        λ 형 Bence Jones 단백질의 N 말단주변(末端周邊)의 아미노산 배열순서에 관한 연구

        Two peptides (Im pr-M, Im ch-M) derived from Im λ-type of Bence Jones Protein and one peptide (Ikch-M) from Ik were separated and purified using the Dowex 50×2 column (1×20 ㎝) and Dowex 1×2(0.9×50 ㎝). The buffer solution was composed of 1% pyridine and IM formic acid in Dowex 1×2 column. The blocked N-terminal was examined with ninhydrin reaction before and after alkaline hydrolysis, which was fractionated by Dowex 1×2 column. Pyrro-glutamic acid in N-terminal residue was identified by comparing with the authentic pyrro-glutamic acid through a high voltage electrophoresis (pH 3.5, 3000 V.) after the peptide Im pr-M (PCA. Ser) was cleavaged at the position of serine with cone. (12 N) HCl and the pyrro-glutamic acid was converted to glutamic acid by treating it with N-NaOH for 116 hours at 27℃. The substractive method was applied to find out the sequence of peptides and carboxypeptidase A was employed to release C-terminal residue from the peptide. In present study PCA. Ser in Im Pr-M was isolated from the pronase digested λ-type Bence Jones protein. The yield of the Im Pr-M was 79.6 percent of its theoretical value, based on the molecular weight of Bence Jones Protein. Im ch-M (PCA. Ser Val. Leu) was isolated from the chymotrypsin digested λ-type Bence Jones Protein. The yield of the Im ch-M was 72.2 percent. based on the molecular weight of Bence Jones Protein. Ik ch-M (PCA. Ser. Ala. Leu) was isolated from the chymotrypsin digested λ-type Bence Jones Protein and its yield was 42% based on the molecular weight of Bence Jones Protein.

        The advance communication is the major difference between the human being and animals. The advancement of the human being is in proportion to the advancement of media. One of the most important concept in communication and media has concerned with Symbolololgy. Media and communication had been developed in various ways, therefore symbology also approaches in various ways, such as TV drama, film, print and/or new media. The purpose of this study is analyzing interface design into Symbolololgy for the effective communication for new media. Interface is the foremost medium between the sender and the receiver. That's why Interface design is so important to the information provider, such as internet, CD-ROM and interactive TV, differnt between the other media. This study is based on effective communication for information provider through Symbological analysis multimedia project(CD-ROM title) into interface design in the New media Era.

      • 밀가루 단백질의 화학적 구성

        밀가루 단백질인 Gluten은 밀가루 반죽의 부피를 크게하는 중요한 역할을 하며 이 Gluten 단백질을 구성한 Gliadin의 분자량은 40,000이며 Glutenin은 2$\~$3백만의 고분자화합물로 Gliadin과 Glutenin은 서로 구조상으로는 다르나 Gliadin이 Glutenin 구성 polypeptide로 S-S 결합에 중합체로 구성하며 이들은 20,000$\~$25,000정도의 공통된 polypeptide로부터 이루어진 것으로 추리하고 있다. Gluten 단백질에 S-S 함량이 7.4$\~$10mole/$10^5$g protein이며 -SH 함량은 0$\~$0.3mole/$10^5$g protein이다. Gluten 표면에 -SH가 적으나 S-S, 및 -SH의 상호교환이 일어나 망상 형성하여 밀가루 반죽내에서 발생하는 $CO_2$의 가스를 들어쌓여 빵의 부피를 크게 한 것이다.

        고구마 β - Amylase 정제에 관한 연구

        Sweet potato β-amylase was purified and crystallized by the suggested acetone fractionation method at room temperature and some physicochemical properties of the enzyme were investigated. The enzyme was purified and crystallized by the suggested method in 3 hours. The purified enzyme showed 26.3 folds of purification, 4,395 unit/㎎ of specific activity and 14.3% of yield, respectively, compared to the crude extract. The crystallized enzyme was found to have a rectangular-sheet form in pure water. The purified enzyme was identified as homogenous by disc PAGE, UV difference spectrum, HPLC analysis and maltase activity assay. The molecular weight of the enzyme subunit was found to be 57,000 by SDS PAGE. The amino acid analysis indicated the enzyme is composed of 478 amino acid residues per 57,000 daltons of the enzyme subunit.

        Bence Jones 단백질의 정제 및 N - 말서검출(末瑞檢出)

        Human Bence Jones Protein could be purified by DEAF-Sephadex A-50 column (2×37㎝) with 0.02M phosphate Buffer (pH 8.0) and gradient increasing with NaCl concentration as in Fig. 2-4. Sample As (K-type Bence Jones Protein) had two component, F-I was major component and its dried weight was 350㎎. of starting material of 500㎎. Other Sample Im and Ik (λ-type Bence Jones Protein) was purified by DEAE-Sephadex A-50 with 0.02M phosphate Buffer(pH 8.0)too. F-I (major component) of Im and F-I of Ik were 242㎎ and 146㎎. its dried weight respectively. K-type of Bence Jones Protein's(As, Ko, Ta.) N-terminal amino acid residue was determined by method of DNP,. K-type of Bence Jones Protein's amino acid residue were either glutamic acid or aspartic acid. Sample Ta was confirmed as glutamic acid its N-Terminal. As and Ko were aspartic acid. Each yellowish spot (DNP-amino acids) were extracted with 4㎖. of pH 8.05% NaHCO₃ solution and calculated its recovery by O.D. (360mμ)using the ε=18.1×10³-DNP Asp. ε=17.4×10³ DNP Gla. considering 50% lose during; the acid (6N-HCI) hydrolysis. Recovery of ko and As were 54.3% and 65% of its starting materials (DNP-Protein). Sample Ta's recovery was 85% of its DNP-protein. λ-type of Bence Jones Protein was rot investigated its N-terminal amino acid residue by DNP-method, probably it was blocked its N-terminal residue with glutamic acid.

        효과적 ODA를 위한 교육 ICT 활용 방안에 대한 연구

        한국은 단기간에 성공적인 경제 성장을 이룩하여 원조 수혜국에서 지원국으로 발전하여 국제적인 모범사례가 되었다. 그러나 세계 무역 10위권인 한국의 대외원조 규모는 다른 OECD 회원국에 비하여 최하위 수준으로 국제사회로부터 인색하다는 비판적 평가를 받고 있다. 이로 인하여 정부에서는 ODA 지원 사업을 확대, 강화하고 있다. ODA 지원에 있어서 경제적이며 효과적인 방안으로는 보유하고 있는 시스템과 노하우를 활용하는 것이다. 한국이 가장 앞서 있는 교육 ICT 시스템과 관련 노하우를 활용한 ODA 사업추진은 수혜국의 미래에 기여한다는 대의적 명분으로 국가 이미지를 극대화 시킬 수 있다. 이와 함께 AV, OA, IT 관련 하드웨어는 물론 다양한 교육, 문화 및 게임 관련 소프트웨어 등 한국의 IT 및 CT 기업들에게 해외 진출의 기회를 제공하여 산업적 실리를 동시에 확보 할 수 있는 정치, 외교, 산업, 경제 및 문화를 함께 어우르는 컨버전스(convergence) 개념이다. 이러한 교육ICT를 통한 ODA 사업 추진 방안을 연구하여 로드맵(Roadmap)과 디자인 시안을 제시하고자 한다. South Korea had accomplished successful economic growth in short period of time comparatively and became international role model of development from aid recipient country to aid donor country. However there comes critical evaluation from international society that the aid is stingy at the level of the bottom compared to other OECD member nations. With this context in mind, the government is reinforcing ODA supporting businesses. Economical and reasonable supporting ways are needed in backing and for these ways; it would be idealistic to exploit our advanced IT experience and knowhow. Especially, it could secure the effect to improve national identity by establishing justification of noble cause that they would bring contribution to not only donor countries but also progress in humankind if we propel aid service based on ICT in education. Also, it could obtain nation industrial profit by offering chance to enter the global market of Korean IT and CT companies as well as various education and culture contents and programs. This paper tries to suggest the roadmap and draft Design that there are some plans to enhance national image and brand as convergence concept for these enterprise to push forward.
