        Improvement effects of a mixed extract of flowers of <i>Pueraria thomsonii</i> Benth. and peels of <i>Citrus unshiu</i> Markovich on postmenopausal symptoms of ovariectomized mice

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Menopausal hot flushes occur frequently in postmenopausal women. In the present study, we investigated a regulatory effect of a mixed extract of flowers of <I>Pueraria thomsonii</I> Benth. and peels of <I>Citrus unshiu</I> Markovich (PCE17), an extract of flowers of <I>Pueraria thomsonii</I> Benth. (PE), an extract of peels of <I>Citrus unshiu</I> Markovich (CE), a mixture of tectorigenin 7-<I>O</I>-xylosylglucoside, tectoridin, and tectorigenin (Tec, the active compounds of PE), and hesperidin (Hes, an active compound of CE) on menopausal hot flushes.</P> <P><B>Methods</B></P> <P>We examined the anti-hot flushes properties of PCE17, PE, CE, Tec, or Hes using a mouse model of ovariectomy-induced hot flushes.</P> <P><B>Results</B></P> <P>The ovariectomy-induced rise in the tail skin temperature was significantly prevented by PCE17, PE, CE, Tec, or Hes. PCE17, PE, CE, Tec, or Hes significantly enhanced 5-HT levels and attenuated RANKL levels in the hypothalamus of ovariectomized (OVX) mice. Treatment with PCE17, PE, CE, Tec, or Hes significantly enhanced the levels of estrogen receptor (ER)-β, 5-HT1A, 5-HT2A, and tryptophan hydroxylase mRNA expression in the hypothalamus of OVX mice. PCE17, PE, or CE decreased follicle-stimulating hormone (FSH) and luteinizing hormone (LH) levels, but did not increase estrogen levels in the serum of OVX mice. Tec or Hes decreased FSH or LH levels and increased estrogen levels. Treatment with PCE17, PE, CE, or Tec ameliorated vaginal atrophy in OVX mice. Finally, PCE17, PE, CE, Tec, or Hes significantly increased norepinephrine and dopamine levels in the hypothalamus of OVX mice.</P> <P><B>Conclusion</B></P> <P>Thus, these results imply that PCE17 has protective effects against hot flushes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PCE17 prevented a rise in the tail skin temperature of OVX mice. </LI> <LI> PCE17 regulated the levels of 5-HT, RANKL, 5-HT1A, 5-HT2A, TPH, and MAO in OVX mice. </LI> <LI> PCE17 ameliorated vaginal atrophy in OVX mice. </LI> <LI> PCE17 has protective effects against hot flushes. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        How authors select covariates in the multivariate analysis of cancer studies in 10 oncology journals in Korea: a descriptive study

        Purpose: Cancer is the leading cause of death in Korea, leading many investigators to focus on cancer research. We present the current practice of variable selection methods for multivariate analyses in cancer studies recently published in major oncology journals in Korea. Methods: We included observational studies investigating associations between exposures and outcomes using multivariate analysis from 10 major oncology journals published in 2021 in KoreaMed, a Korean electronic database. Two reviewers independently and in duplicate performed the reference screening and data extraction. For each study included in this review, we collected important aspects of the variable selection methods in multivariate models, including the study characteristics, analytic methods, and covariate selection methods. The descriptive statistics of the data are presented. Results: In total, 107 studies were included. None used prespecified covariate selection methods, and half of the studies did not provide enough information to classify covariate selection methods. Among the studies reporting selection methods, almost all studies only used datadriven methods, despite having study questions related to causality. The most commonly used method for variable selection was significance in the univariate model, with the outcome as the dependent variable. Conclusion: Half of the included studies did not provide sufficient information to assess the variable selection method, and most used a limited data-driven method. We believe that the reporting of covariate selection methods requires improvement, and our results can be used to educate researchers, editors, and reviewers to increase the transparency and adequacy of covariate selection for multivariable analyses in observational studies.

      • 자율주행차의 음향 인터페이스가 보행자의 심리적 반응에 미치는 영향: 신뢰, 수용, 안전성, 불안감을 중심으로

        자율 주행 기술 상용화를 위해 세계 각국에서 꾸준히 논의 되고 있지만, 여전히 안전사고와 보안, 윤리적 문제 등 해결해야 할 문제 또한 지속해서 제기되고 있다. 자율 주행 차량에는 운전자의 부재로 인한 운전자와 보행자 간의 눈 마주침 같은 의사소통이 일어날 수 없기 때문에 새로운 형식의 의사소통을 위한 인터페이스의 제시가 요구된다. 그 중, 음향 인터페이스를 활용하여 보행자가 자율 주행 차량을 발견하고 길을 건너는 맥락에서 심리적 차이를 분석하였다. 결과를 통하여 음향 인터페이스 종류에 따라 자율주행차와 보행자 사이에 일어나는 심리적 반응에 대한 이해를 제공하고자 한다.

      • Delayed Afterdepolarization in Intact Canine Sinoatrial Node as a Novel Mechanism for Atrial Arrhythmia

        <P> <B>Delayed Afterdepolarization in Sinoatrial Node. <I>Introduction:</I></B> Recent evidence indicates that spontaneous sarcoplasmic reticulum Ca release and Na‐Ca exchanger current activation contribute to the sinoatrial node (SAN) automaticity. These findings suggest that SAN activity may share mechanisms that underlie both automaticity and triggered activity. The aim of this study is to test the hypothesis that spontaneous, nonvoltage gated, intracellular Ca (Ca<SUB>i</SUB>) elevation may induce delayed afterdepolarization (DAD) in intact SAN during isoproterenol infusion.</P><P> <B> <I>Methods and Results:</I> </B> We simultaneously mapped Ca<SUB>i</SUB> and membrane potential in 31 isolated Langendorff‐perfused canine right atriums (RA). Isoproterenol increased heart rate and late diastolic Ca<SUB>i</SUB> elevation (LDCAE) of the superior SAN, leading to consistent SAN automaticity in all 31 RAs. However, DAD‐like diastolic depolarizations (DD) were transiently observed in 4 RAs during isoproterenol infusion. These DAD‐like DDs were preceded by LDCAE, but did not trigger a full action potential. The LDCAE preceding DAD‐like DDs had smaller amplitude (0.41 ± 0.08 AU vs 0.48 ± 0.07 AU, P = 0.001) and less steep slopes (3.7 ± 1.3 AU/s vs 4.8 ± 1.4 AU/s, P = 0.001) than that of sinus beats. The coupling interval of DAD‐like DDs was longer than that of the preceding normal beats (407 ± 48 ms vs 371 ± 44 ms, P = 0.002).</P><P> <B> <I>Conclusion:</I> </B> The isoproterenol‐induced LDCAE of superior SAN induced a full action potential in most cases. However, if the LDCAE was too small to trigger an action potential, then it induces only DAD‐like DD. The failure of DAD‐like DD to consistently trigger a sinus beat is a novel mechanism of atrial arrhythmogenesis. <I>(J Cardiovasc Electrophysiol, Vol. 22, pp. 448‐454)</I></P>

        Attribution to Heterogeneous Risk Factors for Breast Cancer Subtypes Based on Hormone Receptor and Human Epidermal Growth Factor 2 Receptor Expression in Korea

        <▼1><P>Supplemental Digital Content is available in the text</P></▼1><▼2><P><B>Abstract</B></P><P>We conducted a heterogeneous risk assessment of breast cancer based on the hormone receptor (HR) and human epidermal growth factor receptor 2 (HER2) calculating the risks and population-based attributable fractions (PAFs) for modifiable and nonmodifiable factors.</P><P>Using matched case–control study design from the Seoul Breast Cancer Study and the national prevalence of exposure, the risks and PAFs for modifiable and nonmodifiable factors were estimated for total breast cancers and subtypes.</P><P>The attribution to modifiable factors was different for each subtype (luminal A, PAF = 61.4% [95% confidence interval, CI = 54.3%–69.8%]; luminal B, 21.4% [95% CI = 18.6–24.9%]; HER2-overexpression, 59.4% [95% CI = 47.8%–74.3%], and triple negative tumors [TNs], 27.1% [95% CI = 22.9%–32.4%)], and the attribution to the modifiable factors for the luminal A and HER2-overexpression subtypes was higher than that of the luminal B and TN subtypes (<I>P</I> heterogeneity ≤ 0.001). The contribution of modifiable reproductive factors to luminal A type in premenopausal women was higher than that of the other subtypes (18.2% for luminal A; 3.1%, 8.1%, and −3.1% for luminal B, HER2-overexpression, and TN subtypes, respectively; <I>P</I> heterogeneity ≤ 0.001). Physical activity had the highest impact preventing 32.6% of luminal A, 14.5% of luminal B, 38.0% of HER2-overexpression, and 26.9% of TN subtypes (<I>P</I> heterogeneity = 0.014). Total reproductive factors were also heterogeneously attributed to each breast cancer subtype (luminal A, 65.4%; luminal B, 24.1%; HER2-overexpression, 57.9%, and TN subtypes, −3.1%; <I>P</I> heterogeneity ≤ 0.001).</P><P>Each pathological subtype of breast cancer by HRs and HER2 status may be associated with heterogeneous risk factors and their attributable risk, suggesting a different etiology. The luminal B and TN subtypes seemed to be less preventable despite intervention for alleged risk factors, even though physical activity had a high preventable potential against breast cancer.</P></▼2>

        준설 물량 산출을 위한 지반정보 3차원 가시화 프로그램 비교 평가

        국내 및 해외에서 진행되고 있는 간척사업은 대규모 매립재가 필요하나 육상에서 얻을 수 있는 매립재는 한정되어 있어 해상준설토를 활용하기 위한 지반 정보화 관련 연구가 활발히 진행되고 있다. 이러한 연구의 일환으로 준설지역 지반조사 자료의 디지털 공간 정보화를 통해 신뢰도가 고려된 3차원 준설 물량의 자동 산출이 가능한 GIS 기반의 준설 매립지반 DB 구축 및 3차원 준설 매립 정보화 프로그램을 개발하였다. 현장에서 준설된 물량과의 비교 검증에 앞서 본 연구에서는 수집된 새만금 지역의 군장항 인근 지반조사 자료를 바탕으로 개발된 프로그램과 3차원 준설 매립 정보화 상용 프로그램으로 지반정보의 3차원 가시화를 수행하였다. 이를 토대로 가시화 수준, 각 지층의 단면 및 심도, 정량적 준설 물량 산출 결과를 비교하여 개발 프로그램에 대한 적정성을 평가하였으며, 평가결과 각각 산출된 준설 물량은 일치하는 것으로 나타났다. There are many reclamation projects domestically and internationally which requires large quantity of reclaimable materials. To provide enough reclaimable soils which are limited in land, there have been various research focusing on the dredged soils in the marine environments. As a part of this research, a GIS based 3D dredging reclamation visualization program was developed for the volume estimation of dredged soils in 2015. The developed program is based on the digitized spatial information of the site investigation data with a consideration of the reliability of the data. Prior to the validation with the comparisons with the actual dredged volume measurement data, the developed program was compared with the commercial 3D visualization program with 3D visualized results from the test site near the Gunjang harbor. The validation of the developed program was performed in terms of the degree of visualized precision, the sectional and profiling of soil layers and the dredged volume estimation results. Based on the comparisons, both commercial and developed program show similar dredged volume with minor discrepancies in soil layers.

        노천광산 덤프트럭의 최적 운반경로 분석을 위한 지형경사가 고려된 벡터 네트워크 자료의 생성 방법

        최근까지 노천광산을 대상으로 덤프트럭의 최적 운반경로 분석을 수행한 연구들은 주로 래스터 자료를 사용하였다. 그러나 래스터 자료는 격자 해상도에 따라 운반경로가 zigzag 형태로 심하게 왜곡될 수 있으며, 경로의 위상관계를 고려할 수 없는 한계가 있다. 벡터 네트워크 자료를 사용할 경우에는 이러한 문제점들을 해결할 수 있으나, 지형경사에 따른 덤프트럭의 성능 변화 특성이 최적 운반경로 분석시 반영될 수 있도록 할 필요가 있다. 본 연구에서는 노천광산 덤프트럭의 최적 운반경로 분석을 위해 지형경사에 따른 덤프트럭의 이동속도 변화를 반영할 수 있는 벡터 네트워크 자료의 생성 방법을 제시하였다. 수치지형모델 제작, 운반도로 디지타이징, 지형 경사도 계산, 경사도를 반영한 트럭의 이동속도 및 이동시간 계산 순으로 자료의 생성 작업이 진행된다. 제시한 벡터 네트워크 자료 생성 방법의 적용성과 효과를 알아보기 위하여 인도네시아 파시르 석탄노천광산의 로또 남부 채광장을 대상으로 간단한 사례연구를 수행한 결과, 제시한 방법이 ArcGIS Network Analyst 소프트웨어와 쉽게 호환이 가능하며, 최적 운반경로 분석시 지형경사에 따른 덤프트럭의 이동속도 변화를 효과적으로 반영할 수 있음을 확인할 수 있었다. Previous studies for analyzing optimal haulage routes of dump trucks in open pit mines mostly used raster data. However, the raster data has several problems in performing optimal route analyses: ⑴ the jagged appearance of haulage roads according the cell resolution often causes overestimation of the travel cost; ⑵ it difficult to trace the topological relationships among haulage roads. These problems can be eliminated by using vector network data, however a new method is required to reflect the performance characteristics of a dump truck according to terrain gradient changes. This study presents a new method to create vector network data with the consideration of terrain gradient for analyzing optimal haulage routes of dump trucks in open pit mines. It consists of four procedures: ⒜ creating digital elevation models, ⒝ digitizing haulage road networks, ⒞ calculating the terrain gradient of haulage roads, and ⒟ calculating the average speed and travel time of a dump truck along haulage roads. A simple case study at the Roto South pit in the Pasir open pit coal mine, Indonesia is also presented to provide proof that the proposed method is easily compatible to ArcGIS Network Analyst software and is effective in finding optimal haulage routes of dump trucks with considering terrain gradient in open pit mines.
