      • 억간산이 PTU (6-n-propyl-2-thiouracil)로 유발된 Rat 갑상샘 기능저하증에 미치는 영향

        PURPOSE : The object of this study was to evaluate the effect of Ukgansan (UKS), aqueous extracts of mixed herbal formula consisted of nine herbs - Angelicae Gigantis Radix, Atractylodes Macrocephalae Rhizoma, Hoelen, Rhynchophylla Ramulusi, Cnidii Rhizoma, Bupleuri Radix, Glycyrrhizae Radix, Citri Pericarpium and Pinelliae Rhizoma on the PTU-induced rat hypothyroidism METHODS : 6 groups, each of 8 rats per group were used in the present study as follows. Aqueous extracts of UKS were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 200, 100 and 50 ㎎/㎏ (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 ㎎/㎏ for 28 days. The changes on the body weight, thyroid gland, liver, epididymis and testis weights, serum thyroid hormone (TSH, T3 and T4) levels, serum male sex hormone (testosterone, DHT and FSH) levels, serum lipid profiles (total cholesterol, LDL, HDL and triglyceride), liver and testis antioxidant defense system (lipid peroxidation, H2O2, SOD and CAT), serum AST and ALT levels were observed with histopathology of thyroid gland, liver, epididymis and testis. Results were compared with LT4 0.5 ㎎/㎏ treated rats Experimental groups : 6 groups (8 rats per groups were used) 1. Intact control : saline treated instead of PTU, and distilled water orally administered group 2. PTU control : PTU subcutaneously treated and distilled water orally administered group 3. LT4 : PTU subcutaneously treated and LT4 0.5 ㎎/㎏ intraperitoneally treated group 4. UKS200 : PTU subcutaneously treated and UKS extracts 200 ㎎/㎏ orally administered group 5. UKS100 : PTU subcutaneously treated and UKS extracts 100 ㎎/㎏ orally administered group 6. UKS50 : PTU subcutaneously treated and UKS extracts 50 ㎎/㎏ orally administered group. RESULTS : As results of PTU treatment, marked decreases of body weights, increases of thyroid weights, liver, epididymis and testis weight decreases, serum T3, and T4 level decreases with increases of serum TSH levels, serum testosterone and DHT level decreases with increases of serum FSH levels, serum HDL increases and triglyceride content decreases, decreases of liver and testis antioxidant defense system, increases of serum AST levels were observed. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contents and sizes were demonstrated with decreases of hepatocyte numbers due to hypertrophy of hepatocytes and the increases of oligospermatic epididymal tubules at histopathological inspections. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of UKS extracts, and UKS extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. CONCLUSION : The results obtained in this study suggest that UKS extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism related liver and reproductive organ damages mediated by the modulatory effects on the antioxidant defense system.

        황금, 황련, 황백 및 어성초 추출물을 함유한 피부 개선 화장품 조성물

        In our modern society, high level of economic growth and extension of human lifespan have led people to make efforts to maintain their health and be concerned about beautiful and healthy skin. Although various types of cosmetics are being developed and released to satisfy these customers needs, use of essential additives such as emulsifiers and preservatives seems inevitable, considering that the cosmetics are produced through physico-chemical processing of various chemical components and fragrances. Since dangers of these additives have begun to be proved, the maximum permissible amount of them is specified. Nevertheless, the general standards for the hazardous components can still lead expression of symptoms or aggravation of the diseases in sensitive skin or skin diseases. Therefore, it is important to develop the products with safety and effectiveness for continuous use and the symptom relief. Natural herbs used in oriental medicine have a high value as the cosmetic materials, as their excellent benefits such as antiseptic or antimicrobial properties have been reported in several industrial fields including cosmetics and foods, as well as their positive image. In this study, we have examined antioxidative effect of water and ethanol extracts of four natural herbs, Scutellariae radix, Coptidis rhizoma, Phellodendron amurense, and Houttuynia cordata. They all showed a significant antioxidative effect in a dose-dependent manner, and it suggested their potential as the functional ingredients that protect the skin cells from oxidative stress. We expect that further investigation on their anti-inflammatory and anti-microbial effect will provide a strong possibility that they could be used as the active ingredients of functional herbal cosmetics to improve atopic dermatitis.

        황련의 1,3-Butylene glycol과 혼합 추출물의 항주름 및 항산화 활성

        We evaluated the cytotoxicities and anti-wrinkle, antioxidant and whitening capacities of 1,3-butylene glycol and mixed solvent from Coptidis Rhizoma for potential use in cosmetic materials. The collagenase inhibition rates (ie, anti-wrinkle effects) were 81.95% (BGE; 1,3-butylene glycol extract from Coptidis Rhizoma) and 81.28% (WBG; water in 1,3-butylene glycol extract from Coptidis Rhizoma) at 3.0%. The elastase inhibition potential of BGE was higher than that of ascorbic acid (81.96% vs 68.06%, respectively). The tyrosinase inhibition rate of BGE was 11.20%. The DPPH radical scavenging potentials of BGE and EtBG (ethanol in 1,3-butylene glycol extract from Coptidis Rhizoma) were 86.04% and 85.37%, respectively. As for ABTs [2,2′-azinobis-(3-ethylbenzothiazoline-6- sulfonic acid)] radical scavenging and SOD-like activities, WBG showed the highest values-98.98% and 57.03% at a concentration of 3.0%, respectively. Xanthine oxidase inhibitory activities of BGE and EtBG were 94.64% and 94.18%, respectively. Measuring cytotoxicity using the MTT assay, RAW264.7 macrophage viability was increased at 25 ug/mL, however, as the concentration of extracts increased, the viability of RAW264.7 macrophages decreased. HaCaT cell viabilities decreased following treatment with WBG and EtBG at 25 ug/mL, but HaCaT cell viabilities increased at concentrations over 50 ug/mL. Our results revealed that the 1,3-BG and water and ethanol mixed extracts displayed anti-wrinkle and antioxidant activities. These extracts could be used as a functional anti-aging cosmetic material.
