Mast cell heterogeneity occurs not only in different species but also with the same organ in the same species. Two distinct types of mast cells are recognized in the rat. Connective tissue mast cells(CTMCs) and mucosal mast cells(MMCs) of rat contain different proteoglycans, which can be distinguished using histochemical method. The present study was to performed to investigate the mast cell population and histochemical characteristics in the rat tongue. Tissues were taken from the three portions-anterior 1/3, middle 1/3 and posterior 1/,3 portions-of the rat tongue. For optimal preseuation of the MMCs, Carney's solution(pH 2.0) and 10% formalin with 4% lead acetate(pH 4.0) were used. The tissues were embedded in paraffin. The sections(6Um in thickness) were stained with toluidine blue, alcian blue(pH 0.5, 3.0)/safranin, astra blue(pH 0.3, 4.5)/safranin. In each portion of the tongue, mast cells were observed in the lamina propria and muscualar layer. The total number of mast cells in the anterior portion of the tongue was greater than that of the middle or posterior portion of the tongue(p<0.01). No significant difference in the total number of mast cells were found between the middle and the posterior portions(p>0.01). A significantly higher number of mast cells was seen in the deep muscular layer than in the superficial muscular layer of the tongue(p<0.01). The staining characteristics of mast cells of the tongue were influenced by the type and pH of fixatives as well as staining solution. These results indicate that mast cell ehterogenity occurs also in the rat tongue.
