Pseudomonas sp. EL-041 LY와 Alcaligenes sp. EL-041 LW에 의한 Polyvinyl Alcohol의 분해에 관한 연구

Twenty microorganisms capable of biodegradation of polyvinyl alcohol(PVA) were isolated from the waste water by the enrichment culture technique. The strain which had the highest biodegradability was designated as EL-041 L in PVA minimal medium. The strain were found out to be symbiont composed of two kinds of bacteria from the results of pure isolation in nutrient plate and effect of surface area of plate of PAV medium. These isolated strains which had two different kinds of bacteria were designated as EL-041 LY and EL-041 LW. The results experimented on the identification of these isolated strains EL-041 LY and EL-041LW, on the growth characteristics and degradation rate of PVAs, on the oxidation test on PVA compounds and metabolic products formed from PVA by these isolated strains, were as follows. 1.These isolated strains EL-041 LY and EL-041 LW were identified as Pseudomonas and Alcaligenes from the results of bacteriological identification tests. These isolated strains were named temproarily Pseudomonas sp. EL-041 LY and Alcaligenes sp. EL-041 LW for convenience, respectively. 2.The optimal condition of medium for growth and degradation was observed as follows:the initial pH of medium was pH 7.5 ammonium nitrate 0.2% was selected for the source of nitrogen, PVA 0.2% was selected for the carbon source. The degradation rate of PVA 22000 was about 52.1%. Growth on the effect of PVA polymerization grade was showed that degradation rate of lowest PVA polymerization grade, PVA15000(300), was about 68%. The comparision of mixed and pure culture was showed that the one was optical density 0.72 growth rate at 660nm, the other was 0.17 growth rete. 3.Oxidation test was showed the most oxygen uptake to the PVA 100000 substrate of low growth rate. When oxygen uptake was compared with between resting cells of the grown on PVA medium and that of on PVA medium using 0.5% polypeptone as nitrogen and carbon source, only PVA-grown cell could oxidize PVA. This fact coincides with the result of effect of substrate of culture on PVA degradation and suggests that PVA is oxidatively degraded by PVA-grown cells. 4.On the thin layer chromatograph, the metabolic products formed from PVA 22000 by symbiont strains, EL-041L, used as sole carbon and energy soure, were suspected one acidic compounds containing carboxyl group the other alkalic compounds containing ketone group, but both not containing amine group.
