재조합 플라스미드 pCU103 의 수계내 안정성에 미치는 pH 의 영향

The recombinant plasmic pCU103 constructed by cloning the pcbCD genes in pBluscript SK(+) vector was examined for its stability in different water microcosms. In this study, particularly, the effect of different pH of the water on stability of the pCU103 was studied in terms of its structural integrity, concentration, and transformation activity. Different pH value did not affect stability of the plasmid in the waters, such as sterile distilled water(SDW), 0.2㎛ of filtered river water(FW), and autoclaved river water(AW). But the pCU103 plasmid sustained its stability upto 25 days in SDW and AW at any pH value, whereas the plasmid in FW was degraded out after 3 days without regards to pH value, and lost its transformation activity.
