      • Expression of TS, RRM1, ERCC1, TUBB3 and STMN1 Genes in Tissues of Non-small Cell Lung Cancer and its Significance in Guiding Postoperative Adjuvant Chemotherapy

        Background: To explore the expression of TS, RRM, ERCC1, TUBB3 and STMN1 genes in the tissues of patients with non-small cell lung cancer (NSCLC) and its significance in guiding the postoperative adjuvant chemotherapy. Materials and Methods: Real time polymerase chain reaction (PCR) was applied to detect the expression of TS, RRM, ERCC1, TUBB3 and STMN1 genes in the tissues of NSCLC patients so as to analyze the relationship between the expression of each gene and the clinical characteristics and to guide the postoperative individualized chemotherapy according to the detection results of NSCLC patients. Results: Expression of TS gene was evidently higher in patients with adenocarcinoma than those with non-adenocarcinoma (P=0.013) and so was the expression of ERCC1 (P=0.003). The expression of TUBB3 gene was obviously higher in NSCLC patients in phases I/II and IV than those in phase III ($P_1=0.021$; $P_2=0.004$), and it was also markedly higher in patients without lymph node metastasis than those with (P=0.008). The expression of STMN1 gene was apparently higher in patients in phase I/II than those in phase IV (P=0.002). There was no significant difference between the rest gene expression and the clinical characteristics of NSCLC patients (P>0.05). Additionally, the diseasefree survival (DFS) was significantly longer in patients receiving gene detections than those without (P=0.021). Conclusions: The selection of chemotherapeutic protocols based singly on patients' clinical characteristics has certain blindness. However, the detection of tumor-susceptible genes can guide the postoperative adjuvant chemotherapy and prolong the DFS of NSCLC patients.

        Variational B-rep model analysis for direct modeling using geometric perturbation

        The very recent CAD paradigm of direct modeling gives rise to the need of processing 3D geometric con-straint systems defined on boundary representation (B-rep) models. The major issue of processing such variational B-rep models (in the STEP format) is that free motions of a well-constrained model involve more than just rigid-body motions. The fundamental difficulty lies in having a systematic description of what pattern these free motions follow. This paper proposes a geometric perturbation method to study these free motions. This method is a generalization of the witness method, allowing it to directly deal with variational B-rep models represented by the STEP format. This generalization is essentially achieved by using a direct, geometric representation of the free motions, and then expressing the free motions in terms of composites of several basis motions. To demonstrate the effectiveness of the proposed method, a series of comparisons and case studies are presented.

        Robotic Path Planning Based on Episodic-cognitive Map

        Inspired by mammal’s spatial awareness and navigation capabilities, a new episodic-cognitive map buildingand path planning method was proposed, used for navigation tasks of mobile robot under the unstructured environment. Combined with characteristic of cognitive map and simulated the formation mechanism of episodicmemory in the hippocampus, a novel episodic-cognitive map encapsulated the information of scene perception,state neuron and pose perception was built, realized the real-time, incremental accumulative and updating cognitionof the robot to the environment. Based on the episodic-cognitive map, using the minimum distance between events,an algorithm of the event sequence planning was put forward for preferred trajectory choosing. Experimental resultsshowed that the proposed algorithms realized the mobile robot choose the preferred planning path, and using theSIFT-based visual navigation method, the mobile robot can reach the target very well. The method in this paperextended the application of biological cognition theory in the field of robotic planning.

        The Uneven Distribution of Mating Type Genes in Natural and Cultivated Truffle Orchards Contributes to the Fructification of Tuber indicum

        The aim of this study was to investigate the pattern of distribution of mating type (MAT) genes of Tuber indicum in ectomycorhizosphere soils from natural T. indicum-producing areas and cultivated truffle orchards and ascocarp samples from different regions. Quantitative real-time PCR and multiplex PCR were used to weight the copy numbers of MAT1-1-1 and MAT1-2-1 in natural truffle soils and cultivated orchard soils. The effect of limestone on the pattern of truffle MAT genes and the correlation between soil properties and the proportion of MAT genes were also assessed. These results indicated that an uneven and nonrandom distribution of MAT genes was common in truffle-producing areas, cultivated truffle orchards, and ascocarps gleba. The competition between the two mating type genes and the expansion of unbalanced distribution was found to be closely related to truffle fructification. Limestone treatments failed to alter the proportion of the two mating type genes in the soil. The content of available phosphorus in soil was significantly correlated with the value of MAT1-1-1/MAT1-2-1 in cultivated and natural ectomycorhizosphere soils. The application of real-time quantitative PCR can provide reference for monitoring the dynamic changes of mating type genes in soil. This study investigates the distributional pattern of T. indicum MAT genes in the ectomycorhizosphere soil and ascocarp gleba from different regions, which may provide a foundation for the cultivation of T. indicum.

        Research on the Relation between Transformer Oil Flow Electrification and Electrostatic Current

        In order to study and obtain the mathematical relation between the electrification degree of transformer oil flow and the electrostatic current, a small amount of data about the electrification degree of oil flow and the corresponding electrostatic current is studied by linear regression method and grey model method. The results show that the linear correlation between the electrification degree and the electrostatic current was not good, and the relation between the electrification degree of oil flow and electrostatic current (i) could be expressed as ρ<SUP>(0)</SUP> =0.2049 i<SUP>(0)</SUP> +169.4419 according to grey model GM (0, 2) when the electrification degree of oil flow is represented by the charge number generated from transformer oil per unit volume, namely the charge density (ρ).

        Dihydroartemisinin inhibits HepG2.2.15 proliferation by inducing cellular senescence and autophagy

        Dihydroartemisinin (DHA) has been reported to possess anti-cancer activity against many cancers. However, the pharmacologic effect of DHA on HBV-positive hepatocellular carcinoma (HCC) remains unknown. Thus, the objective of the present study was to determine whether DHA could inhibit the proliferation of HepG2.2.15 cells and uncover the underlying mechanisms involved in the effect of DHA on HepG2.2.15 cells. We found that DHA effectively inhibited HepG2.2.15 HCC cell proliferation both in vivo and in vitro. DHA also reduced the migration and tumorigenicity capacity of HepG2.2.15 cells. Regarding the underlying mechanisms, results showed that DHA induced cellular senescence by up-regulating expression levels of proteins such as p-ATM, p-ATR, γ-H<sub>2</sub>AX, P53, and P21 involved in DNA damage response. DHA also induced autophagy (green LC3 puncta gathered together and LC3II/LC3I ratio increased through AKT-mTOR pathway suppression). Results also revealed that DHA-induced autophagy was not linked to senescence or cell death. TPP1 (telomere shelterin) overexpression could not rescue DHA-induced anticancer activity (cell proliferation). Moreover, DHA down-regulated TPP1 expression. Gene knockdown of TPP1 caused similar phenotypes and mechanisms as DHA induced phenotypes and mechanisms in HepG2.2.15 cells. These results demonstrate that DHA might inhibit HepG2.2.15 cells proliferation through inducing cellular senescence and autophagy. [BMB Reports 2019; 52(8): 520-525]

        Influence of Porosity on Mechanical Behavior of Porous Cu Fabricated via De‑Alloying of Cu–Fe Alloy

        We report on a study of the mechanical behavior of porous Cu containing micron-sized pores and fabricated by de-alloyingof a Cu–Fe precursor alloy. Our results show that the minimum volume fraction of pores that can be obtained by using anapproach that involves de-alloying of a Cu–Fe precursor alloy is approximately 40 vol%. Moreover, the average pore sizeformed by de-alloying Cu–Fe of varying compositions is in the range of 1.5–4.0 μm. Our mechanical behavior results revealthat the yield stress increases from 3.9 to 58.6 MPa as the volume fraction of porosity decreases from 78.9% to 39.3%. Moreover, our data shows that the influence of porosity on the relative yield stress and relative Young’s modulus conformsto the scaling equations of Gibson and Ashby as formulated for open-cell porous metals. The pore cell characteristics anddeformation modes of porous Cu produced by de-alloying Cu–Fe alloys were discussed in the context of the observed fluctuationsin the value of the constants C and n in the Gibson-Ashby scaling equation. The evolution of microstructure duringcompressive deformation of porous Cu was studied and the results reveal an increase in the fraction of low-angle grainboundaries, an increase in the number of twins and a decrease in the average grain size with increasing strain from 0% to 70%.

      • Overexpression and Clinicopathological Contribution of DcR3 in Bladder Urothelial Carcinoma Tissues

        Background: To explore the expression of DcR3 protein and its clinicopathological significance in bladder urothelial carcinomas (BUC). Materials and Methods: Immunohistochemistry was performed to detect the expression of DcR3, caspase-3, Bcl-2, VEGF, Ki-67, PCNA and P53 in 166 BUC and 56 normal bladder tissues. Western blotting was used to detect the expression of DcR3 in the supernatants of cultured BUC cells. Results: Overexpression of DcR3 was found in BUC tissues and cell lines, with significant elevation as compared to normal bladder tissues (p<0.0001). Higher DcR3 expression was related to the status of invasion, lymph node metastasis and recurrence. Furthermore, DcR3 expression was negatively correlated with caspase-3 and positively associated with Bcl-2, VEGF, Ki-67 labeling index (LI), PCNA LI and P53 (all p<0.0001), respectively. Conclusions: DcR3 may play a crucial role as an oncogene in tumorigenesis, deterioration and progress of BUC via influencing related pathways of apoptosis, proliferation and angiogenesis. The detection of DcR3 protein in the formalinfixed and paraffin-embedded samples could assist to predict in prognosis of BUC patients.

        Study on the Thermal Transient Response of TSV Considering the Effect of Electronic-Thermal Coupling

        The transmission performance of TSV considering the effect of electronic-thermal coupling is an new challenge in three dimension integrated circuit. This paper presents the thermal equivalent circuit (TEC) model of the TSV, and discussed the thermal equivalent parameters for TSV. Si layer is equivalent to transmission line according to its thermal characteristic. Thermal transient response (TTR) of TSV considering electronic-thermal coupling effects are proposed, iteration flow electronic-thermal coupling for TSV is analyzed. Furthermore, the influences of TTR are investigated with the non-coupling and considering coupling for TSV. Finally, the relationship among temperature, thickness of $SiO_2$, radius of via and frequency of excitation source are addressed, which are verified by the simulation.

        Comparative Study of DC Breakdown and Space Charge Characteristics of Insulation Paper Impregnated with Natural Ester and Mineral Oil

        Natural ester is a suitable substitute for mineral oil and has been widely used in AC transformer in many countries. In order to further application of natural ester in direct current (DC) equipment, it is needed to investigate its long term insulation property under DC condition. In this paper, a thermal ageing experiment was conducted for both mineral oil-paper and natural ester-paper insulation. The DC breakdown and space charge characteristics of insulation paper impregnated with natural ester and mineral oil was compared. Results show that the resistivity of the paper immersed in natural ester and mineral oil both increase as the ageing goes on. While insulation paper impregnated with natural ester has higher resistivity and DC breakdown voltage than the paper impregnated with mineral oil. The DC breakdown voltage for the oil impregnated insulation paper being DC pre-stressing is higher than that without pre-stressing. The average DC breakdown field strength difference between the test with pre-stressing and without pre-stressing clearly shows that there is an apparent enhancement effect for the homo-charge injection on the DC breakdown.
