        Lipid Production by a CO2-Tolerant Green Microalga, Chlorella sp.MRA-1

        Since CO2 concentrations in industrial flue gases are usually 10%-20%, one of the prerequisites for efficient CO2 removal by algae is the level of tolerance of microalgal species to exposure to high concentrations of CO2. A newly isolated microalgal strain, Chlorella sp. MRA-1, could retain growth with high concentrations of CO2 up to 15%. The highest lipid productivity for Chlorella sp. MRA-1 was 0.118 g/l/day with a 5% CO2 concentration. Octadecenoic acid and hexadecanoic acid, the main components of biodiesel, accounted for 70% of the total fatty acids. A lipid content of 52% of dry cell weight was achieved with limited amounts of nitrogen. Chlorella sp. MRA-1 seems to be an ideal candidate for biodiesel production when cultured with high concentrations of CO2.

        Research on profile inversion controlling methods of multi-slug polymer alternative injection

        The development practice of polymer flooding shows that the injection profile easily inverses during the process of single slug polymer flooding. The low effective or ineffective circulation of abundant injecting water in high permeability layer, low producing degree of low permeability layer and large polymer volume have influenced the sweep efficiency and oil recovery of polymer flooding. In view of these questions, the alternative injection method of various viscosity polymer slugs is studied. This paper explains the theory of enhanced oil recovery of multi-slug polymer alternative injection. The experiment results show that the alternative injection of high and low viscosity polymer slugs has a better effect than single slug polymer flooding. The mobility differences between high-low permeability layers can be effectively reduced. For a multi-layer heterogeneous reservoir, there is an optimal alternative cycle. Reasonable slug size can not only inhibit dominant channel forming in high permeability layer but also can effectively produce low permeability layer and control profile inversion to a certain extent. This provides a technical support for studying the controlling methods of profile inversion.

        Silencing of Fanconi Anemia Complementation Group F Exhibits Potent Chemosensitization of Mitomycin C Activity in Breast Cancer Cells

        Purpose: Fanconi anemia complementation group F (FANCF) is a key factor to maintaining the function of Fanconi anaemia/BRCA (FA/BRCA) pathway, a DNA-damage response pathway. However,the functional role of FANCF in breast cancer has not been elucidated. In the present study, we evaluated the chemosensitization effect of FANCF in breast cancer cells. Methods: We performed specific knockdown of the endogenous FANCF in breast cancer cells by transfecting the cells with an FANCF short hairpin RNA (shRNA) vector. Cell viability was measured with a Cell Counting Kit-8, and DNA damage was assessed with the alkaline comet assay. The apoptosis, cell cycle, and drug accumulation were measured by flow cytometric analysis. Protein expression levels were determined by Western blot analysis, using specific antibodies. Results: The analyses of two breast cancer cell lines (MCF-7 and MDA-MB-435S) demonstrated that the FANCF shRNA could effectively block the FA/BRCA pathway through the inhibition of Fanconi anemia complementation group D2ubiquitination. Moreover, FANCF silencing potentiated the sensitivity of cells to mitomycin C (MMC), where combined FANCF shRNA/MMC treatment inhibited cell proliferation, induced Sphase arrest, apoptosis, and DNA fragmentation, and reduced the mitochondrial membrane potential, compared with MMC treatment alone. Conclusion: Taken together, this study demonstrates that the inhibition of FANCF by its shRNA leads to a synergistic enhancement of MMC cytotoxicity in breast cancer cells. These results suggest that the inhibition of the FA/BRCA pathway is a useful adjunct to cytotoxic chemotherapy for the treatment of breast cancer.

        HPLC Determination and Steady-State Bioavailability Study of Levodropropizine Sustained-release Tablets in Dogs

        A simple HPLC method using UV detection was developed and validated for the determination of levodropropizine (LDP) In dog plasma. The sample was prepared for injection using a liquid-liquid extraction method with 1-phenypiperazine as the internal standard. The mobile phase was methanol - diethylamine solution (0.05 M) (20:80, v/v, pH adjusted to 3.0 with $H_3PO_4$) with a detection wavelength of 240 nm. The limit of quantitation (LOQ) of LDP in a biological matrix was determined to be 25.25 ng/mL. The calibration curve was linear across the concentration range of 25.25 to 2020 ng/mL. The intra-day and inter-day precision values (CV%) were within 7% and accuracy (R.E. %) was within 6% of the nominal values for medium (252.5 ng/mL) and high (2020 ng/mL) LDP concentrations. For the LDP concentration at the LOQ, the intra-day and inter-day precision and accuracy were within 20% and 10%, respectively. The average absolute recovery for LDP was 70.28%. This method was successfully used to analyze plasma samples in a steady-state bioavailability study of a newly developed sustained-release LDP tablets (SR) using immediate-release tablets (IR) as the reference. The relative bioavailability of the SR was determined to be $106.3\;{\pm}\;12.8%$ (n=6). The $C_{max}$ of the SR was significantly lower (p<0.05), and the $t_{max}$ was significantly longer than that of the IR (p<0.05). The results of ANOVA and two one-sided tests indicated that the SR exhibited acceptable sustained release properties and was bioequivalent to the IR.
