마이코플라스마에 감염된 대추나무, 뽕나무 및 일일초의 조직절편을 형광염색소인 DAPI(4'-6-diamidino-2-phenylindole$\cdot$2HCl), aniline blue 그리고 quinacrine(quinacrine mustard dihydrochloride)으로 염색하여 형광현미경하에서 관찰함으로서 이들 형광염색소의 마이코프라스마 감염진단에의 효용가치를 비교 조사하였다. 이병식물의 줄기절편의 절부에서 특이형광반응이 나타났으나 건전식물의 조직절편에서는 특이형광반응이 관찰되지 않음으로써, DAPI, anilinc blue 및 quinacrine 등의 형광염색소에 의한 조직염색법은 대추나무, 뽕나무 및 일일초의 마이코플라스마 감염을 식속 정확하게 진단하는 데 매우 유용한 방법임을 보여주었다. 한펀, 이들 형광염색소 중에서는 DAPI가 가장 효과적이었다. Attempts were made to evaluate the efficacy of three fluorochromes, i.e., DAPI (4'-6-diamidino-2-phenylin-dole-2HCl), aniline blue and quinacrine(quinacrine mustard dihydrochloride) for the detection of mycoplasma infections in jujube (Zizyphus jujuba), mulberry (Mows alba) trees and periwinckle (Catharanthus roseus) plant by fluorescence microscopy. Stem sections from these plants infected with mycoplasma-like organisms (MLO) produced distinct fluorescence in the phloem when stained with DAPI, aniline blue or quinacrine, while fluorescence was absent in the healthy plants. The use of these fluorochromes provided simple and efficient techniques for the diagnosis of MLO infections. IOf the three fluorochromes tested, DAPI was found to be most efficient.

Selective breeding and hybridization were attempted to improve shiitake(Lentinus edodes) strains for bed-log cultivation, and the results obtained are as follows. 1. Thirty five strains collected in Korea and abroad were cultivated for one generation(five years) and. fruit body yields were estimated by the amounts produced from 1㎥ of oak tree(Quercus mongolicai logs. Thus, three high-temperature type strains including FRI 177 showing productivity of 139㎏/㎥log, two mid-temperature type strains including FRI 188 of 120㎏/㎥log, and seven low-temperature type strains including FRI 187 of 96㎏/㎥log were selected as superior strains. 2. Twelve hybrid strains confirmed as independent strains through indoor-test with hybrid strains made by Di-mon mating method were cultivated for five years(one generation). Among them, FRI 182 and FRI 181 were turned out to be superior strains. FRI 182 of high-temperature type showed the yield of 108㎏ per 1㎥logs and FRI 184 of low-temperature type produced 103㎏/㎥log. 3. There were no significant differences in productivity among high-, mid- and low-temperature types, but some differences appeared among strain groups. The correlation was not shown in productivities between bed-log and sawdust cultivations.

Post-harvest trunk injection of oxytetracycline-HCl (OTC) was attempted for the control of jujube witches` -broom. Diseased jujube(Zizyphus jujzuba) trees with 9 to 16㎝ trunk diameters were treated with OTC ranged from 2g to 6g according to the size of trunk diameter. OTC dissolved in 0.51 or 1.01 of water was transfused into diseased trees by gravity flow injection during September and October. All these OTC concentrations prevented symptom development for two growing seasons and restored previously severely diseased trees to normal conditions. There was no differences in control effect between 0.51 and 1.01 OTC solutions. With these high OTC concentrations, no phytotoxicity was observed in the new leaves of the following year. Mycoplasma -like organism (MLO)-specific fluorescence was absent in the phloem of recovered tissue when examined by fluorescence microscopy using DAPI (4`-6-diamidino-2-phenylindole.2HCl) staining, indicating the disappearance of MLO by the action of OTC.
우리나라 표고의 생장 , 부후특성 (腐朽特性) 및 톱밥배양에 관한 연구

Studies were made to find out the mycelial growth and rotting characteristics of 91 Lentinus edodes isolates collected or hybridized in Korea and abroad, and their adaptability to bag-culture. The results are as follows. 1. There were little differences in mycelial growth at PDA and oak (Quercus mongolica) sawdust-based medium, and rotting among high-, mid- and low temperature types of L. edodes isolates. But. the fruit yield of high-temperature type was much higher than those of the other two types. The fruit yields of the isolates in the same strain group were apparently different. 2. From the sawdust-based culture of 91 isolates, FRI 221, FRI 208 and FRI 169 were selected as excellent strains in yield and Quality, showing fruiting of 157g, 152g and 119g per 1㎏-medium, respectively. 3. Attempts to culture in various media with different substrates resulted in almost same fruit yields, and the yield was proportional to medium weights as 2.5㎏-medium showed 2.5 times more yield than 1㎏ -medium. Fruit yields were different according to incubation periods and the period of little more than 100days was best. 4. When the correlation analyses among mycelial growth, rotting ability, yield and fruit-shape normality were made with the 91 isolates, mycelial growth at sawdust-based medium showed highly positive correlation with that at PDA, while fruit yield was negatively correlated with mycelial growth in PDA and sawdust-based medium.

New Shiitake (Lentinula edodes) strain “Chunbaegko” was bred by Di-mon method. Bed-log cultivation of “Chunbaegko” was performed. Fruit-body production of “Chunbaegko” was most at spring and autumn. Optimal temperature of fruit-body formation was 14~22oC Baegwhako, the best quality fruit-body, is produced during spring. The diameter of pileus is ca. 52 mm. The total amount of fruit-body production during 4 years(one generation) was 140 kg/m3 log.
