      • 유전자지문법을 이용한 사상체질의 유전적 분석 연구

        VNTR 및 STR 분석은 유전자지문법 (DNA fingerprinting)에서 사용되는 방법으로 개체의 식별 등을 목적으로 하는 범죄수사 및 기타 유전적 분석 연구에서 많이 쓰이고 있는 유전적 분석 방법이다. 본 연구는 사상체질의 판별에 의해서 분류된 각 체질들이 유전적으로 차이가 있는지를 조사하기 위하여 각 체질인의 제놈 DNA를 대상으로 VNTR 및 STR의 분석을 실시하여서 사상체질의학의 객관화 및 그 임상활동에 관한 유용한 자료를 제공함을 목적으로 추진되었다. 본 연구에서 실시한 MCT118, YNZ22 locus를 이용한 VNTR 분석에서는 체질별로 나타난 allele의 분포 양상이 다양할 뿐만 아니라 각 체질에서 나타난 allele의 분포빈도가 체질별로 유의적인 차이를 보이지 않았다. 따라서 VNTR의 분석을 이용하여서는 사상체질의 네 가지 체질 그룹에 대한 유전적 동질성 및 그룹간 유전적 상이성을 유추해 내기가 어려운 것으로 사료되었다. 한편 allel 의 종류가 적고 따라서 그 분포 양상이 VNTR에 비해서 다양하지 않은 STR 중에서 본 연구에서는 TH01과 vWA locus에 대한 분석을 실시하였는데 그 결과 vWA locus에서는 각 allele No.의 분포 양상이 체질별로 다소 다르게 나타났다. VNTR and STR DNA typing are typical genetic analysis methods which are widely used in DNA-fingerprinting for forensic science and other genetic research purposes. In this study, genomic DNA of different constitutions(Taeun, Soyang and Soum) were analyzed by VNTR and STR DNA typing to provide scientific and objective references for Sasang Medicine. It was found out in this study that VNTR-MCT118 and YNZ22 loci showed too many different variation of allele distribution and numbers for each constitution. Therefore, it is thought that VNTR typing can not used for genetic classification study for Sasang Constitution which classifies human body into 4 groups. However, vWA locus, one of the STR loci investigated in this study, showed slight difference in allele distribution for each different constitution.

        In the post-genome era. analysis of the cellular transcriptome using microarray or the cellular proteome using a 2-D gel electrophoresis and MALDI-TOF mass spectrometry are most widely used. Stroke is one of the most important causes of death along with cancer and cardiac disease. When pathological change of cells in developed from cerebral ischemia accompanied by stroke administration of neuroprotective drugs before stroke can decreases the degeneration of neuronal cells. The purpose of the present study was to assess the neuroprotective effect and protein expression after administration of P004, middle cerebral artery model of cerebral ischemia in rats. SD rats were subjected to middle cerebral artery occlusion. P004 (1.000 ㎎/㎏) was administered 2 times at 0, 90 minutes after middle cerebral artery occlusion (MCAo). Rats were killed at 48 hours, and infarct area and volume were determined by histology and computerized image analysis. We investigated the protein expression profile on the global ischemia induced by MCAo. This proteomic analysis enable us to identify several proteins differently expressed in infarct brain tissue. The aims of this study were to do investigation comparing the neuroprotection activities of P004 and to understand the mechanism of acted as neuroprotective drug.

      • 사상의학의 객관화를 위한 유전적 분석 연구

        This main idea of Sasang medicine is that everyone has his own unique constitution. There are four kinds of constitutions and these are Taeyang, Soyang, Teaum and Soum. Although Sasang medicine is a unique and originative medical theory with creative ideas of Korean traditional medicine, the classification of individual constitution mainly depends on the methods which may not be objectively proven and scientifically understood. This study was carried out to establish scientific understanding of Sasang medicine by investigating the possible genetic difference among four constitutions. Sasang constitution classification was performed by Sasang medicine research group in Korea Institute of Oriental Medicine and Kyung Hee University. Genomic DNA ws prepared from the blood of individuals of different constitutions and RAPD(Random amplified polymorphic DNAs) analysis was then carried out to investigate the possible defference among different constitutions on DNA level. For RAPD analysis, then different random primers were applied to Teaum, Soyang and Soum group DNA samples and the presence of DNA markers specific for each constitution was investigated.

      • 허혈/재관류 환경하에서 청폐사간탕의 염증 관련 반응 억제 효과

        Chungpesagan-Tang (CST), a Korean traditional prescription composed of oriental medical herbs, has been used successfully to improve human health and regimen. This study was designed to examine the inhibitory effects of CST against in vitro ischemia/reperfusion-induced inflammatory response. We have characterized the production of prostaglandin E₂and arachidonic acid during ischemia/reperfusion in the human neuroblastoma SK-N-MC and human monocytic macrophage U937 cells and the inhibitory effect of CST on these inflammation-related substance formation has been found out in this study. This result suggested that CST used in this experiment reinforced formation has been found out in this study. This result suggested that CST used in this experiment reinforced antiinflammatory potentials and protected cells against ischemia/reperfusion-induced inflammatory response.

      • 한약내의 리튬이 타이로신 가수분해효소에 미치는 영향에 대한 연구

        Lithium, a simple monovalent cation, is the mainstay in treatment of manic-depressive illness, but despite extensive research, its mechanism of action remains to be elucidated. Because lithium requires chronic administration for therapeutic efficacy and because its beneficial effects last well beyond its discontinuation, it has been postulated that lithium may exert major effects at the genomic level. We have previously shown that Yungmijihwanwon(YM) increase TH protein expression through antioxidant activity and unknown effects in vivo. In the present study, we have sought to determine if lithium in herbal medicine also increases the expression of endogenous gene known to be regulated by AP-1 and have therefore investigated the effects of herbal medicine on tyrosine hydroxylase(TH) levels. Male mice were treated with LiCI and herbal medicine for 30 days, and TH levels and dopamine level were measured in striatum using immunoblotting and ECD-HPLC. Herbal medicine treatment resulted in statistically nonsignificant increase in TH levels in mouse striatum. The precise therapeutic relevance of these effects is presently unknown.

      • 4-Vessel Occlusion 허혈동물모델에서의 대규모 유전자 발현 연구

        Cerebral ischemia, the most prevalent form of clinical stroke, is a medical problem of the first magnitude. Substantial efforts are being made to develop drugs which will protect the brain from the neurodegeneration followed by an ishemic stroke. A key factor in this process is the increasing an evidence that free radical is involved in the mechanisms of ischemic brain damage. We investigated the macro scale gene expression analysis on the global ischemic induced by 4-vessel occlusion in Wister rats. The recent availability of microarrays provides an attractive strategy for elaborating an unbiased molecular profile of large number of genes during ischemic injury. This experimental approach offers the potential to identify molecules or cellular pathways not previously associated with ischemia. ischemia was induced by 4-vessel occlusion for 10 minutes and reperfused again. RNA from sham control brain and time-dependent ischemed brain were hybridized to microarrays containing 4,000 rat genes. 589 genes were found to be at least 2 fold regulated at one or more time points. These survey data provide the foundation studies that should provide convincing proof for ischemia and oxidative stress on gene expression.
