist of phospholipids. Phospholipids are self-assembled in an aqueous solution to compose the bilayer membrane. Bicelle is one of the self-assembled structures of phospholipid with discoid shape. Due to its great potentials in bio-membrane research, it has been dealt with as an important subject in many studies. The conventional methods for the preparation of the bicelle include many steps of the process and harsh conditions, which limit its feasibility and stress the biological substances. For these reasons, a continuous manufacturing method in mild condition has been required. Here, we propose a novel method for bicelle synthesis based on a hydrodynamic focusing microfluidic system. Bicelles were successfully prepared using this continuous method, which was confirmed by the physicochemical properties and morphologies of the synthesized assemblies. By experimental and analytical studies, it was found that there is critical lipid concentration and critical mixing time for bicelle synthesis in this microfluidic system. Furthermore, a linear relationship between the actual composition of bicelle and the initial lipid ratio is deduced, which enables size control of the bicells.
