Olive flounder (Paralichthys olivaceus) has a great potential value for aquaculture in Korea. The theoretical sex ratio of this flounder is close to 1:1 as it is in many other type of fish. However, according to the size selection during artificial seedling production, there is a possibility of unbalanced sex ratio. The actual flounder culturing needs female seedlings because of faster growth than male in economically. However, little is known about the sex ratio of cultured flounder. The six populations consisted of three wild populations and three cultured populations were analyzed for investigating the sex ratio. The sex ratio of wild populations ranged from 1.10 to 1.93 in female and 0.52 to 0.91 in male. And the sex ratio of cultured populations ranged from 0.20 to 2.15 in female and 0.46 to 4.88 in male. This result indicated that the sex ratio of cultured flounder varies depending on the size selection method.
넙치, Paralichthys olivaceus 자연 집단과 양식 집단의 유전학적 다양성 비교

넙치 양식에 이용되는 종묘는 수 세대에 걸친 양식용 어미로부터 생산된 것으로 알려져 있어 넙치 양식 집단의 유전적 다양성은 자연 집단에 비해 크게 낮을 것으로 추정되고 있다. 본 연구는 우리나라에서 양식되고 있는 양식 집단의 유전학적 다양성이 어느 정도인지를 파악하기 위하여 6개의 microsatellite DNA marker를 이용하여 자연 집단과비교하였다. 본 조사에서 관측된 유전자좌별 대립유전자 수의 범위는9~27개였으며, 6개의 유전자좌에 따른 각 집단별 평균대립유전자 수는 자연 집단이 19.7~21.8개, 양식 집단이12.0~14.7개로 나타나, 자연 집단이 21.1개로 양식 집단의전체 평균 9.4개보다 높게 나타났다. 조사된 집단들의 이형접합체율(Ho)의 범위는 0.722~0.959이었으며, 유전자좌별Ho의 범위는 자연 집단에서 0.755~0.918, 양식 집단에서0.722~0.959로 자연 집단과 양식 집단 간 커다란 차이는없었다. 각 유전자좌에 대한 집단별 Markov chain procedure test 를 한 결과, 자연 집단에서는 HWE를 따르는 것으로 나타났으나, 울진 및 완도 양식 집단의 일부 마커들에서 세대를거듭하면서 양식 집단의 유전학적 조성 등이 유전적 평형에서 벗어나고 있는 것으로 나타났다(P⁄0.05). 또한 자연집단인 추자 집단이 양식 집단과 유전적 거리가 가깝게 나타남으로써 방류된 개체들이 상당 부분 자연 집단의 일부로자리 잡아 자연 집단의 유전적 조성에 영향을 주는 것으로판단된다. 따라서 양식 집단의 유전적 다양성 회복 및 자연집단의 유전적 조성의 유지, 보호 등에 대한 지속적인 연구가 필요한 것으로 생각된다. Six microsatellite DNA markers were used to investigate the genetic variability between wild populations and cultured stocks of olive flounder Paralichthys olivaceus. The average of observed (Ho) and expected heterozygosity (He) ranged from 0.722 to 0.959, and from 0.735 to 0.937,respectively. There was no distinguishable difference between the wild populations and cultured stocks in terms of the observed and expected heterozygosities. However, number of alleles per locus differed markedly between the two fish groups: 19.7 to 21.8 for the wild populations and 12.0 to 14.7for the cultured stocks. This result gives important information concerning the production of seedling for the improvement of genetic diversity in this species.
Apostichopus japonicas (Echinodermata; Holothuroidea)에서 온도 스트레스에 의한 Hsp90 및 Ferritin 유전자의 발현

The Apostichopus japonicus is an important species in some Asia countries including Korea, China and Japan. The purpose of the present study was to investigate the differential gene expression of heat shock protein90 (Hsp90) and ferritin as a biomarker for the thermal stress during water temperature rising in the sea cucumber, A. japonicus. The A. japonicus (1.4 g) was cultured in incubator of separate temperature (15°C, 20°C, 25°C and 30°C) for each 0, 3, 6, 12, 24, 48 hours. The mRNA expression levels of Hsp90 and ferritin were examined using RT-PCR assay. Results showed that, the expression of Hsp90 mRNA was not significantly changed at 15°C. The expression of Hsp90 mRNA was significantly increased at high temperature such as 20°C and 25°C. Furthermore, Hsp90 mRNA was early increased at 25°C than 20°C. The ferritin mRNA was similar expression pattern with Hsp90. But, Hsp90 mRNA was more sensitive than ferritin mRNA at high thermal stress. These results indicate that Hsp90 and ferritin mRNAs were involved in the temperature changes response and may be play an important role in mediating the thermal stress in A. japonicas.

The distribution of seaweed was examined in Jeonnam archipelago marine ranching area, the coastal of geumo-do, Yosu. Abundance and distribution of seaweed in dropping site of artificial reef were sampled at 6 station at October 2007. Seaweed community were investigated with quadrat method at line-transect by scientific SCUBA divers. as a result, 30 species in total, 4 green, 9 brown and 15 red seaweed were identified. Range of seaweed biomass in all sampling stations were about 1,600~4,000 g/m<sup>2</sup>. At intertidal zone, appearance of individuals and dominance rate were showed higher than below the subtidal line and at depth from 4m to 6m, individuals, dominance rate and biomass were represented low level. characteristic of water in marine ranching area is turbidity and also compensation depth is low. For that reason, individuals of seaweed community were smaller from depth of 4 meters. Therefore, when equip the artificial reef for preparation of seaweed beds need consider that equipment periods(early spring; before releasing marine seaweed spore) and suitable water depth(3~6m).

In juvenile size, the maximum clearance rate and ingestion rate were 15mm, which is the 70.8 mL/h, 4245.0×104cells/h, respectively. But the lowest clearance rate and ingestion rate were found at 5mm, which is the 4.1 mL/h, 246.0×104cells/h, respectively. The highest clearance rate and ingestion rate of AFDW seen at 7.5mm, with 3.3 mL/mg AFDW/h, 196.0×104cells/mg AFDW/h, respectively. According to the results of water temperature, clearance rate and ingestion rate were lowest at 10℃, showing the 0.3 mL/h, 20.3×104cells/h, respectively. The highest clearance rate and ingestion rate were 25℃, which is the 16.6 mL/h, 993.4×104cells/h, respectively. But in 30℃, clearance rate and ingestion rate decreased to the 12.9 mL/h, 772.6×104cells/h, respectively. With respect to the food density, the experiment groups with 30×104 cells/mL and 240×104 cells/mL showed the clearance rate of 21.1 mL/h and 2.3 mL/h, respectively. But the experimental group with 120×104 cells/mL or more showed decreased clearance rate. While, the maximum ingestion rate was at 60×104 cells/m, showing the 876.2×104cells/h.
