The toluene-tolerant isolate BCNU 106 which is capable of growing in media containing more than 30% (v/v) toluene is gram negative rod. This strain showed oxidase negative, catalase positive. and arginene dihydrolase negative reaction, and didn't accumulate poly-β-hydroxy butyrate. Also this strain utillized L(+)-tartarate, but not L-tyrosine, tripropionin, tricaproin, DL-β-hydroxybutyrate. DL-lactate, erithritol, and quinate. The major 3-hydroxy fatty acids of BCNU 106 is C10:0 3-OH acid and Cl2:0 3-OH acid. The major ubiguinone of BCNU 106 is Q-9. Based on the above date, this toluene-tolerant strain is identified as Pseudomonas savastanoi. The optimal cultivation of strain BCNU 106 can be done in Modified Luria-Bertani media adjusted to pH 7.0 at 37℃ . When strain BCNU 106 was cultivated in the presence of 30% toluene. a 3.3kDa protein was induced. This protein seems to be a major toluene-tolerant protein.

Multinucleated cells resulted from mitosis defect have been noted in pathophysiological statesof the cells such as inflammation, senescence and cancer. Since oxidative stress has been known to correlate with these pathophysiological conditions, we tested the effect of H2O2 on the cell cycle progression and formation of multinucleated cells. H2O2 induced a significant delay in cell cycle progression in Chang liver cells. Interestingly, H2O2 actively induced hyper-amplification of centrosomes (n ≥ 3) and multipolar spindle formationduring mitosis and subsequently increased the generation ofmultinucleated cells. A significant increase of the phospho-ERK level was observed upon H2O2 treatment but PD98059, an MEK1/2 inhibitor, didnt reduce the frequency of cels with hyperamplified centrosomes. On the 2O2 or adriamycin increased intracellular ROS levels and multinu-cleated cels, which were significantly supres-sed by antioxidants, N-acetylcysteine and PDTC. Thus, our results suggest that oxidative stress can triger centrosome hyperamplification and multinucleated cell formation, which may pro-mote pathophysiological progresion.
Trichoderma sp. CNU 301가 생산하는 단백질 분해효소에 관한 연구

The mold which could grow at alkaline pH and thermophilic condition and produced an extracellular proteolytic enzyme was identified as Trichoderma sp.. Trichoderma sp. CNU 301 exhibited the maximum production of proteolytic enzyme at 50℃ in the medium composed of arabinose 1.5%, peptone 1.5% and FeSO₄0.2% (pH 9.0). The partial purified protease by precipitation with ammonium sulfate was most stable at pH 10 and at 60℃. The Trichoderma sp. CNU 301 produced an alkaline and thermostable protease.
