Objective : Heat shock protein family is related to protective mechanism of cells by environmental changes. This study was performed to evaluate the effect of cryopreservation on the heat shock protein 90 (Hsp90) expression in mouse ovarian tissue. Methods : Cryopreservation of mouse ovarian tissue was carried out by slow freezing method. The mRNA level of Hsp90 expression in both fresh and cryopreserved mouse ovarian tissue was analyzed by RT-PCR. The protein expression of Hsp90 was evaluated by Western blot analysis and immunohistochemistry. Results: The mRNA and protein of Hsp90 were expressed in both fresh and cryopreserved mouse ovarian tissue. The amount of Hsp90 mRNA was increased in cryopreserved ovarian tissue after 60 and 90 minutes after thawing and incubation. The amount of Hsp90 protein was increased in the cryopreserved ovarian tissue after 6 hours of the incubation in Western blot analysis. In immunohistochemical study, Hsp90 protein was localized in cytoplasm of oocytes and granulosa cells. Significant level of immunoreactive Hsp90 protein was detected in theca cells contrast to the weak expression in ovarian epithelial cells. Conclusion: This results showed the increase of Hsp90 expression in both mRNA and protein level in the cryopreserved mouse ovarian tissue. It can be suggested that Hsp90 may play a role in the protective or recovery mechanism against the cell damage during cryopreservaion.
빈각을 호소하는 만성 불면 환자에 대한 황련해독탕 약침을 중심으로 한 한방 복합치료 1례 증례보고

Objective : The purpose of this study is to investigate the effect of Hwangryunhaedok-tang pharmacopuncutre on frequent awakening in a 46-year-old female patient who has insomnia. Methods : Patient was treated with Hwangryunhaedok-tang pharmacopuncture on eleven acupoints including GV_{20}, right BL_{66}, ST_{44}, SI_{5}, ST_{41} and both GB_{20}, GB_{21} BL_{15}. Patient s quality of sleep was assessed by Korean version of Pittsburgh Sleep Quality Index(PSQI-K), Korean Sleep Scale(KSS), and Sleep Diary. The treatment was executed once per day between July 6-10, 2017. Results : After five treatment sessions, PSQI-K score changed from 12 to 17, mean value of KSS changed from 20 to 29 and mean value of sleep efficiency changed from 28 to 40. Conclusions : The result suggest that Hwangryunhaedok-tang pharmacopuncture can be a valuable option to relieve insomnia.
우하지냉증을 호소하는 급성 압박골절 환자에 대한 황련해독탕 약침을 중심으로 한 한방 복합치료 1례 증례보고

Objectives The purpose of this study is to investigate the effect of Hwangryunhaedok-tang pharmacopucture on cold hypersensitivity of right leg in a 80-year-old female patient. Methods Patient was treated with Hwangryunhaedok-tang pharmacopuncture on right ST35, GB34, GB39, BL56, BL57, SP9, SP6 and LR4. Treatment progress was assessed by coldness diary, coldness drawing, the skin temperature on ST36, SP6, LR3, KI1, Pittsburgh Sleep Quality Index(PSQI-K) and EQ-5D. The treatment was executed once per a day between August 17- September 9, 2022. Results The range and extent of patient’s subjective symptoms were decreased. The skin temperature on 4 acupoints was increased. The time takes to fall asleep was decreased. The EQ-5D changed from 0.423 to -0.056. Conclusions The result suggest Hwangryunhaedok-tang pharmacopuncture can be a valuable option to relieve acute idiopathic unilateral cold hypersensitivity.
