        시메티딘 및 제산제가 미노싸이클린의 약물동태에 미치는 영향

        Effects of aluminum magnesium hydroxide (A) and cimetidine (C) on the pharmacokinetics of minocycline (M) were investigated in female rats. Blood samples were collected at various time intervals until 36 hrs following oral dosing of drugs. Plasma minocycline concentrations were determined by HPLC. Control group (M), $T_1$ group (M+A), $T_2$ group (A+M after 2 hrs), $T_3$ group (M+A after 2 hrs), $T_4$ group (M+C) and $T_5$ group (C+M after 2 hrs) were divided to examine interaction of the drugs with minocycline. Plasma minocyline level-time curves were well described by two-compartment open model with first-order absorption in rats. Antacid treatment was associated with reduced of 71.0, 45.9, 35.7% in minocycline absorption rate $constant(K_{\alpha})$, maximum plasma $concentration(C_{max})$, and relative $bioavailability(F_{rel})$, respectively. Cimetidine treatment group exhibited no significant changes in plasma level-time curve when compared with control group and did not affect minocycline absorption as by any of these three parameters.

        Inhibitory activity of pesticide-preventing active carbon fiber mask on the penetrations in pesticide spray and its inhibitory effect on acute inhalation toxicity in rats were studied. Pesticides were sprayed into box (1.O x 1.2 x O.6 m) at a dose of 20ml/min of diazinon (DiatoneTM) suspension (340ppm) and 5g/min of BPMC powder (2.0%) for 4 hours, and then captured the penetrated pesticides into water via mask in box under 8l/sec suction. The pesticides were analysed by A.O.A.C. method and GC/MS. Pesticide-preventing active carbon fiber mask significantly inhibited the penetration of diazinon by 93.4% as compared with control, and cotton mask and pesticide-preventing mask did it by 74.2% and 83.7%,respectively. This result shows that pesticide-preventing active carbon fiber mask has the most prominent inhibitory effect in preventing the penetration of diazinone. In the penetration of BPMC, all of three masks, such as active carbon fiber mask, cotton mask and pesticide-preventing mask, have a similar inhibitory effect, which were 99.3%, 98.6% and 97.0%, respectively. The penetrated pesticides via pesticide-preventmg active carbon fiber mask did not exert acute inhalation toxicity.

        생약으로부터 In vitro DNA 결합활성의 검색

        One hundred and seventeen crude drugs were screened for DNA-binding activity in vitro. The DNA-methyl green assay is a useful biochemical screen for the detection or development of biologically active compounds which bind to DNA. This assay is based upon the fact that free methyl green undergoes rapid spontaneous molecular rearrangement to its colorless carbinol base so that the liberation of the dye from DNA by displacement can be followed spectrophotometrically as a decrease in absorbance at 65Onm. Seven methanolic extracts of crude drugs including Cinnamomi Cortex spissus, Coicis Semen, Coptidis Rhizoma, Perillae Semen, Plantaginis Semen, Polygalae Radix and Zanthoxyli Fructus showed less than 2,000mcg/ml as their IC50 values. The active principles of Plantaginis Semen and Zanthoxyli Fructus were transferred into organic solvents, which showed the IC50 values with 588 and 574mcg/ml, respectively. These fractions have been selected for isolation of biologically active constituents.
